Lipids in the development and treatment of human intestinal diseases - Lipids in intestinal diseases

ABSTRACT: Interventions: Group 1: Colon tissue (tumor, control tissue and tissue from diverticulitis patients) which is optained within a surgical intervention at the Asklepios Paulinen Clinic in Wiesbaden is directly transferred to the Clinic of Pathology of the Helios Dr.-Horst-Schmidt-Clinic Wiesbaden, where it is categorized. Within 24 h the tissue is collected from a staff of the Insitute of clinical Pharamcology University of Frankfurt. At the Institute of clinical Pharmacology of the University hospital Frankfurt the tissue is prepared for immunohistochemistry, for detection of lipids by liquid-chromatography-tandem mass spectrometry, for single cell isolation, FACS analysis, and in vitro culture experiments. Reactivation of Human Herpesvirus 6 (HHV-6) in Diverticulitis

Project description:Background: Diverticular disease is a significant healthcare burden in the United States. Younger diverticulitis patients are at increased risk for recurrence. How the molecular pathophysiology differs from those that develop disease at an older age is not understood. We aimed to profile the colonic transcriptome from younger versus older diverticulitis patients to identify differential biological pathways contributing to disease. Methods: We performed RNA-seq on full-thickness sigmoid colon tissue obtained at the time of surgery on diverticulitis patients (n=26) diagnosed at a younger age (<42 years old) or at an older age (>65 years old). Viral reads were identified from the RNA-seq dataset and associated with clinical metadata and the host transcriptome. HHV-6 positivity was evaluated in diverticulitis patients by PCR and immunofluorescence. Patient sera was profiled for HHV-6 using qPCR and ELISA to detect anti-HHV-6 antibodies. Results: Using RNA-seq, diverticulitis patients were profiled for differential expression associated with age of diagnosis. A subset of younger diverticulitis patients (diverticulitis colonic transcriptome-viral signature (DCT-VS)) demonstrated increased expression of anti-viral response genes. We identified viral transcripts in the RNA-seq dataset and found HHV-6 transcripts negatively correlated with DCT-VS. Younger patients more frequently displayed evidence of HHV-6 infection through DNA analysis and immunofluorescence of colonic tissue. During acute disease, HHV-6 DNA was detected in the serum but was absent during disease quiescence. Conclusions: Patients diagnosed with diverticulitis at a younger age demonstrate reactivation of HHV-6 in the sigmoid colon that remains persistent. Future studies to assess the role of pathogenicity and the use of anti-virals for acute uncomplicated diverticulitis should be considered.

Expression RNA-Seq data of matched cases of pre-treatment Fine-needle aspiration (FNA) and the respective post neo-adjuvant treatment operative sample of breast cancer patients.

Project description:Neoadjuvant treated patients were selected from the Genomstudy of the University Women`s Clinic Heidelberg (2009-2016). This study includes primary breast cancer cases that were newly diagnosed at the University women's clinic of Heidelberg and had given their informed consent for participating in this study. This study was approved by the Ethical Committee of the Medical Faculty in Heidelberg. All cases were female and Caucasian. Fresh core needle biopsies and fresh tumor tissue were examined by a pathologist, snap-frozen in liquid nitrogen and stored at -80C within 15 min after sampling. Total RNA as well as DNA and protein were extracted by applying the QIAGEN All Prep Kit. All eluates were stored at -80C until usage. A sample swap most probably occurred for patient 207, so 207-S is actually in the T condition and 207-T in the S condition.

Project description:This CYCling Lynch patients for Exercise and Prevention (CYCLE-P) trial was a prospective, non-randomized controlled trial conducted at the Familial High-Risk Gastrointestinal and Cancer Prevention Clinic of The University of Texas MD Anderson Cancer Center. Twenty-one LS participants with a confirmed pathogenic mutation in one of four DNA mismatch repair (MMR) genes were enrolled between April 2018 and January 2019. Follow-up was conducted 1-year from the start of the intervention. Statistical analysis by both per-protocol and intention-to-treat was performed from March to May 2021.

Integration of genome-wide DNA methylome and transcriptome of human intestinal fibroblasts reveals novel candidate gene signatures in Crohnâs disease-associated fibrosis

Project description:Tissue fibrosis is a serious complication of Crohnâs disease (CD) as well as of a variety of other complex, chronic pathologies. Understanding the underlying pathophysiology of tissue fibrosis is crucial for the development of tissue-specific prevention and interventional treatment strategies. To identify molecular states specific to fibrotic disease, we employed deep sequencing to define the genome-wide DNA methylome and the whole transcriptome of purified human intestinal fibrotic fibroblasts (HIFs) isolated from the colon of patients with fibrotic CD. Integration of this information, via computational tools, identified candidate molecular interactions that could lead to fibrosis pathology. Our definition of a genome-wide fibrosis-specific DNA methylome provides a new paradigm for understanding mechanisms of pathological gene expression that lead to intestinal fibrosis and may have relevance to fibrogenesis in other organs. Human intestinal fibroblasts (HIFs) were extracted and cultured from colon specimens of two groups: Crohnâs disease with associated fibrosis (n=3) and normal fibroblasts from patients with Diverticulitis (n=3). Both RNA-seq and MBD-isolated genome sequencing (MiGS) were performed on every sample.

Role of mitochondria in intestinal epithelial cells

Project description:We aimed to study the role of mitochondria in intestinal epithelial cells by generating and analyzing mice with tamoxifen-inducible intestinal epithelial cell (IEC)-specific knockout of DARS2. DARS2 ablation caused severe mitochondrial dysfunction in IECs and resulted in suppression of epithelial cell proliferation and accumulation of lipids within large lipid droplets in enterocytes. To gain insight into the mechanism underlying the lipid processing defect, we used Lexogen 3' mRNA sequencing to compare the gene expression profiles of the proximal small intestine from mice with IEC-specific DARS2 knockout and control littermates at three days after tamoxifen induction, when the mice do not display yet lipid accumulation, and at seven days after tamoxifen induction, when enterocytes are filled with lipids.

Intestinal Microbiota Influences Non-intestinal Related Autoimmune Diseases.

Project description:The human body is colonized by millions of microorganisms named microbiota that interact with our tissues in a cooperative and non-pathogenic manner. These microorganisms are present in the skin, gut, nasal, oral cavities, and genital tract. In fact, it has been described that the microbiota contributes to balancing the immune system to maintain host homeostasis. The gut is a vital organ where microbiota can influence and determine the function of cells of the immune system and contributes to preserve the wellbeing of the individual. Several articles have emphasized the connection between intestinal autoimmune diseases, such as Crohn's disease with dysbiosis or an imbalance in the microbiota composition in the gut. However, little is known about the role of the microbiota in autoimmune pathologies affecting other tissues than the intestine. This article focuses on what is known about the role that gut microbiota can play in the pathogenesis of non-intestinal autoimmune diseases, such as Grave's diseases, multiple sclerosis, type-1 diabetes, systemic lupus erythematosus, psoriasis, schizophrenia, and autism spectrum disorders. Furthermore, we discuss as to how metabolites derived from bacteria could be used as potential therapies for non-intestinal autoimmune diseases.
