Project description:Regional delivery of oncolytic viruses has been shown to promote immune responses. Malignant pleural effusions comprise an immunosuppressive microenvironment, and the ability of oncolytic viruses to generate immune responses following regional delivery in patients with malignant pleural effusions is unknown. We conducted a phase I clinical trial that studied the intrapleural administration of oncolytic vaccinia virus to establish the safety and feasibility in patients with malignant pleural effusion due to malignant pleural mesothelioma or metastatic disease. In patients with malignant pleural mesothelioma, by correlative analysis of pre- and post-treatment tumor biopsies, we provide insight into tumor-specific viral uptake and associated immune responses.

Project description:Embryo DNA fingerprinting represents an important tool for tracking embryo-specific outcomes after multiple embryo transfer during IVF. The situation in which 2 embryos are transferred and only one implants represents a unique opportunity for the most well-controlled comparison of competent and incompetent embryos. Specifically, this design eliminates all patient-related variables from the comparison of embryos with or without reproductive potential. However, in order to determine which embryo implanted, the investigator must wait until newborn DNA is available upon delivery. This study validates a non-invasive fetal DNA fingerprinting method that reduces the time to identify which embryo implanted by approximately 31 weeks. Thirty-four patients were studied to determine if fingerprinting of fetal DNA extracted from maternal plasma at 9 gestational weeks concurred with the buccal DNA results obtained from the newborn after delivery. This validation required single nucleotide polymorphism (SNP) profiles on each couples’ preimplantation embryos, enriched fetal DNA from maternal plasma at 9 weeks gestation, and newborn DNA obtained from buccal swabs after delivery. The predictions from fetal DNA-based embryo tracking and gender assignments made at 9 weeks gestation were 100% consistent with standardized methods of assessment performed after term delivery. This study demonstrates the first validated fetal DNA fingerprinting method which predicts both gender and which embryo implanted at 9 weeks gestation following multiple embryo transfer. Affymetrix SNP arrays were processed and successfully completed according to the manufacturer's directions on DNA extracted from 136 embryos, 33 parental blood samples, 17 enriched fetal DNA samples and 21 buccal DNA samples.

Project description:The aim of this feasibility study was to assess whether inflammatory status of the lymphatic vessels (LVs) and/or changes in the miRNA profile of the LVs have potential prognostic and predictive value for overall outcome and risk of relapse. Tissue specimens were obtained from the external iliac vessels draining the pelvic region of patients undergoing debulking surgery (n=10). The tissue was transported directly from surgical theatre to the laboratory in ice-cold sterile Phosphate Buffered Saline. LVs were identified and isolated along with a small amount of surrounding tissue under a dissection microscope within a sterile laminar flow cabinet. Each vessel was divided into two sections. One half of each vessel was further cleaned of surrounding fatty tissue, placed in RNA stabilising solution, frozen on dry ice and stored at -20°C. The miRNAs were extracted with an miRNA easy kit and reverse-transcribed to cDNA using a miScript II RT Kit . Subsequent qPCR was carried out using a miScriptSybrGreen real-time PCR kit and pre-defined miScript miRNA PCR Array. Quantification of the inflammatory state (low, medium and high) and presence of cancer-infiltration of each LV was carried out using immunohistochemistry with the remaining half of each vessel. The LV miRNA expression profiling was then analysed in the context of high versus low inflammation, and cancer-infiltrated versus non-cancer-infiltrated. Results were correlated with clinical outcome data including relapse with an average follow-up time of 13.3 months.

Project description:Neurons in posterior parietal cortex contribute to the execution of goal-directed navigation and other decision-making tasks. Although molecular studies have catalogued over fifty cortical cell types, it remains unknown what distinct functions they serve during goal-directed navigation. Here, we identified a molecularly defined subset of somatostatin (Sst) inhibitory neurons that, in mouse posterior parietal cortex, carry a novel cell type-specific error correction signal for navigation. We obtained repeatable experimental access to these cells using an adeno-associated virus (AAV) in which gene expression is driven by an enhancer that functions specifically in a subset of Sst cells. We found that during goal-directed navigation in a virtual environment, this subset of Sst neurons activates in a synchronous pattern that is distinct from the activity of surrounding neurons, including other Sst neurons. Using in vivo two-photon photostimulation and ex vivo paired patch clamp recordings, we show that nearby cells of this Sst subtype excite each other through gap junctions, revealing a self-excitation circuit motif that contributes to the synchronous activity of this cell type. Remarkably, these cells selectively activate as mice execute course corrections for deviations in their virtual heading during navigation toward a reward location, both for self- and experimentally-induced deviations. We propose that this subtype of Sst neurons provides a self-reinforcing and cell type-specific error-correction signal in posterior parietal cortex that may aid the execution and learning of accurate goal-directed navigation trajectories.

Project description:Fiber tracking is a technique that, based on a diffusion tensor magnetic resonance imaging dataset, locates the fiber bundles in the human brain. Because it is a computationally expensive process, the interactivity of current fiber tracking tools is limited. We propose a new approach, which we termed real-time interactive fiber tracking, which aims at providing a rich and intuitive environment for the neuroradiologist. In this approach, fiber tracking is executed automatically every time the user acts upon the application. Particularly, when the volume of interest from which fiber trajectories are calculated is moved on the screen, fiber tracking is executed, even while it is being moved. We present our fiber tracking tool, which implements the real-time fiber tracking concept by using the video card's graphics processing units to execute the fiber tracking algorithm. Results show that real-time interactive fiber tracking is feasible on computers equipped with common, low-cost video cards.

Project description:A number of studies have shown that performing a secondary task while executing a time-judgment task impairs performance on the latter task. However, this turns out not to be the case for certain motor secondary tasks. We show that concomitant secondary motor tasks involving pointing, when performed during a time-judgment task, can actually improve our time-judgment abilities. We compared adult participants' performance in a time-of-movement paradigm with visual pursuit-only and with visual pursuit plus hand pursuit. Rather than interfering with their estimation of stimulus movement duration, the addition of hand pursuit significantly improved their judgment. In addition, we considered the effect of three different movement profiles and four different movement speeds for the moving stimulus. As predicted by Vierordt's law, time judgments of shorter stimuli are overestimated and longer stimuli underestimated. Finally, timing performances appear to improve when the moving target follows a "biological" velocity profile.
