Polyhexamethylene guanidine aerosol causes irreversible changes in blood proteins that associated with the severity of lung injury

Project description:Humidifier disinfectant (HD)-associated lung injury (HDLI) not only associates with increased acute morbidity and mortality, but also affects long term lung function due to lung fibrosis. However, there have been no validated marker capable of detecting exposure to HDs or severity of HDLI. This study aims to discover the differential abundant plasma proteome between them and develop prediction score platform using plasma proteome data from HD-exposed patients in order to classify the severity of HDLI. A total of 483 samples were used in this study. The samples used were 162 children in the normal group, 76 in the 4th stage, 51 in the 3rd stage, 54 in the 2nd stage, 42 in the 1st stage and 50 female adults in the normal group, 20 in the 4th stage, 7 in the 3rd stage, 6 in the 2nd stage, and 14 in 1st stage. The lower the grade, the greater the severity of HDLI.

Transcription profiling of human placentae of different gestational ages: an opru network and uw scor study

Project description:We used a whole genome approach to identify major functional gene categories (including xenobiotic transporters and metabolizing enzymes) whose expression depends on gestational age. STUDY DESIGN: We compared gene expression profiles of 1st (45-59 days) and 2nd trimester (109-115 days), and C-section term placentae. RESULTS: In 1st trimester placentae, genes related to cell cycle, DNA, aminoacids and carbohydrate metabolism were significantly overrepresented, while genes related to signal transduction were downregulated. In the organism defense category, we identified genes involved in chemical response, metabolism, and transport. Analysis of signal transduction pathways suggested, and subsequently confirmed independently, that the Wnt pathway was regulated by gestational age. CONCLUSIONS: Our study will serve as a reference database to gain insight into the regulation of gene expression in the developing placentae and, thus, allow comparisons with placentae from complicated pregnancies such as those in women experiencing gestational diabetes, pre-eclampsia and teratogenic sequelae. EP001: The interferon transcriptome in uterine NK cells is upregulated during the second trimester of pregnancy allowing extravillous trophoblast invasion

Project description:Uterine Natural Killer (uNK) cells regulate essential developmental processes at the maternal-fetal interface during early pregnancy. Uterine NK cell functions are tightly regulated during early placentation to stimulate trophoblast invasion and remodel uterine spiral arteries. Access to human 1st and 2nd trimester uterine tissues allowed us to investigate the transcriptional changes in uNK cells during the early stages of early human pregnancy. Microarray analysis identified 97 upregulated genes in 2nd compared to 1st trimester purified uNK cells of which the majority (61%) clustered as interferon-stimulated-genes (ISG), with ISG15 and ISG20 being upregulated profoundly. Type I interferons (IFNα/β), but not type II interferon (IFNɣ) increased expression of the identified interferon target genes ISG15 and ISG20 in uNK cells in vitro. Moreover, the cytokine-like protein ISG15 stimulated in vitro trophoblast invasion. Second trimester uNK cells promoted trophoblast invasion in vitro, whereas both 1st and 2nd trimester uNK cells stimulated endothelial tube formation. IFNα but not IFNβ stimulation of 1st trimester uNK cells enhanced their capacity to promote trophoblast invasion. In conclusion, the uNK cell interferon transcriptome is upregulated during the 2nd trimester allowing uNK cells to promote trophoblast invasion. Type I interferon signaling regulates uNK cell-induced trophoblast invasion via induction of effector molecules like ISG15. First trimester uNK cells can be induced to act like second trimester uNK cells by IFNα with respect to promotion of trophoblast invasion. Project description:Intervention1: Warm foot bath therapy: In experimental arm, Warm foot bath therapy Each session-15min. Participants will receive warm foot bath therapy prior oxaliplatin for 15min.and post oxaliplatin for 15min.on 1st day of 2nd,3rd,4th,5th,6th chemotherapy cycle and in home at night once for 15min.till completion of 6cycles. In control arm, Participants will not receive the above intervention; but only receive routine treatment. Control Intervention1: standard routine treatment No interventions: In experimental arm, Warm foot bath therapy Each session-15min. Participants will receive warm foot bath therapy prior oxaliplatin for 15min.and post oxaliplatin for 15min.on 1st day of 2nd,3rd,4th,5th,6th chemotherapy cycle and in home at night once for 15min.till completion of 6cycles. Gene expression in fetal mesenchymal stem cells for identification of epitopes suitable for non-invasive isolation

Like other fetal cell types they cross the placenta, and can be found in maternal organs decades later. To determine potential ligands in human fetal mesenchymal stem cells not present in maternal blood, the gene expression of 1st trimester human fetal bone marrow, liver and blood derived mesenchymal stem cells will be compared to blood mononuclear cells from pregnant women using a Affymetrix human gene array system.
