Velidation research of colonoscopy using artificial intelligence

ABSTRACT: Interventions: We enrolled 415 subjects who under went colonoscopy at our institution from January 2021 to March 2022.The randomization method used in this study was the numbered container method according to the CONSORT guideline. The allocation message was generated by the computer program of the Python random module and then sealed in sequentially numbered identical containers according to the allocation sequence. Evaluation of the therapeutic effect of crocin along with standard regimes in colorectal cancer

Project description:Intervention 1: Intervention group: Crocin along with standard regime will be assigned to intervention group. The daily dose of crocin will be 30 mg (two 15 mg tablets) during 12 weeks. Comparing malignant monocytosis across the updated WHO and ICC classifications of 2022

Project description:The World Health Organization Classification of Hematolymphoid Tumors (WHO) and the International Consensus Classification (ICC) of 2022 introduced major changes to the definition of CMML. To assess qualitative and quantitative implications for patient care, we started with 3,311 established CMML cases (according to WHO 2017 criteria) and included also 2,130 oligomonocytosis cases fulfilling the new CMML diagnostic criteria. Applying both classification systems from 2022, 356 and 241 of oligomonocytosis cases were newly classified as myelodysplastic (MD)-CMML (WHO and ICC 2022, respectively), most of which were diagnosed as MDS according to WHO 2017. Importantly, 1.5 times more oligomonocytosis cases were classified as CMML according to WHO 2022 than based on ICC, due to different diagnostic criteria. Genetic analyses of the newly classified CMML cases showed a distinct mutational profile with strong enrichment of MDS-typical alterations, resulting in a transcriptional subgroup separated from established MD- and myeloproliferative (MP)-CMML. Despite a different cytogenetic, molecular, immunophenotypic, and transcriptional landscape, no differences in overall survival were found between newly classified and established MD-CMML cases. To the best of our knowledge, this study represents the most comprehensive analysis of routine CMML cases to date, both in terms of clinical characterization and transcriptomic analysis, placing newly classified CMML cases on a disease continuum between MDS and previously established CMML.

Comparison of the root transcriptome from plants grown on different growing substrates (hydroponics and soil).

Project description:In order to decipher the molecular alterations caused by the growing medium, we harvested the roots of plants grown in hydroponics and on soil for 6 weeks under 10-hour short days. The temperature was 20Â°C (day and night), the relative humidity was 70 %, and the light was provided by cool-white fluorescent tubes at 100 Î¼E.mâ2.sâ1 during the first 10 days, 60 Î¼E.mâ2.sâ1 later. The soil was composed of a mixture of peat compost and sand (3:1 v/v), which was watered daily with tap water. Plants were transferred to 1-liter containers (six plants/container) 18 days after sowing. Plants grown in hydroponics were sown on agar-containing (6.7 g.lâ1; Kalys, France) seed-holders placed on 2-liter black containers and accessories (http://www.araponics.com). The hydroponic solution, changed every two weeks, was prepared as described in Tocquin et al. (2003). During the first three weeks of growth, the number of plants per 2-liter tank was 35 and 18 afterwards. Harvesting was performed 8 hours after the beginning of the light period (16h30), and the roots from 35 to 45 individuals were pooled. Three independent biological replicates were performed for plants grown on both media.

High-Throughput Single-Cell Labeling (Hi-SCL) for RNA-Seq using drop-based microfluidics

Project description:The importance of single-cell level data is increasingly appreciated, and significant advances in this direction have been made in recent years. Common to these technologies is the need to physically segregate individual cells into containers, such as wells or chambers of a micro-fluidics chip. High-throughput Single-Cell Labeling (Hi-SCL) in drops is a novel method that uses drop-based libraries of oligonucleotide barcodes to index individual cells in a population. The use of drops as containers, and a microfluidics platform to manipulate them en-masse, yields a highly scalable methodological framework. Once tagged, labeled molecules from different cells may be mixed without losing the cell-of-origin information. Here we demonstrate an application of the method for generating RNA-sequencing data for multiple individual cells within a population. Barcoded oligonucleotides are used to prime cDNA synthesis within drops. Barcoded cDNAs are then combined and subjected to second generation sequencing. The data are deconvoluted based on the barcodes, yielding single-cell mRNA expression data. In a proof-of-concept set of experiments we show that this method yields data comparable to other existing methods, but with unique potential for assaying very large numbers of cells.

Drosophila melanogaster gene expression changes after spaceflight.

Project description:Gene expression levels were determined in 3rd instar and adult Drosophila melanogaster reared during spaceflight, to elucidate the genetic and molecular mechanisms underpinning the effects of microgravity on the immune system. The goal was to validate the Drosophila model for understanding alterations of innate immune responses in humans due to spaceflight. Five containers of flies, with ten female and five male fruit flies in each container, were housed and bred on the space shuttle (average orbit altitude of 330.35 km) for 12 days and 18.5 hours, with a new generation reared in microgravity. RNA was extracted on the day of shuttle landing from whole body animals (3rd instar larvae and adults), hybridized to Drosophila 2.0 Affymetrix genome arrays, and the expression level of all genes was normalized against the gene expression level from the corresponding developmental stage animals raised on ground. Spaceflight altered the expression of larval genes involved in the maturation of plasmatocytes (macrophages) and their phagocytic response, as well as the level of constitutive expression of pattern recognition receptors and opsonins that specifically recognize bacteria, and of lysozymes, antimicrobial peptide pathway and immune stress genes, hallmarks of humoral immunity. Larval microarrays (FL 6 samples) are based on RNA extracted from 6 independent sets of 50 mid 3rd instar larvae reared in microgravity and collected on the day of landing after 12 days and 18.5 hours on the space shuttle and the same number of control larvae raised on ground (GL 6 samples). Adults microarrays (F1 3 samples) are based on RNA from 3 sets of 20 adult females each, that emerged during spaceflight and within 4 hours of landing and the same number of adult females from the corresponding ground control containers (G1 3 samples).
