Fluorescent Probe VGT-309 to ID Cancerous Colorectal Lesions During Augmented Colonoscopy

ABSTRACT: The purpose of this study is to determine the safety and feasibility of VGT-309 for the visualization of colorectal tumors in real-time using near-infrared (NIR) fluorescence endoscopy. In addition, signatures of 50+ biomarkers will be evaluated in biopsies using CODEX multi-plexing.

Project description:Adult male and female w1118 flies were inflicted with traumatic brain injury using high-impact trauma device. Brains were dissected at control, 1, 2 and 4 hours post-TBI and exposed to near-infrared (NIR) light at 750 nm for 2 hours. Total RNA was extracted from fly brains and 3'mRNA-seq was performed to assess gene expression changes in response to TBI and NIR eposure in both sexes. 3 replicates were sequenced for each condition.

Photothermal Modulation of Human Stem Cells Using Light-responsive 2D Nanomaterials

Project description:Two-dimensional (2D) molybdenum disulfide (MoS2) nanomaterials are an emerging class of biomaterials that are photo-responsive at near-infrared wavelengths (NIR). Here, we demonstrate for the first time the ability of 2D MoS2 to modulate cellular functions of human stem cells through photothermal mechanisms. The interaction of MoS2 with human stem cells is investigated using whole transcriptome sequencing (RNA-seq). Global gene expression profile of stem cells reveals significant influence by NIR stimulation of MoS2 on integrins, cellular migration and wound healing. Overall, the combination of MoS2 and NIR light may provide new approach to control and regulate cellular migration and functions.

Molecular Fluorescence Endoscopy in Patients With Familial Adenomatous Polyposis, Using Bevacizumab-IRDye800CW

Project description:There is a need for better visualization of polyps during surveillance endoscopy in patients with hereditary colon cancer syndromes like Familial Adenomatous Polyposis (FAP) and Lynch Syndrome (LS), to improve the adenoma detection rate. Optical molecular imaging of adenoma associated biomarkers is a promising technique to accommodate this need. The biomarker Vascular Endothelial Growth Factor (VEGF) is overexpressed in adenomatous colon tissue versus normal tissue and has proven to be a valid target for molecular imaging. The University Medical Center Groningen (UMCG) developed a fluorescent tracer by labeling the VEGF-targeting humanized monoclonal antibody bevacizumab, currently used in anti-cancer therapy, with the fluorescent dye IRDye800CW. The investigators hypothesize that when bevacizumab-IRDye800CW is administered to patients, it accumulates in VEGF expressing adenomas, enabling adenoma visualization using a newly developed near-infrared (NIR) fluorescence endoscopy platform (NL43407.042.13). This hypothesis will be tested in this feasibility study, next to the determination of the optimal tracer dose.

Project description:To clarify the effects of near-infrared radiation, we assessed DNA microarray after water-filtered broad-spectrum near-infrared (1100-1800 nm together with a water-filter that excludes wavelengths 1400-1500 nm) irradiation. We performed 5 rounds of near-infrared irradiation (at 10 J âcm2) using 2 sets of transparent polycarbonate plates, one to block UV and the other to block both UV and near-infrared.

HSP90 specific nIR probe identifies aggressive prostate cancers: a translational study from preclinical models to a human pilot study

Project description:Current screening modalities for prostate cancer rely on serologic testing for PSA and transrectal biopsy; however, many prostate cancers behave indolently.Because aggressively growing tumors depend on oncogenic drivers, we hypothesized that we could identify early, but aggressive prostate cancer,by an imaging modality targeting Hsp90, a chaperone for many molecules involved in oncogenic signaling. Previously we observedgreater avidity ofa probe,consisting of a near infrared dye tethered to an Hsp90 inhibitor,for cancers with greatermetastatic potential. We now report that an Hsp90 inhibitor-linked near infrared dye with greater tissue penetration could detect prostate cancers in preclinical models and in a phase I human clinical trial. These data, demonstrating specific uptake into aggressive tumors, in conjunction with our prior observations on photodynamic therapy guided by Hsp90 expression support a theranostic approach todetect and treatearly aggressive prostatecancers.
