WTCCC3 Anorexia Nervosa

ABSTRACT: As part of the Genetic Consortium for Anorexia Nervosa (GCAN) and Wellcome Trust Case Control Consortium 3 (WTCCC3), we have amassed the largest anorexia nervosa (AN) sample in the world (~4,000). Following the WTCCC3 GWAS, we secured funding from the Klarman Family Foundation to extend the genetic analyses to variants on the exome chip (CoreExome array). This pre-lim relates to carrying out genotyping on the CoreExome array on up to a maximum of 580 new samples.

Mammalian Methylation Consortium

Project description:The Mammalian Methylation Consortium aimed to characterize the relationship between cytosine methylation levels and a) species characteristics such as maximum lifespan and b) individual sample characteristics such as age, sex, tissue type. Both supervised machine learning approaches and unsupervised machine learning approaches were applied to the data as described in the citations. To facilitate comparative analyses across species, the mammalian methylation consortium applied a single measurement platform (the mammalian methylation array, GPL28271) to n=15216 DNA samples derived from 70 tissue types of 348 different mammalian species (331 eutherian-, 15 marsupial-, and 2 monotreme species). Most of the CpGs are located in highly conserved stretches of DNA but not all CpGs apply to all species as detailed in the description of the platform, GPL28271 and on https://github.com/shorvath/MammalianMethylationConsortium/.

NC HHEAR Hub Pilot Study within the HHEAR Consortium

Project description:This data generation, and subsequent data analysis was conducted by the NC HHEAR Hub as part of a larger HHEAR Consortium Pilot Study. A goal of this study to compare the metabolite identifications and annotations provided by HHEAR Consortium Laboratories as well as non-HHEAR Consortium Laboratories. Urine samples were provided by the Duke HHEAR Hub. This included pools of 3 spot urines collected over 48 hours, which were selected based on maximum overlap of collected data on other exposure matrices (targeted PFAS panel (serum), passive air sampling, wristband data). Dr. Yuan Li of the NC HHEAR Hub had primary responsibility for the generation of the untargeted data.

Project description:<p>DNA samples were obtained from participants of the Geisinger MyCode biobank. Phenotype data to determine case or control status for abdominal aortic aneurysm or extreme obesity were derived from Geisinger electronic medical record data through an enterprise data warehouse. Samples were genotyped using the Illumina Human CoreExome-12 v1.0 Array. Genotype data were filtered using standard quality control measures.</p>

The heat shock response of killifish (Fundulus heteroclitus): candidate gene and heterologous microarray approaches.

Project description:Subspecies of the Atlantic killifish, Fundulus heteroclitus, differ in their maximum thermal tolerance. To determine whether there is a link between the heat shock response (HSR) and maximum thermal tolerance, we exposed 20ºC acclimated killifish from these subspecies to a 2hr heat shock at 34ºC and examined gene expression during heat shock and recovery using real time quantitative PCR and a heterologous cDNA microarray designed for salmonid fishes.
