Genetic determinants of glycated hemoglobin levels in the Greenlandic Inuit population

ABSTRACT: We recently showed that a common genetic variant leads to a remarkably increased risk of type 2 diabetes (T2D) in the small and historically isolated Greenlandic population. Motivated by this, we aimed at investigating the genetic determinants for glycated hemoglobin (HbA1C). We found no novel associations but saw that for 10 out of 15 known HbA1C loci that the effects in Greenlandic Inuit were similar to previously reported. One locus, ANK1, showed a significant ancestral population specific effect. Our results shed light on the genetics effects across ethnicites.

Whole RNA profiling of Tuberculosis-diabetes comorbidity linked to diabetes with poor glycemic control

Project description:Type 2 diabetes (DM2) is a disease that hinders tuberculosis (TB) control strategies worldwide. TB infection, progression, morbidity, treatment success, adverse effects, relapse, and mortality are worse in these susceptible populations. Therefore, expanding the partially known molecular mechanisms leading to TB-DM2 comorbidity may be essential to face both epidemics. Consequently, a transcriptomic study was conducted on TB-DM2 comorbidity, associated with patients with DM2 who have poor glycaemic control (PDM2), since they have a higher risk of getting TB. Human blood samples from healthy controls (CTRL, HbA1c < 6.5%), TB ( HbA1c < 6.5%), TB-DM2, DM2 (HbA1c < 8.9%), and PDM2 (HbA1c > 10%) groups (n=4 each) were analyzed using high-density human GE 4X44K v2 differential expression microarrays.

Evaluation of volumetric absorptive microsampling and mass spectrometry data-independent acquisition in hemoglobin related clinical markers

Project description:Data-independent acquisition (DIA) allows comprehensive proteome coverage, and it has the potential to work as a unified protocol to determine a multitude of proteins in blood. Because of its high specificity, mass spectrometry has a high potential to reduce the interferences presented by other assays in the evaluation of blood markers. Here, we combined DIA with volumetric absorptive microsampling (VAMS) and automated proteomics sample processing as a platform to perform clinical markers determination. We evaluate, as a proof-of-concept, two hemoglobin-related clinical biomarkers: glycated hemoglobin (HbA1c) and hemoglobin variants. HbA1c by DIA showed good correlation with the reference method, but method imprecision did not meet the very low imprecision specification for the clinical evaluation of this biomarker. A strategy for identification of Hb variants was developed based on a customized database combined with a workflow for DIA data extraction and rigorous evaluation of peptides. This approach revealed Hb variants not identified by routinely clinical methods such as Hb Woodville and Hb Okayama.

Intra- and inter-individual metabolic profiling highlights carnitine and lysophosphatidylcholine pathways as key molecular defects in type-2 diabetes

Project description:In this study we explored a unique cohort of 43 samples to build a map of metabolites varying in T2D across five different tissues (subcutaneous adipose tissue, liver, pancreatic islets, skeletal muscle and blood serum). The samples originate from the Excellence in Diabetes biobank (EXODIAB) in Sweden. To our knowledge, this is the first attempt to create a map of metabolites across various metabolic-relevant tissues for the same cohort of individuals. In total we identified 286 unique metabolites, 32% of which were significantly altered between non-diabetes and T2D. We found evidence that amino acids (AAs) and bile acids are elevated for specific tissues in T2D and significantly associated to the percentage of glycosylated hemoglobin A1c (HbA1c). We showed that carnitines and lysophosphatidylcholines (LPCs) are the most significantly altered metabolites in muscle, liver and serum in T2D. Finally, we attempted to explore the progression to overt T2D and suggest potential biomarkers in the early development of T2D, i.e. from non-diabetes to pre-diabetes to T2D.

Project description:Human serum albumin is the most abundant plasma protein with a large number of lysine and arginine residues. Hence, it is highly susceptible to glycation in vivo. In hyperglycemic conditions, such as diabetes, the level of HSA glycation increases. Here we quantified glycated HSA peptides in a subject population of healthy and type 2 diabetes with and without nephropathy to assess its performance for the diagnosis of diabetic nephropathy compared to HbA1c.

Project description:Men and women with type 2 diabetes, all born in Europe, were recruited to this phase II clinical trial to examine the possible effects of tadalafil on insulin resistance. The study had a cross-over design and participants were randomised to either start with tadalafil or placebo for 6 weeks. At the end of the tretment period participants were extensively examined, including a hyperinsulinemic clamp. Initial treatment period was followed by 8 weeks of wash-out, before participants started with the other treatment for 6 weeks, and a new examination. Tadalafil showed no effect on the primary endpoint (clamp), but did reduce HbA1c, a secondary endpoint.

Project description:Analysis of evolved changes in transcriptional plasticity and parental effects on plasticity induced by mild heat stress in the nematode Caenorhabditis remanei. Results of this study highlight the importance of the broad environmental context of an organism and its influence on phenotypic plasticity, parental effects, and evolutionary responses. mRNA profiles of ancestral and two experimentally evolved populations of C. remanei. Parents of the sampled worms were raised at either 20Â°C or 30Â°C, then the resulting embryos were divided and reared at either 20Â°C or 30Â°C prior to collection (as L1 larvae). 6 replicates/larval temperature for each population if the parents were raised at 20Â°C, and 2 replicates/larval temperature for each population if the parents were raised at 30Â°C.

Quantitative Proteome Analysis Reveals Increased Content of Basement Membrane Proteins in Arteries from Patients with Type 2 Diabetes and Lower Levels among Metformin Users

Project description:We analyzed non-atherosclerotic repair arteries gathered at coronary by-pass operations from 30 patients with type 2 diabetes, as well as from 30 age- and gender-matched non-diabetic individuals. Quantitative proteome analysis was done by iTRAQ-labelling and LC-MS/MS analysis on individual arterial samples. The amounts of the basement membrane (BM) components, alpha-1- and alpha-2- type IV collagen, gamma-1- and beta-2-laminin were significantly increased in patients with diabetes. Moreover, the expressions of basement membrane components and other vascular proteins were significantly lower among metformin users, as compared to non-users. Patients treated with or without metformin had similar levels of HbA1c, cholesterol and blood pressure. In addition, quantitative histomorphometry showed increased area fractions of collagen-stainable material in tunica intima and media among patients with diabetes.

Immune dysfunction in intermediate hyperglycaemia and diabetes patients in tuberculosis

Project description:People living with diabetes have an increased risk of developing active tuberculosis. The effects of diabetes (HbA1c ≥6.5%) and intermediate hyperglycaemia (HbA1c 5.7-6.5%), on this transcriptomic signature were investigated by RNA-seq, to enhance understanding of immunological susceptibility in diabetes-tuberculosis comorbidity.Diabetes increased the magnitude of gene expression change in the host transcriptome in tuberculosis, characterised by an increase in innate, and decrease in adaptive immune responses. Strikingly, patients with intermediate hyperglycaemia and tuberculosis exhibited blood transcriptomes much more similar to diabetes-tuberculosis patients than to uncomplicated tuberculosis patients. Aberrant transcriptomes unveil a susceptibility mechanism of DM patients to TB of enhanced inflammation and reduced interferon responses.
