Project description:HCA Endometrium_LM The endometrium regenerates monthly and its transformation is executed through dynamic changes in states and interactions of multiple cell types. Using transcriptomics methods we seek to profile changes of the endometrium across the menstrual cycle. Our map will have implications in women's health and cancer, by enabling the interpretation of GWAS analyses or the studying functional consequences of somatic mutations.

Project description:Cell atlas of the human placenta
The Human Cell Atlas initiative is an ambitious global research effort which aims to describe every cell in the human body across all developmental stages to generate a reference map to accelerate progress in science and medicine particularly relating to human development and disease. The initiative is led by investigators based at the Wellcome Sanger Institute and the Broad Institute of MIT and Harvard and funding support from the Wellcome and NIH. This specific project will assess human development. We aim to interrogate the cellular composition and molecular regulators underpinning development and maturation through single cell RNA-sequencing/spatial transcriptomics and computational algorithms to predict cellular developmental trajectories and cell-cell interactions. We will subsequent validate these findings in situ using allied imaging technologies e.g. immunohistochemistry and RNAscope and in vitro culture of human intestinal organoids (HIOs).

For information on the proper use of pre-publication data shared by the Wellcome Trust Sanger Institute (including details of any publication moratoria), please see http://www.sanger.ac.uk/datasharing/ Schistosomes are blood dwelling digenean trematodes that mature as adults in the intestinal or urinary veins, predominantly of mammals. Adult female Schistosomes produce eggs of which approximately 40% fail to pass into faeces or urine (species-dependent) but are dispersed by the blood stream into different organs where they provoke severe inflammation. This parasitic infection is known as schistosomiasis and considered by the WHO as the second most socioeconomically devastating parasitic disease, next only to malaria, with hundreds of millions infected worldwide. As the eggs represent the causative pathogenic agents, the understanding of egg forming processes, and therefore of the schistosomal reproductive biology in general, is of fundamental interest. Additionally the differences between immature (pre-mating) and mature (post-mating) adult worms are of significant interest; adults only reach maturity upon mating. Trasncriptomic sequencing of gonad-specific cellular material will help to unravel signal transduction cascades involved in e.g. gametogenesis and/or vitellogenesis. This project aims to characterize the transcriptome profiles of immature and mature adult ovaries and adult testes.

Project description:Cell Atlas of human fetal gonads
The HUGODECA consortium brings together leading European academic and industrial experts (from 5 different EU countries). It includes active contributors and executives of the Human Cell Atlas (HCA) which will ensure complementarity with other ongoing HCA efforts. The overall HUGODECA concept is grounded on the integration of multiple synergistic expertise and technologies: Single cell profiling, spatial transcriptomic, 2D Mass cytometry and cyclic immunofluorescence and 3D imaging of optically cleared gonads. HUGODECA will implement novel tools, analytical and computational methods to process and integrate multidimensional OMICS and image data across different platforms. It will evaluate the accuracy of ex vivo culture models of human gonadal development and assess consequences of altering key signalling pathways.

Project description:HCA Placental Infection Atlas
Self-renewing human trophoblast stem cells (hTSC) can give rise to major trophoblast lineages, including syncytiotrophoblast (SCT) and extravillous trophoblast (EVT). hTSC has facilitated the study of placental development and function in vitro. Here, we aim to utilise the hTSC line (HMDMC 20/0005) to study human placental infection and inflammation. This study will provide the opportunity to better understand the pathogenesis of human placenta in the event of infection and inflammation. In brief, differentiated hTSC will be infected with relevant pathogens and stimulated with a panel of inflammatory cytokines. We will use single-cell genomic and spatial approaches to characterise cellular heterogeneity in response to infection or cytokines stimulus. In parallel, we will investigate the interaction between iPSC-derived macrophages with different trophoblast lineages during the course of infection using single-cell genomic and spatial methods.

Project description:Fertility depends on the coordination of complex interactions between male and female reproductive proteins inside the female reproductive tract (FRT). These interactions mediate a suite of changes in female behavior, morphology, and physiology after mating, yet little is known about how the molecular environment of the FRT may differ among species and coordinate species-specific female post-mating responses. We used semi-quantitative proteomics to compare the FRT protein composition between virgin and mated females in Drosophila melanogaster. These results are compared to those from quantitative TMT proteomic analyses of the mating-induced changes in D. simulans and D. mauritiana, and after conspecific and heterospecific inseminations. Our study highlights the value of using quantitative proteomics approaches to study the molecular composition of the FRT environment, and how its divergence may inform mechanistic studies of post-mating pre-zygotic reproductive isolation between species.

Project description:The present study aims at understanding differential gene-expression patterns in the female mouse brain tissues at different stages of reproductive life cycle Methods: Total RNA from brain tissues of 3, 12 and 40 week old female mice was subjected to cDNA and labelled cRNA synthesis and hybridized on 4X44k array. For the same samples, transcriptome sequencing was done followed by qPCR validation of the common genes. Data was analyzed using Gene spring GX 11.5 with MM10 reference along with GSEA and STRING analysis. We primarily focused on understanding gene expression changes in female mice brain which occur during various reproductive life stages viz. 3, 12, 40 weeks which is akin to adolescent, mature and pre-menopausal states.The genes involved in metabolic activity, neurological activity, inflammation and signalling along with genes involved in reproductive regulation show differential expression. The changes observed provide physiological insights in changes in the female brain associated with development, fertility and progressing age.
