An atlas of mRNA expression in pig adipose and muscle tissues

Project description:Obese and lean-type pig breeds show obvious differences in adipose deposition and muscle growth; however, the molecular mechanisms underlying this phenotypic variation remains unclear. Landrace (a leaner, Western breed), Rongchang (a fatty, Chinese breed) and Tibetan (a feral, indigenous Chinese breed that has not undergone artificial selection) pig breeds were used in this study. We collected eight diverse adipose tissues and two phenotypically distinct skeletal muscle tissues from three well-defined pig models with distinct fat rates, and studied mRNA expression differences among breeds, males and females, and tissues. These results highlight some possible candidate genes for porcine adipose deposition and muscle growth and provide some data on which to base further studies of the molecular basis of energy metabolism. The mRNA expression differences of eight diverse adipose tissues and two phenotypically distinct skeletal muscle tissues from three well-defined pig models with distinct fat rates are measured.

Identification and analysis of circRNAs in the prefrontal cortex of wild boar and domestic pig

Project description:Domestication caused significant differences in morphology and behavior between wild and domestic animals, and gene expression changes played an important role in this event. circRNA is a class of non-coding RNA that exerts a wide range of functions in biological processes through the regulation of gene expression. However, the regulatory role of circRNA in the process of domestication is still unclear. Here, we analyzed circRNA expression patterns in the prefrontal cortices of wild boar and domestic pig to determine the potential role of circRNAs in domestication. We identified a total of 11,375 circRNAs and found that 349 and 354 circRNAs were up-regulated in wild boar and Rongchang pig, respectively. This study lays the groundwork for exploring the regulatory role of circRNA in the process of domestication and provides new insights that contribute to further investigation of the molecular mechanism of pig domestication.

Phenotype heterogeneity and maturation of alveolar macrophages in the domestic pig

Project description:The macrophages of the airways are the first line of innate immune defense against inhaled pathogens. We showed that pig bronchoalveolar cells are composed of an heterogeneous alveolar macrophage (AM) population. The adherent AM non-adherent produce more TNF when stimulated with bacterial compound and are more efficient at phagocytosis than the non-adherent AM.The micro-array analysis revealed two different gene expression and would suggest that the non-adherent population would be more adapted to anti-viral responses. 5 Affymetrix Snowball microarray were analyzed from 1 pig breed (HAMSHIRE). Gene expression from non-adherent AM was compared to previous Array of Adherent AM from the same individuals (Series GSE45145). Cells were left untreated.
