Project description:We conducted prospective clinical studies to validate the importance of CD4+ T cells in 13 diseases from the following ICD-10-CM chapters: Neoplasms (breast cancer, chronic lymphocytic leukemia); endocrine, nutritional and metabolic diseases (type I diabetes, obesity); diseases of the circulatory system (atherosclerosis); diseases of the respiratory system (acute tonsillitis, influenza, seasonal allergic rhinitis, asthma); diseases of the digestive system (Crohn’s disease [CD], ulcerative colitis [UC]); and diseases of the skin and subcutaneous tissue (atopic eczema, psoriatic diseases). Study participants were recruited by clinical specialists based on diagnostic criteria defined by organizations representing each specialist’s discipline. Age and gender matched healthy controls (n = 127 and 39, respectively) were recruited in the Southeast region of Sweden from outpatient clinics at the University Hospital, Linköping; Ryhov County Hospital, Jönköping, a primary health care center in Jönköping; and a medical specialist unit for children in Värnamo. Study participants represented both urban and rural populations with an age range of 8–94 years. Patients with type I diabetes and obesity had an age range of 8–18 years. 12 patients had more than one diagnosis.

Project description:Diabetes microbiota study from a representative Saudi Population

Project description:The FINRISK cohorts comprise the respondents of representative, cross-sectional population
surveys that are carried out every 5 years since 1972, to assess the risk factors of chronic
diseases (e.g. CVD, diabetes, obesity, cancer) and health behavior in the working age
population, in 3-5 large study areas of Finland. DNA samples were collected in the following
survey years: 1987, 1992, 1997, 2002, 2007, and 2012. The MONICA and EHES (EU)
procedures were applied in phenotype collection (cf. MORGAM) and a wide spectrum of
laboratory tests was carried out from serum and plasma samples. Background information on
socioeconomic status, medical history, diet, exercise, measured anthropometric measures,
etc. was collected by questionnaires and during a clinical visit. Plasma/serum samples are
still available for the 2002-2012 cohorts. The cohort sizes are 6000-8800 per survey. The
cohorts have been followed up by linking them to the national hospital discharge register,
causes-of-death register and cancer register.
This project is an extension to previous efforts to build a catalogue of Finnish genome wide
data on population-based Finsrisk samples with rich phenotypic characterisations and health
registry link-up. These samples will extend the current Sequencing Initiative Suomi (SISu)
samples with a combination of genotyping using Illumina HumanCoreExome array and SISu-
based imputation. This will lead to high confidence common and low frequency variant
catalogue. The project will be funded by Aarno Palotie’s remaining faculty funds
complemented by Finnish funding from FIMM.

Project description:With increasing age of the global population, age-associated diseases such as cardiovascular diseases, cancer or diabetes are on the rise in many countries. To identify cellular processes and markers of aging in humans, this study performed comparative whole genome microarray gene expression profiling of peripheral blood mononuclear cells (PBMC) isolated from two age groups, i.e., an old age group (age: 75-89 years) and a younger, middle-aged group (age: 35-50 years). Total RNA was isolated from the PBMC and subjected to whole genome microarray gene expression profiling. The whole genome gene expression data of human PBMC reveal age-associated changes in transcript levels and contribute to a better understanding of biological processes underlying aging and the pathogenesis of age-associated diseases.

Project description:Obesity and its co-morbidities, such as diabetes and hypertension, can significantly reduce a person’s quality of life and place huge pressure on healthcare resources. When we eat a meal our gut and brain release hormones to control the amount of food and fluid we ingest to prevent overeating. One of these hormones is called glucagon-like peptide 1 (GLP-1) and is released from intestinal cells in response to food intake, but also produced and released in the brain. Drug analogues of GLP-1 are already in use in the clinic to treat both diabetes and obesity. The aim of this work was to obtain fundamental knowledge about a GLP-1 receptor population in nerve terminals of the posterior pituitary gland. We have investigated the pharmacological actions of GLP-1 using a selective receptor agonist called liraglutide, a drug that is approved for diabetes and obesity treatment in humans. Our work has focussed on the phosphoproteome of the neurointermediate lobe (posterior pituitary + intermediate lobe) of the rat pituitary gland 30 minutes after intraperitoneal injection of liraglutide (100 µg/kg) compared to vehicle controls (n = 6 animals per group). New understanding of this GLP-1 receptor population is essential for our knowledge of current treatments of diabetes and obesity that use stable peptide analogues in humans.

Project description:In several models of obesity-induced diabetes, increased lipid accumulation in the liver has been associated with decreased diabetes susceptibility. For instance, deficiency in leptin receptor (db/db) leads to hyperphagia and obesity in both C57BL/6 and C57BLKS mice but, only on the C57BLKS background do the mice develop beta-cell loss leading to severe diabetes while C57BL/6 mice are relatively resistant. Liver triglyceride levels in the resistant C57BL/6 mice are 3 to 4 fold higher than in C57BLKS. To better understand the mechanisms contributing to metabolic dysfunction in obesity-induced diabetes, we used microarrays to comprehensively profile gene expression livers of F2 mice (B57BL/6 X DBA/2) deficient in leptin receptor (db/db) DBA/2J females were mated to C57BL/6 males carrying leptin receptor deficiency (db/+) and, F1 (db/+) offspring were interbred to produce F2 mice. Offspring deficient in leptin receptor (db/db) were fed on a chow diet until 5 weeks or 12 weeks of age and then euthanized for collection of liver tissue for RNA profiling along with other diabetes-related phenotypes.

Project description:Insulin resistance is necessary but not sufficient for the development of type 2 diabetes. Diabetes results when pancreatic beta-cells fail to compensate for insulin resistance by increasing insulin production through an expansion of beta-cell mass or increased insulin secretion. Communication between insulin target tissues and beta-cells may initiate this compensatory response. Correlated changes in gene expression between tissues can provide evidence for such intercellular communication. We profiled gene expression in six tissues of mice from an obesity-induced diabetes-resistant and a diabetes-susceptible strain before and after the onset of diabetes. We studied the correlation structure of mRNA abundance and identified 105 co-expression gene modules. We provide an interactive gene network model showing the correlation structure between the expression modules within and among the six tissues. This resource also provides a searchable database of gene expression profiles for all genes in six tissues in lean and obese diabetes-resistant and diabetes-susceptible mice, at 4 and 10 weeks of age. A cell cycle regulatory module in islets predicts diabetes susceptibility. The module predicts islet replication; we found a strong correlation between ^2 H_2 O incorporation into islet DNA /in vivo/ and the expression pattern of the cell cycle module. This pattern is highly correlated with that of several individual genes in insulin target tissues, including IGF2, which has been shown to promote beta-cell proliferation, suggesting that these genes may provide a link between insulin resistance and beta-cell proliferation. Keywords: time course, mouse strain comparison, effect of obesity, Type 2 diabetes is a disorder that involves an increased demand for insulin brought about by insulin resistance, together with a failure to compensate with sufficient insulin production. Although Insulin resistance occurs in most obese individuals, diabetes is generally forestalled through compensation with increased insulin. This increase in insulin occurs through an expansion of beta-cell mass and/or increased insulin secretion by individual beta-cells. Failure to compensate for insulin resistance leads to type 2 diabetes. One way to understand the pathophysiology of diabetes is to examine the coordinate changes in gene expression that occur in insulin-responsive tissues and pancreatic islets in obese animals that either compensate for insulin resistance or progress to type 2 diabetes. In each case, there are groups of genes that undergo changes in expression in a highly correlated fashion. By identifying groups of correlated transcripts (gene expression modules) during the compensation and development of diabetes, we can gain insight into potential pathways and regulatory networks in obesity-induced diabetes. We study two strains of mice that differ in obesity-induced diabetes susceptibility. In this study, we surveyed gene expression in six tissues of lean and obese C57BL/6 (B6) and BTBR mice aged 4 wks and 10 wks. B6 mice remain essentially non-diabetic at all ages, irrespective of obesity. When obese, BTBR mice become severely diabetic by 10 weeks of age. By analyzing the correlation structure of the genes under three contrast conditions, obesity, strain, and age, we identified gene expression modules associated with the onset of diabetes and provide an interactive co-expression network model of type 2 diabetes. We found a key module that is comprised of cell cycle regulatory genes. In the islet, the expression profile of these transcripts accurately predicts diabetes and is highly correlated with islet cell proliferation.

Project description:The anti-diabetes activity of Rhizoma Coptidis was documented in the book âNotes of Elite Physiciansâ written by Hongjing Tao 1500 years ago. Berberine (BBR) is the major isoquinoline alkaloid constituent of the Chinese herb Rhizoma Coptidis, which has the beneficial characteristic of regulating glucose and lipid metabolism and has been extensively used in the treatment of obesity, diabetes and hypercholesterolemia. However, the effect of berberine on specific diabetes treatment targets is unknown. This study was conducted to elucidate the genome-wide gene expression profile in Zucker Diabetic Fatty Rats liver tissues in response to Berberine treatment.
