Expression information of splicing factor OsSKIPa knock-down and overexpressed rice

ABSTRACT: OsSKIPa is an SNW/SKIP protein in rice, which homolog to SKIP(human) and PRP45(yeast). OsSKIPa can rescue the lethal of yeast prp45 mutants. OsSKIPa-suppressed plants (S59R) show decrease of cell viability. Overexpression of OsSKIPa (S58S) increase the osmotic stresses tolerance. OsSKIPa is an important regulator and interacting with a lot of proteins which have varient functions. In order to find the genes expression information when OsSKIPa changed. Arabidopsis germination and two week growth on ionic vs osmotic stress

Project description:Arabidopsis Col-0 seeds were germinated and grown for two weeks on Arabidopsis thaliana salt media (ATS, control) or ATS media supplemented 50, 75, 100 or 125 mM NaCl that imposes both an ionic and osmotic stress; or ATS media supplemented with iso-osmolar concentrations of sorbitol (100, 150, 200 or 250 mM) that imposes only an osmotic stress. The aim of the study was to identify genes involved in plant growth and adaptation to ionic stress compared to genes involved in growth and adaptation to osmotic stress conditions. To do this we identified lists of genes that are differentially expressed in plants grown in NaCl (A) and lists of genes differentially expressed in plants grown in sorbitol (B). We then compared these lists to find ionic/salt-specific genes that are only expressed in plants grown in NaCl and not in plants grown in sorbitol; and osmotic genes that are expressed both in plants grown in NaCl and in plants grown in sorbitol. Comparative proteomic analysis of Arabidopsis plants treated with osmotic stress

Project description:To evaluate the differences in the acclimation of gcd1 gcd3 and WT plants to osmotic stress, we conducted an LC-MS/MS-based label-free quantitative proteomics analysis of WT and gcd1 gcd3 seedlings grown under mock conditions or osmotic stress imposed by 300 mM mannitol treatment. Comparative proteomic analysis of plants treated with osmotic stress reveals the basis of the underlying acclimation mechanism.

Expression data from NexCre:Atg7 conditional knockout mice brain

Project description:Transcriptome analysis of NesCre:Atg7 conditional knockout mice. Autophagy plays an important role in regulating protein metabolism and tissue homeostasis. Recent studies have reported that neural stem cell-specific Atg7 knockout mice (NesCre:Atg7f/f cKO mice) exhibit neonatal lethal due to severe neurodegeneration. However, the precise mechanisms of how neuronal fate is regulated by the autophagic pathway have not been elucidated. Here, we performed microarray experiments to analyze the changes in gene expression patterns in NesCre:Atg7 cKO mice. As a result, we could find a lot of candidate genes changed by Atg7 deficiency.

Multi-omics analysis of green lineage osmotic stress pathways unveils crucial roles of different cellular compartments

Project description:Maintenance of water homeostasis is a fundamental cellular process required by all living organisms. Here, we use the single-celled green alga Chlamydomonas reinhardtii to establish a foundational understanding of osmotic-stress signaling pathways through transcriptomics, phosphoproteomics, and functional genomics approaches. Comparison of pathways identified through these analyses with yeast and Arabidopsis allowed us to infer their evolutionary conservation and divergence across these lineages. 76 genes, acting across diverse cellular compartments, were found to be important for osmotic-stress tolerance in Chlamydomonas through their functions in cytoskeletal organization, potassium transport, vesicle trafficking, mitogen-activated protein kinase and chloroplast signaling. We show that homologs for five of these genes have conserved functions in stress tolerance in Arabidopsis and reveal a novel PROFILIN-dependent stage of acclimation affecting the actin cytoskeleton that ensures tissue integrity upon osmotic stress. This study highlights the conservation of the stress response in algae and land plants and establishes Chlamydomonas as a unicellular plant model system to dissect the osmotic stress signaling pathway.

Transcript profiling of transgenic rice lines where the OsMADS26 gene is over-expressed or down growing cultivated in standard or osmotic stress condition

Project description:Functional analyses of MADS-box transcription factors in plants have unraveled their role in major developmental programs (e.g; flowering and floral organ identity), in stress-related developmental processes such as abscission, fruit ripening and senescence and the role of some of them in stress response regulation was reported. The aim of this study was to decipher the genes that are under the control of the OsMADS26 transcription factor in rice in standard or osmotic stress condition. Transcriptome analysis in over-expressor and down-regulated OsMADS26 rice transgenic lines revealed that several major regulators of biotic and abiotic stress responses are mis-regulated compared to control plants, suggesting that OsMADS26 acts as an upstream negative regulator of stress-associated genes. Transgenic plants over-expressing or down-regulated (by RNA interference) for OsMADS26 were obtained and used to study the impact of this transcription factor on gene regulation in plants cultivated in standard or osmotic stress (100 mM manitol) condition.

Over-expression of bHLH122 in Arabidopsis

Project description:bHLH122 could be induced by salt, osmotic and drought stress except ABA, the transgentic Arabidopsis were more tolerant to abiotic stresses, such as drought and salt. What's more, in the overexpression plants, the endogenesis ABA contents were higher than WT. We found there existed an interaction between bHLH122 and CYP707A3 by virtue of EMSA and ChIP assays. We wanted to learn more about the molecular mechanism of bHLH122 and to explore what had changed in the over-expression plant through Genechips.
