Project description:We perceive the environment through an elaborate mental representation based on a constant integration of sensory inputs, knowledge, and expectations. Previous studies of astronauts on board the International Space Station have shown that the mental representation of space, such as the perception of object size, distance, and depth, is altered in orbit. Because the mental representations of space and time have some overlap in neural networks, we hypothesized that perception of time would also be affected by spaceflight. Ten astronauts were tested before, during, and after a 6-8-month spaceflight. Temporal tasks included judging when one minute had passed and how long it had been since the start of the workday, lunch, docking of a vehicle, and a spacewalk. Compared to pre-flight estimates, there is a relative overestimation for the 1-min interval during the flight and a relative underestimation of intervals of hours in duration. However, the astronauts quite accurately estimated the number of days since vehicle dockings and spacewalks. Prolonged isolation in confined areas, stress related to workload, and high-performance expectations are potential factors contributing to altered time perception of daily events. However, reduced vestibular stimulations and slower motions in weightlessness, as well as constant references to their timeline and work schedule could also account for the change in the estimation of time by the astronauts in space.

Project description:Background. While significant attention has been paid to the potential risk of pathogenic microbes aboard crewed spacecraft, the non-pathogenic microbes in these habitats have received less consideration. Preliminary work has demonstrated that the interior of the International Space Station (ISS) has a microbial community resembling those of built environments on Earth. Here we report the results of sending 48 bacterial strains, collected from built environments on Earth, for a growth experiment on the ISS. This project was a component of Project MERCCURI (Microbial Ecology Research Combining Citizen and University Researchers on ISS). Results. Of the 48 strains sent to the ISS, 45 of them showed similar growth in space and on Earth using a relative growth measurement adapted for microgravity. The vast majority of species tested in this experiment have also been found in culture-independent surveys of the ISS. Only one bacterial strain showed significantly different growth in space. Bacillus safensis JPL-MERTA-8-2 grew 60% better in space than on Earth. Conclusions. The majority of bacteria tested were not affected by conditions aboard the ISS in this experiment (e.g., microgravity, cosmic radiation). Further work on Bacillus safensis could lead to interesting insights on why this strain grew so much better in space.

Project description:The distance and duration of human spaceflight missions is set to markedly increase over the coming decade as we prepare to send astronauts to Mars. However, the health impact of long-term exposure to cosmic radiation and microgravity is not fully understood. In order to identify the molecular mechanisms underpinning the effects of space travel on human health, we must develop the capacity to monitor changes in gene expression and DNA integrity in space. Here, we report successful implementation of three molecular biology procedures on board the International Space Station (ISS) using a miniaturized thermal cycler system and C. elegans as a model organism: first, DNA extraction-the initial step for any type of DNA analysis; second, reverse transcription of RNA to generate complementary DNA (cDNA); and third, the subsequent semi-quantitative PCR amplification of cDNA to analyze gene expression changes in space. These molecular procedures represent a significant expansion of the budding molecular biology capabilities of the ISS and will permit more complex analyses of space-induced genetic changes during spaceflight missions aboard the ISS and beyond.

Project description:The GEROS-ISS (GNSS rEflectometry, Radio Occultation and Scatterometry onboard International Space Station) is an innovative experiment for climate research, proposed in 2011 within a call of the European Space Agency (ESA). This proposal was the only one selected for further studies by ESA out of ~25 ones that were submitted. In this work, the instrument performance for the near-nadir altimetry (GNSS-R) mode is assessed, including the effects of multi-path in the ISS structure, the electromagnetic-bias, and the orbital height decay. In the absence of ionospheric scintillations, the altimetry rms error is <50 cm for a swath <~250 km and for U10 <10 m/s. If the transmitted power is 3 dB higher (likely to happen at beginning of life of the GNSS spacecrafts), mission requirements (rms error is <50 cm) are met for all ISS heights and for U10 up to 15 m/s. However, around 1.5 GHz, the ionosphere can induce significant fading, from 2 to >20 dB at equatorial regions, mainly after sunset, which will seriously degrade the altimetry and the scatterometry performances of the instrument.

Project description:The "GENARA A" experiment was designed to monitor global changes in the proteome of membranes of Arabidopsis thaliana seedlings subjected to microgravity on board the International Space Station (ISS). For this purpose, 12-day-old seedlings were grown either in space, in the European Modular Cultivation System (EMCS) under microgravity or on a 1 g centrifuge, or on the ground. Proteins associated to membranes were selectively extracted from microsomes and identified and quantified through LC-MS-MS using a label-free method. Among the 1484 proteins identified and quantified in the 3 conditions mentioned above, 80 membrane-associated proteins were significantly more abundant in seedlings grown under microgravity in space than under 1 g (space and ground) and 69 were less abundant. Clustering of these proteins according to their predicted function indicates that proteins associated to auxin metabolism and trafficking were depleted in the microsomal fraction in µg space conditions, whereas proteins associated to stress responses, defence and metabolism were more abundant in µg than in 1 g indicating that microgravity is perceived by plants as a stressful environment. These results clearly indicate that a global membrane proteomics approach gives a snapshot of the cell status and its signaling activity in response to microgravity and highlight the major processes affected.

The findings of reduced T-cell function, cytokine imbalance, and increased stress hormones which preceded the event are detailed. Relevance for deep space countermeasures is discussed.
