Project description:Insulin binds the insulin receptor (IR), which in turn has showed to form nanoclusters at the cell membrane. Trying to exploit the nanoscale spatial organization of IR, we developed rod-like insulin-DNA-origami nanostructures carrying different numbers of insulin molecules. These structures (referred to as NanoRods, or NR) were then utilized to investigate receptor activity to spatial distribution of insulin molecules. One part of this investigation was to study the transcriptional response of brown adipocytes treated with NR bound to one insulin (NR-1) and NR bound to seven insulin (NR-7), and compare to untreated controls. Furthermore, free insulin was also used to ensure that similar pathways were activated when using NR-bound insulin.

Project description:Glucocorticoids are critical components of combination chemotherapy regimens in pediatric acute lymphoblastic leukemia (ALL). However, the signaling pathways regulating apoptosis in glucocorticoid-treated lymphoid cells remain unclear. In this study, pediatric ALL patient-derived xenograft inherently sensitive to glucocorticoids were exposed to dexamethasone in vivo. Whole-genome GR binding sites and histone acetylation status were detected using chromatin immunoprecipitation sequencing analyses. This provided a global understanding of dexamethasone-induced DNA modulations in ALL cells in vivo, which is likely to be important in the understanding of mechanisms of glucocorticoid response in lymphoid malignancies. One xenograft (ALL-54) was derived from a patient of dexamethasone-good responder. The xenograft was innoculated into 2 NOD/SCID mice, and treated with dexamethasone (15 mg/kg) or vehicle control. Binding of glucocorticoid receptor (GR), histone acetylation and IgG control in spleen-harvest xenograft samples were detected using ChIP-seq.

We found that IR affects 86 genes, including those specific to granulocyte (Lyz2 and MMP8) and nuclear architecture (Lmnb1 and Lbr). IR was associated with the decrease in protein levels measured by mass spectrometry (P=0.0015, binomial test). There was a significant overlap of IR between human and mouse (P=2.85E-22, hypergeometric test), showing that IR is conserved.Inhibition of NMD in granulocytes resulted in marked accumulation of 39/86 intron retaining mRNAs (P<0.05, RUV procedure with Holm-Bonferroni correction), indicating that IR triggers NMD to downregulate mRNA and protein expression.

Project description:Background: Preclinical studies showed thatmesenchymal stem cells (MSCs) ameliorate tau phosphorylation, amyloid-betaaccumulation, and inflammation in Alzheimer’s disease (AD) mouse models viasecretion of neurotrophic factors and cytokines. We aimed to identify CSFbiomarkers that can be used to predict or monitor the response to MSCs inpatients with AD. Methods: AD patients were injected withhuman umbilical cord blood-MSCs (n=22) or placebo (n=12). The cerebrospinalfluid (CSF) samples were collected at baseline, one day after the firstinjection, and one day after the third injection. The patients injected withMSCs were classified into good responder (GR) or poor responder (PR) groupsbased on the rate of changes in the ratio of total-tau and phosphorylated-tauin the CSF. We selected three typical participants in each group, and their CSFprotein levels were analyzed using liquid chromatography-tandem massspectrometry (LC-MS/MS). Results: In the LC-MS/MS analysis 1,667proteins were identified. 11 proteins showed significant differences betweenthe typical GR and PR at baseline. Based on their significance level and knownfunctions two proteins, reticulocalbin-3 (RCN3) and follistatin-related protein3 (FSTL3), were selected as potential biomarkers to predict MSC response. 173proteins showed significant change one day after the third injection comparedto the baseline in typical GR. We excluded 45 proteins that showed significantchange after the third injection compared to the baseline in the typical PR.Based on their significance level and known function, four proteins,scrapie-responsive protein 1 (SCRG1), neural proliferation and differentiationcontrol protein (NPDC1), apolipoprotein E (ApoE), and cystatin C (CysC), wereselected as potential biomarker to monitor MSC response. Additionally,functional analysis revealed that the increased CSF proteins after the thirdinjection compared to the baseline in the typical GR were associated withsynaptogenesis. Conclusions: This study identified twoproteins (RCN3 and FSTL3) that may be potential biomarkers for predicting MSCresponse and four proteins (SCRG1, NPDC1, ApoE, CysC) that may be potentialbiomarkers for monitoring MSC response in patients with AD. Further studies areneeded to validate our results.

NR blunts IFNg and IL-17 secretion with greater effects on TH17 polarization. RNA-seq analysis implicates NR blunting of sequestosome 1 (SQSTM1/p62)-coupled oxidative stress. NR administration increases SQSTM1 and reduces reactive oxygen species (ROS) levels. Furthermore NR activates NRF2, and genetic knockdown of NRF2 and of the NRF2-dependent gene, SQSTM1 diminish NR amelioratory effects. Metabolomic analysis identify that NAD+-boosting increases arginine and fumarate biosynthesis and genetic knockdown of argininosuccinate lyase ameliorates NR-effects on IL-17 production. Hence, NR via amino acid metabolites orchestrate NRF2 activation, augments CD4+ T cell antioxidant defenses and attenuates TH17 responsiveness. Oral NR supplementation in healthy volunteers similarly increase serum arginine, SQSTM1 and antioxidant enzyme gene expression and blunts TH17 immune responsiveness, supporting evaluation of NAD+-boosting in CD4+ T cell linked inflammation.

Project description:Glucocorticoid receptor (GR) was demonstrated to undergo an N/C interaction between the 26VMDFY30 motif in the N-terminus and C-terminal LBD in the presence of its co-chaperone cytoplasmic constitutive active/androstane receptor retention protein (CCRP). Utilizing the fact that phosphor-mimic mutation of tyrosine 30 to glutamic acid abolished this N/C interaction, CCRP and either GR Y30F or GR Y30E mutant was ectopically expressed in COS-1 cells for subsequent cDNA microarray analysis. Dexamethasone, a potent GR ligand, significantly induced or repressed 673 and 689 genes in Y30F and Y30E, respectively. Among them, only 166 genes were commonly regulated. This analysis strongly suggested that the GR N/C interaction serves to regulate distinct gene sets.
