Gene expression data from mouse primary stomach tissues

ABSTRACT: We investigated gene expression profiles of gastric cancer tissues from Pdx-1-Cre;Cdh1F/+;Trp53F/F;Smad4F/F mice (pChePS_GC), gastric cancer tissues from Pdx-1-Cre;Cdh1F/F;Trp53F/F mice (pCP_GC), and normal gastric epithelium (NGE) from Pdx1-1-Cre-negative mice. We used microarrays to detail the global programme of gene expression underlying gastric carcinogenesis and identified distinct classes of up and down-regulated genes during this process.

Gene expression analysis of pancreatic cancer tissues in mice

Project description:To examine effects of environmental risk factors on pancreatic cancer development, we fed control Pdx1-Cre (WT) and Pdx1-Cre; Kras-flox (Kras) mice with a Western alcohol diet containing high-cholesterol and high-saturated fat diet and 3.5% alcohol for 5 months. To enhance alcohol-induced pancreatic injury, we added a low dose of lipopolysaccharide (LPS, 1 μg/ml) to the diet. Mice were also given 7 hourly injections of cerulein (50 μg/kg) 4 times from the 1st to 4th month. To mimic tobacco smoking, we injected tobacco-derived carcinogen 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK, 100 mg/kg) 4 times from the 1st to 4th month. Pancreas tissues were collected from Pdx1-Cre (n=3, normal) and Pdx1-Cre; Kras-flox (n=6, all) mice. Kras-flox (Kras) mice were also fed a regular chow (n=3, control) or 3.5% alcohol diet (N=3, alcohol) for 5 months. RNA-seq analysis revealed induction of cancer-associated genes by these risk factors in the pancreatic cancer tissues.

Pancreatic cancer-induced cachexia syndrome

Project description:Cancer cachexia syndrome is observed in 80% of patients with advanced-stage cancer, and it is one of the most frequent causes of death. Severe wasting accounts for more than 80% in patients with advanced pancreatic cancer. Here we wanted to define, by using an microarray approach and the Pdx1-cre;LSL-KrasG12D;INK4a/arffl/fl, the pathways involved in muscle, liver and white adipose tissue wasting. The aim of our work was to characterize as extensively as possible the pathways activated by the pancreatic cancer-induced cachectic tissues. For this purpose, we generated and compared genome-wide expression profiles of white adipose tissue, skeletal muscle and liver, from Pdx1-cre;LSL-KrasG12D;INK4a/arffl/fl and LSL-KrasG12D;INK4a/arffl/fl mice at 10 weeks-old. Tissue samples by triplicate was obtained from liver, muscle and adipose tissues in both groups, controls and cachectic mice. Total RNA samples was processed and profiled on Affymetrix Mouse Gene 1.0 ST arrays as previously described (Cano et al, 2012)

Comparison of exon-wise expression profiling between normal and cancer tissues of human stomach

Project description:Gastric cancers account for the fourth most frequent cancer death worldwide. Although many differential gene expression profiles are reported for gastric cancers, their variation at the post-transcriptional level has not been provided yet. In this study, we compared the gene expressions of normal stomach vs. stomach cancer in an exon-wise manner and compared alternatively spliced transcripts. The RNA from normal and cancer tissues of gastric cancer patients were subjected to Exon 1.0 ST microarrays. Transcriptome analysis of RNAs from normal and cancer tissues of human stomach by exon array. We analyzed 30 pairs of normal-cancer stomach tissues using the Affymetrix Human Exon 1.0 ST platform. Array data was processed by the Affymetrix Exon Array Computational Tool.
