MiR-975 overexpression effect on Drosophila cell lines

ABSTRACT: We overexpressed the conspecific miR-975 in the cell lines derived from multiple Drosophila species (i.e. S2 cells from Drosophila melanogaster; ML82-19a cells from Drosophila simulans and WR-Dv-1 cells from Drosophila virilis). Comparison of the miR-975 effect between different cell lines provide insight into the rewiring of regulatory network through the evolution of new microRNA.

Project description:Transcriptional profiling of courtship song stimulated females in Drosophila melanogaster comparing females exposed to conspecific song to those exposed to either white-noise (control) or heterospecific song (D. simulans) Three condition experiment, D. melanogaster (mel_song) song stimulated vs. Control (no_song) and vs. D. simulans (sim_song) stimulated females. Biological replicates: 7 for D_mel_song, 4 for no_song, 3 for D_sim_song. One replicate per array. Each sample contains 120 pooled female heads, collected from three independent experiments. Technical replication: two arrays with reverse labeling for each contrast.

mRNA-Seq of whole flies from Drosophila

Project description:We sequenced mRNA from whole flies of females and male of Drosophila melanogaster, D. simulans, D. yakuba, D. ananassae, D. pseudobscura, D. mojavensis, and D. virilis as part of the modENCODE project to validate novel gene models found in D. melanogaster, identify genes differentially expressed between the sexes and sex-specific alternative splicing events, and examine the evolution of gene expression.

Quantitative proteomics reveals rapid divergence in the postmating response of female reproductive tracts among sibling species

Project description:Fertility depends, in part, on interactions between male and female reproductive proteins inside the female reproductive tract (FRT) that mediate postmating changes in female behavior, morphology, and physiology. Coevolution between interacting proteins within species may drive reproductive incompatibilities between species, yet the mechanisms underlying postmating-prezygotic isolating barriers remain poorly resolved. Here, we used quantitative proteomics in sibling Drosophila species to investigate the molecular composition of the FRT environment and its role in mediating species-specific postmating responses. We found that (1) FRT proteomes in D. simulans and D. mauritiana virgin females express unique combinations of secreted proteins and are enriched for distinct functional categories, (2) mating induces substantial changes to the FRT proteome in D. mauritiana but not in D. simulans, and (3) the D. simulans FRT proteome exhibits limited postmating changes irrespective of whether females mate with conspecific or heterospecific males, suggesting an active female role in mediating reproductive interactions. Our study suggests that divergence in the FRT extracellular environment and postmating response contribute to previously described patterns of postmating-prezygotic isolation and the maintenance of species boundaries.

Four lines (Ya19, Ya23, Ya24, Ya26) carry a D. simulans Y chromosome (from a Cameroon population) and four lines (Sec01, Sec03, Sec08, Sec27) carry a D. sechellia Y chromosome. All lines are otherwise identical with a D. simulans background (UCSD stock center line 14021-0251.092).

A global transcriptome analysis of human epidermal keratinocytes upon overexpression of microRNA-19a or microRNA-19b or microRNA-20a

Project description:Several members from microRNA 17-92 cluster, i.e. miR-19a, miR-19b and miR-20a, were found up-regulated in human epidermal keratinocytes at wound-edges compared to the intact skin; however their biological role in keratinocytes during wound repair has not been studied. To study the genes regulated by miR-19a, miR-19b and miR-20a, we transfected miRNA specific mimics, i.e. pre-miR-19a, pre-miR-19b or pre-miR-20a into human primary epidermal keratinocytes to overexpress them. We performed a global transcriptome analysis of keratinocytes upon overexpression of miR-19a or miR-19b or miR-20a using Affymetrix arrays.
