Derivation of kidney organoids from human pluripotent stem cells [RNA-Seq: Data Set 1]

ABSTRACT: We have developed a robust methodology for the derivation of kidney organoids from human pluripotent stem cells further vascularized taking advantage of several in vivo approaches. Our methodology suffices for the generation of kidney derivatives transcriptomically comparable with 22 weeks of gestation kidney tissues and highlight the use of longer periods of exposure to 3D in order to promote renal differentiation in a time frame of 15 days.

In order to ascertain the impact of extracellular matrix microenvironment in renal differentiation undifferentiated human pluripotent stem cells were further cultured in compliant substrates mirroring embryo-ECM microenvironment (1kPa) in order to promote renal differentiation and compared these conditions in front of rigid substrates (60 kPa) mimicking hard substrates normally used when differentiating human pluripotent stem cells

Thyroid progenitors are robustly derived from embryonic stem cells through transient, developmental stage-specific overexpression of Nkx2-1

Project description:The clinical importance of anterior foregut endoderm (AFE) derivatives, such as thyrocytes, has led to intense research efforts for their derivation through directed differentiation of pluripotent stem cells (PSCs). Here we identify transient overexpression of the transcription factor (TF) NKX2-1 as a powerful inductive signal for the robust derivation of thyrocyte-like cells from mouse PSC-derived AFE. This effect is highly developmental stage-specific and dependent upon FOXA2 expression levels and precise modulation of BMP and FGF signaling. The majority of the resulting cells express thyroid TFs (Nkx2-1, Pax8, Foxe1, Hhex) and thyroid hormone synthesis-related genes (Tg, Tpo, Nis, Iyd) at levels similar to adult mouse thyroid and give rise to functional follicle-like epithelial structures in Matrigel culture. Our findings demonstrate that NKX2-1 overexpression converts AFE to thyroid epithelium in a developmental time-sensitive manner and suggest a general methodology for manipulation of cell fate decisions of developmental intermediates.

Vascularized And Complex Organ Buds From Diverse Tissues Via Mesenchymal Cell-Driven Condensation

Project description:Here, we demonstrate a generalized method for organ bud formation from diverse tissues by combining pluripotent stem cell-derived tissue-specific progenitors or relevant tissue samples with endothelial cells and mesenchymal stem cells (MSCs). The MSCs initiated condensation within these heterotypic cell mixtures, which was dependent upon substrate matrix stiffness. Defining optimal mechanical properties promoted formation of 3D, transplantable organ buds from tissues including kidney, pancreas, intestine, heart, lung, and brain. Transplanted pancreatic and renal buds were rapidly vascularized and self-organized into functional, tissue-specific structures. These findings provide a general platform for harnessing mechanical properties to generate vascularized, complex organ buds with broad applications for regenerative medicine.
