Project description:Although there have been studies conducted on cornea and retina growth and development, postnatal gene expression studies on sclera growth during postnatal growth has not been well characterised. Given that the mouse genome has 85% homology to the human genome and has been completely sequenced, mouse model for the study of ocular growth has advantages over other animal models. Thus, we aimed to study the biology and genetics behind sclera growth during post-natal development in Balb/cJ mice as a means to understand genetic changes that cause scleral growth and development during post-natal eye development The purpose of this study was to identify the genes underlying the development of mouse sclera post-natal growth of the posterior chamber of the eye using microarray Total RNA was isolated from single cryogenically ground mouse sclera (n=30, 6 samples each from week 1-, 2-, 3-, 6-and 8-old mice), reverse-transcribed and hybridized onto a Affymetrix mouse gene 1.0 ST array.We sought to use microarray to determine the gene expression profiles of mouse sclera from different age groups and identify the developmental genes that are involved in postnatal ocular growth.

Project description:The optic nerve is a white matter tract that conveys visual information to the brain. The sclera comprises the white, protective outer layer of the eye. A characterization of the proteome of normal human retrobulbar optic nerve and sclera may facilitate studies of the eye. We conducted a proteomic analysis of optic nerve and sclera from five adults. Proteins were fractionated using SDS-PAGE. After in-gel digestion, peptides were analyzed using LC-MS/MS on an Orbitrap Elite mass spectrometer. We identified 2,711 non-redundant proteins in retrobulbar optic nerve and 1945 non-redundant proteins in sclera. Optic nerve proteins included proteins expressed by oligodendrocytes (laminin, proteolipid protein, fibronectin), myelin proteins (myelin basic protein, myelin-associated glycoprotein), and paranodal structural proteins (ankyrin β, spectrin). Sclera included 18 collagen protein chains, small leucine-rich proteoglycans (decorin, biglycan, lumican, keratocan, prolargin, fibromodulin, mimecan), non-collagenous glycoproteins (fibronectin, vitronectin, laminin), extracellular matrix proteins (thrombospondins 1-4, dystroglycan, transgelins 1-3), and integrins alpha-V, alpha-1 and 2, beta-1, -2, and -5. Twenty-one unambiguous protein isoforms were identified in optic nerve and ten unambiguous protein isoforms were identified in sclera. The mass spectrometry proteomics data have been deposited to the ProteomeXchange Consortium via the PRIDE partner repository with the optic nerve dataset identifier PXD001581.

Project description:The sclera is white, tough outer covering, known for providing structural support to the eyeball and the intraocular contents. Any abnormality in sclera results in disorders such as conjunctivitis, episcleritis, scleritis and hence a proper investigation will help in preventing the same. In the current study, we carried out an in-depth proteomic analysis of human sclera in which the proteins were extracted from the sclera tissue and subjected to both in-gel and in-solution digestion using trypsin as the proteolytic enzyme. The peptides were analyzed using Orbitrap Fusion Tribrid mass spectrometerand the data acquired was searched against Human RefSeq 75 protein database using SEQUEST HT and Mascot which resulted in the identification of 4,493 non-redundant proteins.The outcome of this study will facilitate in discovery of potential candidate biomarkers aiding in curbing the scleral disorders.

A detailed investigation of the proteome of the normal human sclera may provide insights into the biology and pathophysiology of the sclera. Project description:Introduction: In human eyes, ocular enlargement/ growth, reflects active scleral extracellular matrix remodeling. miRNAs are small non-coding RNAs that regulate gene expression by base pairing with target sequences, and serve as nodes of signaling networks. We hypothesized that the sclera, like most tissues, expresses miRNAs, some of which modulate genes regulating ocular growth. In this study, the scleral miRNA expression profile of rapidly growing human fetal eyes was compared with that of stable adult donor eyes using high-throughput microarray and quantitative PCR analyses. Results: Human sclera expressed several miRNAs. Microarray comparison of adult and fetal samples revealed many to be differentially expressed (p<0.01, min p= 6.5x10^11), with increased expression of collagen specific mir-214, let-7c, let-7e, mir-103, mir-107, and mir-98 in fetal sclera subsequently confirmed (1.5 to 4 fold changes, p<0.01). For both adult and fetal samples, no significant differences in miRNA expression profiles of sclera from posterior and peripheral ocular regions were observed. Conclusion: This is the first study to catalogue miRNA expression in human sclera. The sclera expresses several miRNAs, some of which show age-related differential regulation, higher in rapidly growing fetal eyes, consistent with a role in ocular growth regulation. These findings may be useful for linking scleral miRNA expression with potential manipulation in disorders such as scleral ectasia/ axial myopia.

Project description:Scleritis is a severe inflammatory ocular disorder with unknown pathogenesis. Using a mass spectrometry approach, we investigated healthy sclera and sclera affected by non-infectious scleritis for differentially expressed proteins. Therefore, we collected scleral samples of enucleated eyes due to severe non-infectious scleritis (n=3) and control scleral tissues (n=5), all exenterated eyes for eyelid carcinomas without scleral invasion. Nano LC-MS mass spectrometry identified 629 proteins within the healthy and diseased scleral tissues, of which collagen type I was the most abundantly expressed protein. Collagen type II-XII was also present. Filaggrin-2, a protein that plays a crucial role in epidermal barrier function, was upregulated in all scleritis cases. In addition, other epithelial-associated proteins were upregulated (such as Keratin 33b, 34, and 85, Epiplakin, transglutaminase-3, Galectin 7, and Caspase-14) in scleritis. Further, upregulated proteins involved in regulation of the cytoskeleton (Vinculin and Myosin 9) and housekeeping proteins were found (elongation factor-2 and cytoplasmic dynein 1). We selected two proteins for validation with immunohistochemistry: Fillagrin-2 and myosin 9. Upregulation of both proteins was confirmed, the latter protein showing co-localization with the endothelial cell marker ETC-related gene (ERG), indicating neovascularization in scleral tissue affected in scleritis. Further research, preferably with multiple scleritis cases, is needed to validate our findings, including identification of the specific role of Filaggrin-2 and angiogenesis in the pathogenesis of scleritis.
