We normalize this ratio to the ratio measured at time t=0h and transform into log2ratios. The experiment is carried out in triplicates, from three independant cultures.

Stability of meiosis-specific messenger RNA provides a novel paradigm for the regulation of meiosis

Project description:Compared to mitosis, much remains to be elucidated about molecular regulation of meiosis, although increasing light has been shed recently on its signal transduction, cell cycle regulation and chromosome dynamics. Here we show that some transcripts required exclusively for meiosis in fission yeast, such as mei4 mRNA encoding a transcription factor or rec8 mRNA encoding a cohesin subunit, are highly unstable if expressed during the mitotic cell cycle. These transcripts carry a cis-acting region responsible for the mitotic instability, which we name DSR. A YTH-family protein encoded by the mmi1 gene is essential for manifestation of their instability. Deletion of mmi1 impairs cell growth severely, apparently due to detriments brought by untimely expression of meiotic messages. Microarray analysis using a temperature-sensitive mmi1 mutant has revealed that at least a dozen of meiosis-specific transcripts are governed by the DSR-mmi1 system. Gene expression profiles of WT and ime4-/- mutant yeast cells, under vegetative and meiosis-inducing conditions

Project description:Inactivation of the yeast IME4 gene, the yeast homologue of METTL3, was shown to result in the loss of m6A in mRNA of mutant cells grown in sporulation medium. We attempted to characterize the effects of ime4 deletion on gene expression under vegetative and meiosis-inducing conditions. The results show that in vegetatively-growing ime4-/- cells there is an increased expression of the RME1 gene (repressor of meiosis) which prevents precocious entry into the meiotic program. Mutant yeast cells showed reduced expression levels of genes involved in ribosome biogenesis and gene expression processes. Surprisingly, despite the fact that a diploid strain was analyzed, there was also a striking change in the expression level of haploid-specific genes, suggesting that RNA methylation may be used to enforce the sexual identity of diploid cells, required for the implementation of the gametogenesis program. Consistently, when cells were induced to undergo meiosis, ime4-/- diploids failed to undergo the meiotic divisions. Among the genes showing reduced expression in the mutant were IME1 and IME2, the two known inducers of meiosis. Thus, the yeast IME4 gene plays an important role in the regulation of the developmental switch from vegetative cells into gametogenesis.

High-resolution mapping reveals a conserved, widespread, dynamic mRNA methylation program in yeast meiosis

Project description:N6-methyladenosine (m6A) is the most ubiquitous mRNA base modification, but little is known about its precise location, temporal dynamics, and regulation. Here, we generated genomic maps of m6A sites in meiotic yeast transcripts at nearly single-nucleotide resolution, identifying 1,308 putatively methylated sites within 1,183 transcripts. We validated 8/8 methylation sites in different genes with direct genetic analysis, demonstrated that methylated sites are significantly conserved in a related species, and built a model that predicts methylated sites directly from sequence. Sites vary in their methylation profiles along a dense meiotic time-course, and are regulated both locally, via predictable methylatability of each site, and globally, through the core meiotic circuitry. The methyltransferase complex components localize to the yeast nucleolus, and this localization is essential for mRNA methylation. Our data illuminates a conserved, dynamically regulated methylation program in yeast meiosis, and provides an important resource for studying the function of this epitranscriptomic modification.
