Next generation sequencing based transcriptome comparison between wild-type Drosophila and an Alzheimer's disease model

ABSTRACT: Gene expression profiling of wild-type (WT) fruit flies and transgenic flies that express human amyloid precursor protein was performed to investigate how Aβ42 affects the transcriptome of Drosophila.

Quantitative proteomics unravels molecular networks of neuroprotection conferred by Brahmi extract in Aβ42 -induced toxicity model of Drosophila melanogaster

Project description:Ayurvedic drug formulations Bacopa monnieri and Centella asiatica are known to have neuroprotective effects. These have been traditionally used in the treatment of Alzhemeir’s disease, and other neurological deficits. Using pan neuronal Aβ42 model of Drosophila melanogaster, a mass spectrometry based quantitative proteomic analysis platform was used to generate the data on proteins altered in response to the Aβ42 toxicity and restoration of altered proteins by consumption of aqueous extracts of two Ayurvedic drug formulations Bacopa monnieri and Centella asiatica aqueous extract. Quantitative proteomic analysis resulted in 0.67 million mass spectra corresponding to 2,59,168 peptide-spectrum matches (PSM) mapping to 24,305 non- redundant peptides corresponding to 11,480 Drosophila melanogaster proteins. Proteins were filtered for >3 PSMs, resulting in 9,540 proteins. Flies expressing Aβ42 significantly altered 517 proteins which were involved in maintaining essential neuronal functions. Supplementing flies with Bacopa monnieri or Centella asiatica extract commonly rescued 224 proteins from Aβ42 toxicity, moreover, extract supplemented group significantly altered proteins which were additionally supporting neuronal maintenance in flies with Aβ42 stress.

Measuring ageing-dependent transcriptional responses to human Aβ42 peptide in Drosophila brain

Project description:We have shown previously that older flies are intrinsically more susceptible to Aβ42 toxicity. Building upon these findings, this study aimed to determine the mechanisms by which ageing increases this vulnerability to damage in the brain. A fixed dose of Aβ42 peptide was induced in young (5d) versus older (20d) fly neurons, and then gene and protein expression changes examined in dissected fly brains using microarray analyses. This unbiased approach has revealed genes and pathways that correlate with increased susceptibility of the ageing brain to proteotoxicity.

Dynamic changes in the brain protein interaction network correlates with progression of Aβ42 pathology in Drosophila

Project description:Alzheimer’s disease (AD), the most prevalent form of dementia, is a progressive and devastating neurodegenerative condition for which there are no effective treatments. Understanding the molecular pathology of AD during disease progression may identify new ways to reduce neuronal damage. Here, we present a longitudinal study tracking dynamic proteomic alterations in the brains of an inducible Drosophila melanogastermodel of AD expressing the Arctic mutant Aβ42 gene. We identified 3093 proteins from flies that were induced to express Aβ42 and age-matched healthy controls using label-free quantitative ion-mobility data independent analysis mass spectrometry. Of these, 228 proteins were significantly altered by Aβ42 accumulation and were enriched for AD-associated processes. Network analyses further revealed that these proteins have distinct hub and bottleneck properties in the brain protein interaction network, suggesting that several may have significant effects on brain function. Our unbiased analysis provides useful insights into the key processes governing the progression of amyloid toxicity and forms a basis for further functional analyses in model organisms and translation to mammalian systems

Project description:We found some novel miRNA that targets SIRT1 may contribute to the pathogenesis in AD mouse model. We first identified that SIRT1 is significantly reduced in Alzheimer patient`s precentral gyrus and 5XFAD mice. To determine whether the strategy of inhibiting some miRNA affects AD pathology, we synthesized antisense oligonucleotides of miRNA (miRNA ASO) and studied this sequence in the brain of 5XFAD through direct intracerebral ventricular injection using stereotaxic instrument. We identified miRNA ASO significantly reduced amyloid beta plaque and amyloid production enzyme. Importantly, the attenuation of amyloid beta plaques through miRNA ASO was caused by a phagocytic activity of glial cells, by which it can directly target CD36. miRNA ASO also decreases inflammatory responses. These results inhibit neuronal loss caused by amyloid beta and leading to improvement of cognitive impairment. These insights of miRNA ASO suggests as therapeutic scope against AD pathology.

Effect of Bacteroides fragilis administration on microglia in WT and the APP-PS1 model of Alzheimer's disease [Bfrag]

Project description:The gut microbiota and innate immune system play critical roles in Alzheimer’s disease (AD). Bacteroides is elevated in AD patients and correlates with cerebrospinal fluid levels of Aβ and tau. We found that increased amyloid-β (Aβ) plaques in Bacteroides fragilis treated APP/PS1-21 mice were associated with altered cortical expression Aβ processing genes. B. fragilis suppressed peripheral CD4+ T cell production of GM-CSF and IL-4 and transcriptional changes in microglia related to GM-CSF and IL-4 signaling, phagocytosis, and protein degradation. Furthermore, B. fragilis impaired the microglial uptake of intracranially injected Aβ42, whereas Erysipelotrichaceae strains increased uptake. Depleting murine Bacteroidetes with metronidazole decreased amyloid load in aged 5xFAD mice, increased CD4+ T cell GM-CSF production, and activated microglial pathways related to cytokine signaling, phagocytosis and lysosomal degradation. These data suggest that the gut microbiome may contribute to AD pathogenesis by suppressing peripheral cytokines and microglia phagocytic function, leading to impaired immune-mediated Aβ clearance.

A diet-induced type 2 diabetes model in Drosophila

Project description:We showed in this present study that HSD feeding could induce type 2 diabetes-like phenotype in fruit flies, resembling the key disease features including hyperglycemia, glucosuria, increased hunger and thirsty, unexplained weight loss, and reduced female fecundity. We also showed that HSD-fed flies shared similar molecular mechanisms underlying type 2 diabetes patients, including both insulin deficiency and insulin resistance. Last but not least, disrupting insulin signaling in fruit flies could mimic the effect of HSD feeding to induce type 2 diabetes-like phenotype, further confirming that these phenotypes were a result of compromised insulin signaling upon HSD feeding. Taken together, we demonstrated that this diet-induced type 2 diabetes model in fruit flies rightly and comprehensively recapitulated the physiological, metabolic, behavioral, and molecular features of human type 2 diabetes, offering a valid and accessible model to study the underlying mechanism of this disease and to validate potential diagnostics and therapeutic tools. Given the highly conserved regulatory framework of physiology and metabolism between flies and human, our work has paved a way to generate novel insights into type 2 diabetes, a common, severe, and yet poorly understood disease in human.
