Project description:Largemouth bass (Micropterus salmoides) (LMB) inhabiting Lake Apopka, Florida are exposed to high levels of organochlorine pesticides (OCPs) that exert effects through multiple modes of action. The objectives of this study were to assess the impacts of two OCPs (p,p'-Dichlorodiphenyldichloroethylene; p,pM-bM-^@M-^Y DDE and methoxychlor; MXC) on the reproductive axis of LMB. Female LMB were fed coated feed over 84 days of either 125 (p,pM-bM-^@M-^Y DDE), 10 (MXC), or 0 (control) ppm (mg/kg). In addition, due to the fact that p,pM-bM-^@M-^Y DDE and MXC are suspected anti-androgens, the potent anti-androgenic pharmaceutical flutamide (FLUT) was also fed to a separate group of LMB at 750 ppm. After 84 days, LMB significantly incorporated p,pM-bM-^@M-^Y DDE and MXC into muscle and ovary tissue. Gonadal histology and plasma sex steroids were not significantly different than controls after being fed contaminants. Microarray analysis revealed that p,pM-bM-^@M-^Y DDE and FLUT regulated more genes in common that either chemical compared to MXC, consistent with data suggesting that p,pM-bM-^@M-^Y DDE is a more potent anti-androgen than MXC. There was a significant positive correlation between 71 commonly regulated genes in all three treatments, suggesting that these transcripts may be responding to a common anti-androgenic mode of action. Cell processes affected by all three treatments included leukocyte cell adhesion, ossification, platelet function and inhibition, xenobiotic metabolism, fibrinolysis, and thermoregulation. Most striking was that all three chemicals depressed gene networks related to immune function and inflammation. Project description:The increase of aged population makes it necessary to find effective prevention and treatment procedures to help the elderly to enjoy a healthy and long life. The organochlorine compound p,p´-DDE (1,1-dichloro-2,2-bis(p-chlorophenyl) ethylene) is a DDT metabolic byproduct that generates great concern because its deleterious effects on both human and environment health. It is an endocrine disruptor agent and causes alterations in immune response, reproductive system, development, nervous system and endocrine homeostasis. Nutritional interventions may improve health and prolong life. A transcriptional study was carried on in order to analyze the biological responses caused by p,p´-DDE exposure in the liver of aged Mus spretus mice. Data showed that the elderly are especially sensitive to the toxicity of environmental contaminants. This work also evaluated the possible hepatoprotective effect of including sun-dried Pedro Ximenez must in the diet when p,p´-DDE exposure happens simultaneously. Our results showed that this dietary intervention controls the hepatic metabolism of carbohydrates and lipids through restoring bile acid homeostasis, improving the overall health in the elderly.

Project description:Evidence that persistent environmental pollutants may target the male reproductive system is increasing. The male reproductive system is regulated by secretion of testosterone by testicular Leydig cells, and perturbation of Leydig cells function may have ultimate consequences. 3-methylsulfonyl-DDE (3-MeSO2-DDE) is a potent adrenal toxicants formed from the persistent insecticide DDT. Although studies have revealed endocrine disruptive effect of 3-MeSO2-DDE, the underlying mechanisms at cellular level in steroidogenic Leydig cells remains to be established. The current study addresses the effect of 3-MeSO2-DDE viability, hormone production and proteome response of primary neonatal porcine Leydig cells. The AlamarBlue™ assay was used to evaluate cell viability. Solid phase radioimmunoassay was used to measure concentration of hormones produced by both unstimulated and luteinizing hormone (LH)-stimulated Leydig cells following 48 h exposure. Protein samples from Leydig cells exposed to a non-cytotoxic concentration of 3-MeSO2-DDE (10µM) were subjected to nano-LC-MS/MS and analyzed on a Q Exactive mass spectrometer and quantified using label-free quantitative algorithm. Gene Ontology (GO) and Ingenuity Pathway Analysis (IPA) were carried out for functional annotation and identification of protein interaction networks. 3-MeSO2-DDE regulated Leydig cell steroidogenesis differentially depending on cell culture condition. Whereas its effect on testosterone secretion at basal condition was stimulatory, the effect on LH-stimulated cells was inhibitory. From triplicate experiments, a total of 7540 proteins were identified in which the abundance of 87 proteins in unstimulated Leydig cells and 146 proteins in LH-stimulated Leydig cells were found to be significantly regulated in response to 3-MeSO2-DDE exposure. These proteins not only are the first reported in relation to 3-MeSO2-DDE exposure, but also display small number of proteins shared between culture conditions, suggesting the action of 3-MeSO2-DDE on several targeted pathways, including mitochondrial dysfunction, oxidative phosphorylation, EIF2-signaling, and glutathion-mediated detoxification. Further identification and characterization of these proteins and pathways may build our understanding to the molecular basis of 3-MeSO2-DDE induced endocrine disruption in Leydig cells.

Project description:The consistent cold temperatures and large amount of precipitation in the Olympic and Cascade ranges of Washington State are thought to increase atmospheric deposition of contaminants in these high elevation locations. Total mercury and 28 organochlorine compounds were measured in composite, whole fish samples collected from 14 remote lakes in the Olympic, Mt. Rainer, and North Cascades National Parks. Mercury was detected in fish from all lakes sampled and ranged in concentration from 17 to 262 ug/kg wet weight. Only two organochlorines, total polychlorinated biphenyls (tPCB) and dichlorodiphenyldichloroethylene (DDE), were detected in fish tissues (concentrations <25 ug/kg wet weight). No organochlorines were detected in sediments (MRL ≈1-5 ug/kg), while median total and methyl mercury in sediments were 30.4 and 0.34 ug/kg (dry weight), respectively. Using a targeted rainbow trout cDNA microarray with known genes, we detected significant differences in liver transcriptional responses, including metabolic, endocrine, and immune-related genes, in fish collected from a contaminated lake compared to a lake with a lower contaminant load. Overall, our results suggest that local urban areas are contributing to the observed contaminant patterns, while the transcriptional changes point to a biological response associated with exposure to these contaminants in fish. Specifically, the gene expression pattern leads us to hypothesize a role for mercury in disrupting the metabolic and reproductive pathways in fish from high elevation lakes in western Washington. Project description:Coastal monitoring in the 21st century faces challenges as measuring exposure using traditional toxicity approaches is not practical or financially feasible for the numerous contaminants entering oceans. Transcriptomics, high-throughput assays, and adverse outcome pathways (AOP) have been one approach to overcome these obstacles but development of these methods is challenging for non-model organisms and contaminants with unknown mechanisms. Endocrine Disrupting Compounds (EDCs) have been found to cause reproductive impairments such as intersex in bivalves, however, the mechanism linked to this adverse outcome (AO) is unknown as the estrogen receptor (ER) has been identified to be unresponsive to EDCs. To develop mechanism-based biomarkers that may be linked through an AOP we exposed Mytilus edulis to 17-alpha-ethinylestradiol (5 and 50ng/L) and 4-nonylphenol (1 and 100μg/L), and performed gene expression analysis on digestive gland tissue using a M. edulis microarray. Through network and targeted analyses, we identified the non-genomic estrogen signaling pathway and steroidogenesis pathway as the likely mechanism of action and identified biomarkers and potential key events (KE) for an AOP of endocrine disruption in marine mussels. The mechanism-based biomarkers were validated in the laboratory, tested in the field, and were responsive in areas known to be contaminated with EDCs.

Project description:Although autism spectrum disorder (ASD) is among the most heritable of neurodevelopmental disorders, the rapidly rising prevalence of ASD suggests that environmental factors may interact with genetic risk for ASD. Environmental factors may impact both gene expression and phenotypes in ASD through epigenetic modifications that, in turn, could lead to intergenerational effects influencing risk for ASDs. Endocrine disrupting compounds (EDCs), such as the long-lived organochlorines, are of particular interest with respect to risk for autism because of their ability to interfere with sex hormones that have been implicated in the regulation of RORA, a dysregulated gene in ASD that is a master regulator of many other ASD risk genes. The specific aims of this study are to: 1) investigate whether high versus low exposures to the persistent organochlorine 1,1-dichloro-2,2-bis(p-chlorophenyl)ethylene (DDE) are associated with differentially methylated regions (DMRs) in sperm from a Faroese cohort whose natural diet of pilot whale meat and blubber exposes them to higher than average levels of organic pollutants; 2) determine if genes associated with DDE DMRs are enriched for ASD risk genes; 3) identify pathways and functions over-represented among genes associated with DMRs. Whole genome bisulfite sequencing (WGBS) was used to identify genome-wide DMRs in sperm from individuals divided by high and low exposure levels. Gene ontology and pathway analyses were used to determine enrichment in functional relationships to ASD. Genes in DMRs not only could discriminate between high and low exposures to DDE, but also were enriched in autism risk genes. Gene ontology and pathway analyses of these genes show significant enrichment for neurodevelopmental processes frequently impacted by ASD. Results of this study show that elevated exposure to certain organochlorines is associated with genome-wide DNA methylation patterns in sperm affecting genes involved in neurological functions and developmental disorders, including ASD.

Project description:In this study, both male and female sexually regressed largemouth bass (Micropterus salmoides) (LMB) were fed a nominal concentration of 3.0 mg dieldrin/kg in feed for 60 days. A third group of male LMB was fed both 3.0 mg dieldrin/kg dieldrin and 0.7 mg E2/kg in feed. E2 was used as a model compound to elicit estrogenic effects via ER signaling. There were four samples analyzed for 1) control males 2) control females 3) males fed 3 mg/kg dieldrin 4) females fed 3.0 mg/kg dieldrin, and 5) males fed 3.0 mg/kg dieldrin + 0.7 mg/kg E2. There was a total of 20 microarrays and sample processed.
