Gene expression in epigonadal white adipose tissue of LDKO and LTKO mice

ABSTRACT: We compared RNA expression profiles of eWAT obtained from Cntr- and LDKO-mice, and Cntr3- and LTKO-mice. Compared against the controls, the expression of 82 genes decreased, and 83 genes increased more than 2-fold in eWAT of LDKO-mice, most of which returned to normal expression in eWAT of LTKO-mice

Crtc1 deficiency aggravates fat accumulation in the white adipose tissue

Project description:Obesity is described as excessive fat accumulation that drives the development of glucose and lipid metabolism disorders, which is linked to multiple diseases. Crtc1, known as a transducer to regulate Creb activity, plays an important role in several basic physiological functions. Previous studies have shown visible hyperappetite and obesity in Crtc1 knockout (Crtc1-/-) mice. To investigate the effect of Crtc1 on fat accumulation in different organs, we generated Crtc1-/- mice by CRISPR/Cas9 system and regarded Crtc1+/+ as control under the normal feeding conditions. Compared with Crtc1+/+ mice, Crtc1-/- mice exhibited increase of bodyweight which was resulted from the abnormal expansion of white adipocyte. In addition, Crtc1-/- mice were more prone to hyperglycemia and dyslipidemia, supported by the levels of plasma glucose and FABP4. The results of RNA-seq and qRT-PCR in liver and epididymal white adipose tissue (eWAT) showed that the fat accumulation caused by Crtc1 deletion was mainly related to the lipid metabolism of adipose tissue, not liver. Moreover, the up-regulation of lipid metabolism in eWAT induced by Crtc1 deficiency was closely related to the up-regulation of Pparγ signaling pathway. Our findings suggested that endogenous Crtc1 had a protective role in obesity development and Crtc1 deficiency aggravates the progression of fat accumulation and related co-morbidities, which introduced a new insight for treatment of obesity.

Dietary intake of antioxidant curcumin reduces eIF2 phosphorylation and diacylglycerol and glycerolipid contents in white adipose tissue of obese mice

Project description:To elucidate the bioactive property of the dietary antioxidant curcumin, we examined tissue distribution and the gene expression- and lipidomic-profiles in epididymal white adipose tissue (eWAT) of the diet-induced obese mice. Dietary intake of curcumin (0.1% W/W) didn’t affect the eWAT weight and the plasma lipid levels but reduced the levels of lipid peroxidation marker in eWAT. Curcumin was a slightly accumulated in eWAT and altered the gene expression associated with eukaryotic translation initiation factor 2 (EIF2) signaling. Curcumin suppressed the endoplasmic reticulum (ER) stress-related eIF2 phospholyration, the accumulation of macrophages and the expression of oxidative stress-sensitive transcription factor NF-κB p65 and leptin, whereas anti-inflammatory effect wasn’t enough to reduce the TNF-α and IFN-γ levels. Lipidomic- and gene expression analysis suggests that curcumin reduced the contents of some diacylglyverols (DAGs) and DAG derived glycerophospholipids by suppressing the expressions of lipogenesis-related glycerol-3-phosphate acyltransferase 1 and lipolysis-related adipose triglyceride lipase.

The white adipose tissue transcriptional response to withdrawal of vitamin B3

Project description:Distinct markers for early, mild vitamin B3 deficiency are lacking. To identify these, we examined the molecular responses of white adipose tissue to vitamin B3 withdrawal. We performed a dietary intervention in male C57Bl/6JRcc mice. A diet with a low but adequate level of tryptophan without nicotinamide riboside (NR) was compared to the same diet with NR at the recommended vitamin B3 (30 mg NR per kg diet). Physiological and circulating parameters were determined and global transcriptomics, qRT-PCR and histology of epididymal white adipose tissue (eWAT) were done. We observed a decreased insulin sensitivity and a shift from carbohydrate to fatty acid oxidation. This was consistent with molecular changes in eWAT, where we observed an altered MEK/ERK signalling, a lowering of glucose utilization markers and an increase in makers of fatty acid catabolism, which may be related to the consistent reduction of mitochondrial OXPHOS Complex I (mRNAs and protein). The synthesis pathway of tetrahydropteridine (BH4), an essential cofactor for neurotransmitter synthesis, was found to be increased. Based on our results, we propose the technically validated downregulation of Anp32a, Tnk2 and the upregulation of Mapk1, Map2k1, Mthfs, Mthfsl and Qdpr as a WAT transcriptional signature marker for mild vitamin B3 deficiency.

Cx3cl1 expression in the adipose tissue is downregulated during development of obesity

Project description:To assess changes in expression level of various chemokines and their receptors on diet-induced obesity, we analysed gene expression in adipose tissue of C56BL/6J mice fed a high-fat (HF) diet or normal chow diet for 8 weeks. HF diet-induced obese (DIO) mice showed adipose tissue inflammation and insulin resistance. Comprehensive gene expression analysis showed that MCP-1–CCR2 and CCL5–CCR5 signalling in epididymal white adipose tissue (eWAT) were enhanced during the development of obesity. Surprisingly, the gene expression of Cx3cl1 was decreased in the eWAT of DIO mice compared with lean mice. While Cx3cr1 expression showed no significant difference between DIO and lean mice. Decreased CX3CL1-CX3CR1 signalling in adipose tissue may also be involved in the development of obesity-induced adipose tissue inflammation and insulin resistance.
