Sex of Blood

ABSTRACT: Blood samples were taken at 1:00 pm +/- 1 h (Potassium-EDTA-monovettes, Sarstedt, Numbrecht, Germany) to control for circadian influences on gene expression. Blood was taken from normal male controls, normal female controls as well as different patients with disorders of sex development leading to a dissection of sex chromosomes and external genital phenotype. All samples were analyzed by a blood cell counter (Beckmann-Coulter, Krefeld, Germany) and white blood cell counts did not differ between the normal males and the normal females (by t-test). PBMCs were isolated from whole blood using PBS (pH 7.2) and Ficoll (Biochrom, Berlin, Germany) gradient centrifugation. Comparative proteomics analysis of microalgae Prototheca associated with severe, mild and non-infectious strains

Project description:Three strains, P. zopfii genotype 1 (SAG 2063), P. zopfii genotype 2 (SAG 2021) and P. blaschkeae (SAG 2064) from the culture collection at the Institute of Animal Hygiene and Environmental Health, Freie Universität Berlin, Germany were cultured six times independently at 37°C in Sabouraud dextrose liquid medium until OD600 measurement reached 0.6 to 0.9. Following the cell harvest by brief centrifugation, the cells were lysed and label free quantitative proteomics analysis were carried out to investigate the differences between these strains.

Normal blood cell subtypes

Project description:We used antibodies coupled to magnetic beads (MACS) to purify cell types out of whole blood from healthy donors (CD4+ T cells, CD8+ T cells, and B cells). We also profiled whole blood (collected in Pax tubes) and Peripheral Blood Mononuclear cells obtained by Ficoll-Paque based separation. RNA was extracted using Trizol, amplified linearly twice, labeled with Cy5 and co-hybridized with amplified Stratagene human reference (labeled with Cy3). Molecular profiling of myeloid leukemia cell lines

Project description:Cell lines. The myeloid cell lines CMK, HEL, HL-60, K-562, KASUMI-1, KG-1, LAMA-84, M-07e, MONO-MAC-1, MV4-11, NB-4, OCI-AML2, OCI-AML5, SIG-M5, THP-1, and UT-7 were obtained from the DSMZ in Braunschweig, Germany (http://www.dsmz.de). The cell line ME-1 was kindly provided by Dr. Ikuya Sakai, Ehime University, Japan. Cell lines were cultured under standard conditions in RPMI 1640 containing 10% FCS, 2 mM L-glutamine, 100 units/ml penicillin and 100 units/ml streptomycin. For subsequent DNA and RNA isolation 1x107 cells of the respective cell lines were washed twice in phosphate-buffered saline, and stored as cell pellets at 800C. Gene expression profiling. Gene expression profiling (GEP) was performed essentially as described using the previously described cDNA microarray platform (Bullinger et al., N Engl J Med 2004;350:1605-1616). Microarray Coverage of Universal Human Reference RNA on microarrays with yeast controls

Project description:Microarray coverage of UHRR was determined by counting the number of spots with intensities exceeding the 1% and 5% false positive rates defined using the yeast negative control spots. Seven microarrays from different print runs were used. In four experiments, the UHRR was labeled with Cy5 and in three experiments with Cy3.
