Single cell RNA sequencing of cultured CCR10+ and CCR10- lung airway epithelial cells

ABSTRACT: Mechanically dissociated IPF lung explant derived cellular suspensions were cultured in 50% senescent lung fibroblast conditioned medium + 50% fibroblast complete medium (DMEM + 15% FBS + antiboitics) and 10 µM of Y27632. After approximately 2 weeks, cells were passaged and cultured overnight in Pneumacult Ex Plus medium (STEMCELL technologies).

Project description:Trihydroxyphenolic compounds, such as Epigallocatechin Gallate (EGCG), cause fibroblast-specific TGFb1 receptor kinase and lysyl oxidase like 2 (LOXL2) inhibition, reducing fibroblast activation and collagen cross-linking in fibrotic mice lungs and in cultured human precision cut lung slices (PCLS) derived from Interstitial Lung Disease (ILD) / Idiopathic Pulmonary Fibrosis (IPF) patients. When taken for two weeks, EGCG reduced protein markers of pro-fibrotic signaling in spare tissue from diagnostic surgical biopsies from ILD patients. Here, we report the result of single-cell RNA sequencing on lung tissue from ILD patients who took EGCG (600mg by mouth daily) for two weeks prior to diagnostic surgical biopsy and donated spare tissue and from control/untreated biopsy samples, including comparison to normal donor and end-stage ILD explant lung tissues.

Project description:Studies of human fetal lung in explant culture and in isolated epithelial cells have demonstrated that both glucocorticoids and cyclic AMP promote differentiated alveolar type II cell phenotype as assessed by ultrastructural morphology and surfactant production. This project profiles changes in gene expression associated with hormone induced differentiation. Undifferentiated human fetal lung (13-20 wk) epithelial cells were cultured in serum-free medium (control) or with dexamethasone/8-Bromo cyclic AMP/isobutylmethylxanthine (DCI) to promote type II cell differentiation. RNA from five sets of experiments (10 samples) was evaluated using the U133A Affymetrix GeneChip set. Keywords: Hormone treatment

scRNA-seq of canalicular stage human fetal lung cultured under air-liquid interface

Project description:Small pieces (<0.5 cm of diameter) were isolated from distal regions of 125 day male human fetal lung and cultured on floating poly-carbonate membranes on top of serum and growth factor free medium under air-liquid interface conditons (ALI). Next generation sequence analysis of cells derived from mouse hair follicles after long term culture

Project description:Follicle epithelial stem cells (HFSCs) have important functions for homeostasis of skin as well as hair follicle morphogenesis. In the present study, we screened different types of commercially available culture medium for culturing HFSCs. Among those tested, one type was shown capable of supporting the expression of stem cell markers in cultured HFSCs. However, the differentiation ability of HFSCs serially passaged using that optimal medium was found to be impaired, leading to unsuccessful formation of hair follicles in vivo.

Amniotic fluid stem cell extracellular vesicle therapy improves lung growth and branching morphogenesis in fetal hypoplastic lungs secondary to oligohydramnios

Project description:Oligohydramnios is known to cause fetal pulmonary hypoplasia. Amniotic fluid stem cell extracellular vesicles (AFSC-EVs) have shown beneficial effects on hypoplastic lungs secondary to congenital diaphragmatic hernia. Herein, we investigated, whether AFSC-EVs also foster antenatal lung development in hypoplastic lungs secondary to oligohydramnion. We therefore induced surgically oligohydramnion in rat dams and harvested their fetuses. Controls underwent surgery without amniotic fluid drainage. The hypoplastic lungs were assigned to ex vivo to either medium alone or medium + AFSC-EV as a lung explant. Those samples were harvested 24h later and subjected to RNA extraction for subsequent RNA-seq.
