Expression data from FACS-isolated liver endothelial cells

ABSTRACT: Cellular sources of liver endothelial cells remain elusive. Here, we used irradiation-conditioned bone marrow chimeric mice to lineage trace the endothelial cells. Bone marrow-derived (YFP+) and resident (YFP-) liver endothelial cells were isolated and total RNA were used for microarray analysis using Clariom S mouse assays (Applied Biosystems).

Project description:Mouse tissues (including heart, skeletal muscle, brain, lung, stomach, small instestine, colon, liver, kidney, testis, uterus, bone marrow, thymus, lymph node, spleen and embryo) were isolated from three C57BL/6 mice (eight-week-old, uterus from female and other tissues from male). Project description:Bone metastases is a common severe complication for breast cancer. We previously showed that conditioned medium (CM) from osteocytes stimulated with oscillatory fluid flow, mimicking bone mechanical loading during routine physical activities, reduced breast cancer cell extravasation across endothelial monolayers. Endothelial cells are situated at an ideal location to mediate signals between osteocytes in the bone matrix and metastasizing cancer cells in the blood vessels. Therefore, we used RNA sequencing to show that CM from endothelial cells conditioned in CM from flow-stimulated osteocytes significantly altered gene expression in bone-metastatic breast cancer cells. This This provides insights into the capability of bone-loading activity in preventing bone metastases.

Project description:Comprehensive transcriptional characterization of bone marrow endothelial cells by RNA sequencing was performed to determine the molecular properties/signatures of endothelium during bone marrow recovery and niche formation. Regenerative bone marrow endothelium was FACS-isolated from bone marrow aspirates of Acute Myeloid Leukemia patients 17 days after receiving chemotherapy (n=3). Niche-forming endothelial cells were FACS-isolated from fetal bones (gestational age 15-20 weeks) (n=3). Healthy adult bone marrow endothelial cells (n=7) were used as steady-state controls. cDNA was prepared using the SMARTer procedure (SMARTer Ultra Low RNA Kit, Clonetech). Project description:A comparative analysis of gene expression of 3 different experiments; 1. Perinate or adult-tagged GFP+YFP+ and bulk GFP+YFP- Tregs, 2. FL or BM-derived Tregs 3. Perinate or adult thymic Tregs. 1. Foxp3-eGFP-Cre-ERT2 x Rosa26-YFP lineage-reporter mice were injected ip with tamoxifen (4 injections, every third day) between 0-10 (perinate) or 35-45 (adult) days of age, and keep until 8 weeks of age. Perinate or adult-tagged GFP+YFP+ cells and bulk GFP+YFP- were double-sorted. 2. Hematopoietic progenitor cells were isolated from E18.5 fetal liver of CD45.1 congenic mice or from bone marrow of 5 weeks old CD45.2 B6 mice via negative selection of Thy1.2+ cells. The two populations were mixed at a 1:1 ratio, and were iv-transferred into irradiated (1000 rad) Rag-1-KO mice. FL or BM-derived Tregs were double sorted after reconstitution. 3. Foxp3-GFP+ Tregs were double-sorted from perinate (4 days old) or adult (5 weeks old) thymi. RNA from whole samples was amplified, labeled, and hybridized to Affymetrix Mouse Gene 1.0 ST Arrays.

Transcriptome profiling of adult mouse bone marrow endothelial cells

Project description:We performed RNA sequencing analyses of adult mouse bone marrow endothelial cells. Especially, we investigated gene expression profiling of endothelial cells before and after lethal irradiation or hematopoietic cell depletion. We also analyzed mouse bone marrow endothelial cell subtypes, Apln+ and diaphyseal endothelial cells. Whole bone marrow cells, lineage negative hematopoietic stem and progenitor cells, Lin- Sca1+ cKit+ cells were used as controls for the differential gene expression analyses.
