Halobacterium NRC-1, H2O2 Stress Response

Project description:50mM H2O2 was added to mid-log phase Halobacterium NRC-1 cultures. After constant stress of H2O2 for 30 minutes, cultures were spun down and pellets were resuspended in same volume of GN101 media. Samples for RNA preparation were collected during recovery time points at 0, 10, 20, 30, 40, 60 and 120 minutes. 16 samples (8 exposed to H2O2 and 8 non-exposed controls) were each hybridized on duplicate arrays (as dye-flips) against the same standard control sample.

Expression profile for PA14 lasR rhlR double mutant with H2O2 stress

Project description:Quorum sensing controls the expression of multiple virulence factors. PA14 genes lasR and rhlR are necessary for quorum sensing via homoserine lactones. A PA14 lasR rhlR deficient mutant exhibits a reduced oxidative stress response. Here we conducted a microarray to determine oxidative stress response gene regulation mediated by the homoserine lactone quorum sensing circuits. A PA14 lasR rhlR deficient mutant was compared to the wild-type with and without H2O2 stress.

Global gene expression profiling and senescence biomarker analysis of hESC exposed to H2O2 induced non-cytotoxic oxidative stress

Project description:Human embryonic stem cells (hESCs) potentially offer new routes to study, on the basis of the Developmental Origins of Health and Disease (DOHaD) concept, how the maternal environment during pregnancy influences the offspring health and can predispose to chronic disease in later life. Reactive Oxygen Species (ROS), antioxidant defences, and cellular redox status state play an important role in the regulation of gene expression and are involved in diabetes and metabolic syndromes as in aging. We designed an in vitro cell model of oxidative stress by exposing hESCs to hydrogen peroxide (H2O2) for 72h. To resemble the period of preimplantation embryonic development, we have analysed the global gene expression profiles of hESCs (HUES3) exposed to non-cytotoxic H2O2 concentrations (see main text), using Illumina HT-12 v4 expression beadarray.
