Gene expression profile of N2 and HPX-2 mutant C.elegans strains when exposed to E.coli and E.faecalis

ABSTRACT: We use RNAseq analysis as an un-biased and highly sensitive measurement of global transcriptomic changes upon the loss of HPX-2. The RNAseq result provided insights into the potential physiological processes HPX-2 is involved in.

Total proteome analysis of engineered/adapted E.coli strains with glucose/xylose co-utilization ability

Project description:This project was about developing a bacterial strain that could consume a mixture of glucose and xylose simultaneously with higher ethanol productivity. In this study, an ethanologenic strain (SSK42) was made deficient in Carbon Catabolite Repression (CCR) by deleting the ptsG gene encoding EIIBCGlc component of PTS transport system. This strain (SCD00) was then evolved for several generations on xylose containing minimal media. A strain (SCD78) was finally obtained that, unlike its parent strain could consume glucose and xylose simultaneously. Then we performed proteomics analysis of evolved and un-evolved strain. The strain SSK42 was also considered for proteome analysis as a reference for analysis. The starting strain – SSK42, is the derivative of E.coli B.

Project description:The aim of the study was to compare the transcriptome of E. coli K12 MG1655 cells lacking hydroperoxidase (Hpx-) and therefore unable to detoxify hydrogen peroxide, to wild-type cells; both when the Rtc RNA repair system is active and when it is inhibited; at 8 and 24 hpi. A very large number of genes (up to 1/3) were found to be differentially expressed in Hpx- as compared to wild-type. Inhibition of the Rtc system had dramatic effects on Hpx- but not on wild-type.
