RNA-seq of diabetic or non-diabetic murine bone marrow derived macrophages (BMDM)

ABSTRACT: The aim of this study was to interrogate whether BMDM basal or pro-inflammatory stimulated gene expression is altered by diabetes and persists despite in vitro glucose normalisation. Unstimulated BMDM from diabetic mice differentially express 632 genes (324 increased and 308 downregulated) compared to unstimulated control BMDM; this increases to 1,348 genes (802 increased and 546 decreased) upon stimulation (FDR<0.05, FC>1.5).

MicroRNA-802 regulates the innate immune response through Paneth cell expansion and Wnt signaling

Project description:Paneth cells are important for maintaining epithelial cell renewal and modulating innate immune function in the intestine through secretion of growth factors and antimicrobial peptides, which help sustain epithelial stem and progenitor cells and contribute to the intestinal barrier and protection against pathogenic bacteria. Here we show that the intestine-enriched miR-802 is a central regulator of intestinal epithelial cell proliferation and Paneth cell function. Genetic ablation of mir-802 in mice leads to amplified ROS generation and Notch/Wnt signaling, increased intestinal epithelial turnover, impaired enterocyte differentiation and nutrient uptake, and increased enterocyte apoptosis. Mice lacking mir-802 in the intestine also exhibit Paneth cell expansion, increased antimicrobial peptide production, and protection against Salmonella infection. This phenotype relies on the miR-802 target gene Tmed9.

Project description:Insulin resistance represents a hallmark during the development of type 2 diabetes mellitus (T2D) and in the pathogenesis of obesity-associated disturbances of glucose and lipid metabolism 1,2,3. MicroRNA (miR)-dependent posttranscriptional gene silencing has recently been recognized to control gene expression in disease development and progression including that of insulin-resistant T2D. MiRs, whose deregulation alters hepatic insulin sensitivity include miR-143, miR-181 and miR-103/107. Here we report that expression of miR-802 is increased in liver of two obese mouse models and of obese human subjects. Inducible transgenic overexpression of miR-802 in mice causes impaired glucose tolerance and attenuates insulin sensitivity, while reduction of miR-802 expression improves glucose tolerance and insulin action. We identify Hnf1b as a target of miR-802-dependent silencing and shRNA-mediated reduction of Hnf1b in liver causes glucose intolerance, impairs insulin signaling and promotes hepatic gluconeogenesis. In turn, hepatic overexpression of Hnf1b improves insulin sensitivity in db/db mice. Thus, the present study defines a critical role for deregulated expression of miR-802 in the development of obesity-associated impairment of glucose metabolism via targeting Hnf1b and assigns Hnf1b an unexpected role in the control of hepatic insulin sensitivity.

miR-324 regulates both osteoblast and osteoclast differentiation and function to control bone homeostasis II

Project description:microRNAs are non-coding RNAs which modulate the expression of other RNA molecules. One microRNA can target many transcripts, allowing each microRNA to play key roles in many biological pathways. miR-324 is a microRNA previously implicated in bone and cartilage maintenance, defects of which result in common age-related diseases, such as osteoporosis or osteoarthritis (OA). Cartilage damage was increased in both surgically and ageing-induced OA however changes in the cartilage transcriptome were minimal, with few miR-324 predicted targets dysregulated. However, in vivo micro-computed tomography and histology demonstrated that global miR-324-null mice had an increase in bone mineral density, trabecular thickness and cortical thickness, with many parameters increasing with age. The bones of the miR-null mice also had decreased osteocytes numbers and lipid droplets. In vivo TRAP staining revealed a decrease in osteoclasts with histomorphometry demonstrating an increased rate of bone formation in miR-324-null mice. Ex vivo assays revealed that the high bone mass phenotype of the miR-324-null mice resulted from increased osteoblast activity and decreased osteoclastogenesis. RNA-seq and qRT-PCR followed by miR-324 target prediction and validation in osteoblasts, bone marrow macrophages and osteocytes, revealed that the osteoclast fusion regulator Pin1 was a miR-324 target in the osteoclast lineage, Hoxa9 and Samd5 were osteocyte target genes and the master osteogenic regulator Runx2 was a target of miR-324-5p in osteoblasts, the in vitro overexpression of which recapitulated the increased osteogenesis and decreased adipogenesis phenotype observed in vivo. These data point to important roles of miR-324 in skeletal biology. Elucidation of pathways regulated by miR-324 offers promise for the treatment of bone diseases such as osteoporosis.
