Nutrient sensing controlling gut motility in the stomachless fish ballan wrasse (Labrus berggylta)

ABSTRACT: Ex vivo intestines from 15-20 g ballan wrass (Labrus bergylta) were administered a bolus (lipid, protein, inert matter or empty control) before being mounted in oxygenated physiological buffer inside a glass vial. The intestinal motility patterns were documented with time lapse imaging for 14 hours. The images were used to construct spatio-temporal maps for gut motility analysis; i.e. to determine the speed, amplitude, frequency, travelled distance, direction, and initiation site of waves of muscle contractions. The experiments were repeated for RNA sampling and deep seq analysis. Gene expression analysis showed that intestines administered intact lipids clustered closer to the empty intestines compared to those given hydrolyzed lipids.

CEBPD suppresses motility-associated gene signatures and reduces pancreatic ductal adenocarcinoma cell migration

Project description:To investigate the effects of CEBPD on pancreatic ductal cell tumorigenicity, we established doxycycline-inducible MIA PaCa-2 cells for CEBPD over-expression, as well as non-control cells with an empty inducible construct. RNAseq was performed on these cells in the presence and absence of doxycycline and we showed that CEBPD activates gene signatures associated with cell motility.

Transcriptome responses to dexamethasone of the hypothalamus of pigs with different genotypes at the GRAla610Val mutation

Project description:The main aims of the experiment were to 1) chart gene networks governed by glucocorticoid receptor on a genome-wide scale, 2) to provide insights into mechanisms modulating glucocorticoid sensitivity and 3) to obtain better understanding of molecular and phenotypic consequences of the GRAla610Val substitution by examining its transcriptome signature in untreated and dexamethasone-treated animals. A total of 48 purebred German Landrace piglets were used in the experiment. At the time of the experiment the piglets were 7-weeks old. A bolus injection of either dexamethasone or sterile saline was administered intramuscularly into the neck muscle. Twenty piglets received saline, sixteen piglets were treated with 10 μg/kg dexamethasone, and twelve piglets were treated with 60 μg/kg dexamethasone. All three treatment groups were balanced for sex and genotype.

Muscle directs diurnal energy homeostasis through a myokine-dependent hormone module in Drosophila

Project description:Inter-tissue communication is critical to control organismal energy homeostasis in response to temporal changes in feeding and activity or external challenges. Muscle is emerging as a key mediator of this homeostatic control through consumption of lipids, carbohydrates, and amino acids, as well as governing systemic signaling networks. However, it remains less clear how energy substrate usage tissues, such as muscle, communicate with energy substrate storage tissues in order to adapt with diurnal changes in energy supply and demand. Using Drosophila, we show here that muscle plays a crucial physiological role in promoting systemic synthesis and accumulation of lipids in fat storage tissues through Foxo. To gain more insight into these tissue-specific changes in lipid metabolism, we generated genome-wide expression profiles from dissected thoraces (enriched in indirect flight muscle), carcass (enriched in fat body), and intestines of Act88FG4>FoxoRNAi flies and control animals.

Transcription profiling of mouse small intestine from PlagL2-/- animals reveals transcription factor PlagL2 regulates steps in chylomicron metabolism

Project description:Enterocytes assemble dietary lipids into chylomicron particles that are taken up by intestinal lacteal vessels and peripheral tissues. Although chylomicrons are known to assemble in part within membrane secretory pathways, the modifications required for efficient vascular uptake are unknown. We report that the transcription factor Pleomorphic adenoma gene-like 2 (PLAGL2) is essential for this aspect of dietary lipid metabolism. PlagL2-/- mice die from post-natal wasting owing to failure of fat absorption. Lipids modified in the absence of PlagL2 exit from enterocytes but fail to enter interstitial lacteal vessels. Dysregulation of enterocyte genes closely linked to intracellular membrane transport identified candidate regulators of critical steps in chylomicron assembly. PlagL2 thus regulates essential and poorly understood aspects of dietary lipid absorption and its deficiency represents an authentic animal model with implications for amelioration of obesity or the metabolic syndrome. Experiment Overall Design: Total RNA was extracted from 4 knockout and 4 wild-type mouse small intestines at 18.5 dpc using the Macherey-Nagel Nucleospin kit. cRNA synthesis and labeling, hybridization to Affymetrix (Santa Clara, CA) MOE430 2.0 expression arrays, and data acquisition occurred on the Affymetrix GeneChip Instrument System.
