Gene expression signatures of megakaryocytes post-IR

Project description:Platelets are anucleated blood cells that are produced by their progenitor cell, the megakaryocyte (MK). Platelets are centrally positioned in hemostasis and thrombosis and, according to the recent findings, play key roles in innate immunity, inflammation, atherogenesis, and cancer metastasis. The quantitative and qualitative properties of platelets are crucial determinants of their hemostatic function. While the regulatory mechanisms of platelet number and size have been studied separately, critical questions remain regarding the interplay of platelet number and size in hemostasis. In this study, using a mouse model of irradiation (IR)-induced thrombocytopenia, we show that mature MKs (mMKs) are resistant to IR and maintain the nadir platelet levels post IR. To identify the phenotype switching and function variation in BM MKs post IR, we performed microarray expression profiling of MKs from the bone marrow of mice at 1- or 3-days post IR and control mice (Ctrl).

Mammary Tissue: Wild type mammary glands vs IGF-IR induced mammary tumors vs IGF-IR independent tumors

Project description:Molecular profiling was used to classify mammary tumors that develop in MTB-IGFIR transgenic mice. It was determined that the primary mammary tumors (PMT), which develop due to elevated expression of the type I insulin-like growth factor receptor (IGF-IR) in mammary epithelial cells, most closely resemble murine tumors with basal-like or mixed gene expression profiles and with human basal-like breast cancers. Downregulation of IGF-IR transgene in MTB-IGFIR tumor-bearing mice leads to the regression of most of the tumors followed by tumor re-appearance in some of the mice. These tumors that re-appear following IGF-IR transgene downregulation do not express the IGF-IR transgene and cluster with murine mammary tumors that express a mesenchymal gene expression profile and with human claudin-low breast cancers. Therefore, IGF-IR overexpression in murine mammary epithelial cells induces mammary tumors with primarily basal-like characteristics while tumors that develop following IGF-IR downregulation express a gene signature that most closely resembles human claudin-low breast tumors.

Project description:The cellular response to DNA damage is vital for maintaining genomic stability and preventing undue cell death or cancer formation. The DNA damage response (DDR), most robustly mobilized by double-strand breaks (DSBs), rapidly activates an extensive signaling network that affects numerous cellular systems, leading to cell survival or programmed cell death. A major component of the DDR is the widespread modulation of gene expression. We analyzed transcriptional responses to ionizing radiation (IR) in 5 human cell lines to elucidate the scope of this response and identify its gene targets. According to the mRNA expression profiles most of the responses were cell line-specific. Data analysis identified significant enrichment for p53 target genes and cell cycle-related pathways among groups of up-regulated and down-regulated genes, respectively. Expression profiles were measured using affymetrix chips in IR- irradiated G361 cells and their time-matched untreated controls. Time points recorded were 0, 3 and 6 hrs.
