RNA sequencing of mouse placenta in control and protein restricted pregnancies

ABSTRACT: We used RNA sequencing to identify differentially expressed genes in whole mouse placentas. Pregnant mice were fed either a 20% or 6% protein (isocaloric) two-weeks before mating and throughout pregnancy. Placental lysates are derived from C57BL/6 conceptuses at E16.5 of gestation.

Project description:STOX1 overexpressing placentas generate preeclampsia-like symptoms in pregnant mice. These symptoms are reverted by aspirin. The experiment aims at analyzing placental expression in Transgenic/non transgenic with and withou aspirin, in the placentas at 16,5 days post-coïtum. There are two strains of transgenic mice that express the transgene at different levels: STOX42 about 15 fold that of STOX13. STOX13 expressed the human STOX1 roughly at the level of the mouse endogeneous Stox1 in the placenta.

Project description:External ionizing Irradiation of pregnant mice at E13.5 4 Gy, gene expression at E17.5 On E13.5, female C57Bl/6HN mice were irradiated (total body) at 0 Gy (sham) or 4 Gy using a Gamma Cell cesium irradiator (JL Shepherd, San Fernando, CA), with a dose rate of 70 cGy/min. The mice were monitored for 4 days, and on E17.5, they were sacrificed, and placentas were procured and processed for RNA extrtaction.

Transcriptional profiles of placentas from systemic lupus erythematosus and normal full-term pregnancies.

Project description:We report transcriptional profiles of placentas from systemic lupus erythematosus pregnancies and normal full-term pregnancies. We recruited 8 pregnant patients with SLE, and 8 age-matched healthy pregnant women as normal controls. RNA-seq strand-specific libraries were constructed using the VAHTS Total RNA-seq (H/M/R)Library Prep Kit. Differentially expressed genes were identified using Cuffdiff. The P-value significance threshold in multiple tests was set by the false discovery rate (FDR). The fold-changes were also estimated according to the FPKM in each sample.The differentially expressed genes were selected using the following filter criteria: FDR <0.05 and fold-change >2.

Effect of maternal iron deficiency on placental gene expression on gestational day 18.5 in rats

Project description:Iron deficiency occurs when iron demands chronically exceed intake, and is particularly prevalent in pregnant women. Iron deficiency during pregnancy poses health risks for the baby. The placenta serves as the interface between a pregnant mother and her baby; thus, maternal iron deficiency may indirectly impact fetal growth and development by altering placental function. In this study, pregnant Sprague-Dawley rats were fed either a low-iron or iron-replete diet starting two weeks before mating. On gestational day 18.5, RNA was collected, and a Clariom S microarray was performed to elucidate differences in gene expression between gestaional day 18.5 placentas isolated from dams fed iron replete or iron deficient diets.

Epigenome analysis of normal and preeclamptic placenta samples

Project description:Preeclampsia (PE), a hypertensive disorder of pregnancy, is hypothesized to be associated with, if not mechanistically related to abnormal placental function. However, the exact mechanisms regulating the pathogenesis of PE remain unclear. While many studies have investigated changes in gene expression in the PE placenta, the role of epigenetics in PE associated placental dysfunction remains unclear. Using the genome-wide Illumina Infinium Methylation 450 BeadChip array, we analyzed gene-specific alterations in DNA methylation in placental biopsies collected from normal pregnant women delivering at term (n=14), with term PE (≥37 weeks; n=19) or with preterm PE (<37 weeks, n=12). Of the 485,582 gene loci on the array, compared to controls, 229 loci were differentially methylated in PE placentas and 3411 loci were differentially methylated in preterm PE (step up p-value <0.05 and >5% methylation difference). Functional annotation of the differentially methylated genes in preterm PE placentas revealed a 32 gene cluster in the cadherin and cell adhesion functional groups (Benjamini p<0.00001). Hypermethylation of CDH11 (p=0.0143), COL5A1 (p=0.0127) and TNF (p=0.0098) and hypomethylation of NCAM1 (p=0.0158) was associated with altered mRNA expression in preterm PE placentas. These studies demonstrate aberrant methylation, correlating with disease severity, in PE placentas. Project description:Pregnant rats were randomly divided into two groups. Dams were then housed individually and fed throughout gestation a 22 % protein diet, representing the control group (C, n=5), or an 9% isocaloric protein diet, representing the low-protein (LP, n=6). Both diets were isocaloric, because the protein deficiency in the LP diet was compensated by the addition of carbohydrates. Feeding of LP diet began on the day of conception and continuing throughout pregnancy. Placental gene expression in pregnancies established after the transfer of day 7 blastocysts derived from in vitro (IVP), somatic cell nuclear transfer (SCNT) and in vivo (AI) embryos

Project description:Aberrant placental gene expression associated with culture condition and/or deficiencies in transcriptome reprogramming are hypothesized to be the major cause of SCNT and IVP inefficiencies. Therefore, the main objective of this study was to invesitgate the dysregulated genes, molecular pathways and functional alteration in bovine placentas derived from SCNT and IVP pregnancies compared to their AI counterparts Day 7 blastocysts derived from AI, IVP or SCNT were transferred to synchronized cows. The pregnant animals were slaughtered at day 50 of the gestation period and the placentas were then recovered and used for transcriptome analysis using GeneChip bovine genome array
