Project description:This experiment set contains the complete set of raw data for Whitfield et al. (2002)Mol.Biol.Cell. HeLa S3 cells were synchronized by three different methods (double thymidine block, thymidine-nocodazole block or mitotic shake-off)in five independent time courses. mRNA from the first and second double thymidine block experiments were converted to cDNA by standard methods using an oligo-dT/random hexamer mix and hybridized to 24K arrays; mRNA from the third double thymidine and thymidine-nocodazole block experiments were converted to cDNA using olido-dT alone and hybridized to 43K arrays; total RNA from the mitotic shake-off experiment was first amplified using a modified eberwine protocol, aRNA converted to cDNA by reverse transcription with random hexamer and hybridized to 43K arrays. Project description:[Hela cells]: We performed cdr2 knockdown with a pool of 4 cdr2-specific siRNAs to test whether cdr2 may regulate c-myc target genes as cells passage through mitosis. Project description:Analysis of synchronized HCT116 cells at various time points up to 10 hours following treatment with DMSO or Nocodazole. Experiment Overall Design: HCT116 colorectal cancer cell line was obtained from the American Type Culture Collection (ATCC) and grown in McCOYâS 5A MEDIUM MODIFIED (SIGMA) with 10% FBS (JBS). It was maintained at 37°C and 5% CO2. Synchronous culture was obtained by incubating cells for 19 h in 2 mM of thymidine, followed by a 9 h-incubation in normal medium and a second 16 h-incubation in thymidine (2 mM). Cells were washed with normal medium followed by treatment with DMSO for 0, 2, 4, 6, 7, 8, 9 and 10 h as a control or 0.1 mg/ml Nocodazole (Sigma) for 7, 8, 9 and 10 h.

Project description:We compared the mRNAs expression profile of HeLa cells between two phases of the mitotic cell cycle: S and G2/M phases. Results provide insight into the regulation of transcript levels during mitotic cell cycle progression. HeLa cells were synchronized with double thymidine blockade (12 hours with 2 mM thymidine, 12 hours release, and 12 hours with 2 mM thymidine), and cells were taken after 2 hours release (S phase) and 8 hours release (G2/M phase). Project description:HeLa cells were synchronized with a double thymidine block procedure. Briefly, the exponentially growing HeLa cells were maintained with 2 mM thymidine for 18 h, followed by a release of 9 h in fresh medium, and then cells were re-cultured in 2 mM thymidine for additional 15 h. After a release of 7.5 h in fresh medium, DMSO or G6PD inhibitor was added to the medium. 1 h later, the cells entered into M phase. Mitotic cells were harvested by mitotic shake-off. Then, the samples were subjected to LC-MS/MS analysis. Finally, a Kinase-Substrate Enrichment was performed, which could infer the changes of upstream kinase activity upon the treatment of G6PD inhibitors.
