Project description:High-throughput sequencing technique was used to screen the miRNAs which has potential role in the aging delaying effect of olmesartan on human smooth muscle cells. We cultured human vascular smooth muscle cells and used the third to fifth generation cells as unaging cells (Zhengchang-1,-2,-3), natural culture to passage 10 to 15 as aging group samples (Bo-1,-2,-3), and cells treated with olmesartan for the same time as aging group cells as treatment group (bo-ao-1,-2,-3). After collecting cells of each group, RNA was extracted and reversed. After the quality inspection was qualified, miRNA sequencing was carried out. Finally, we confirmed that the miRNA expression profile of human vascular smooth muscle cells changed after olmesartan treatment. This study confirmed that miRNA plays a potential role in the aging process of human smooth muscle cells and that olmesartan may play a role in delaying aging by regulating miRNA expression profile.

Project description:This experiment is part of the FunGenES project (FunGenES - Functional Genomics in Embryonic Stem Cells partially funded by the 6th Framework Programme of the European Union, http://www.fungenes.org). The experiment was conducted at the University Nice Sophia-Antipolis, Nice, France. The aim of this project is to identify new regulators, and new markers of the early steps of adipocyte commitment.Addition of RA on early-differentiating mES cells (Hanging drops, Day 3) orientates their commitment to the adipocyte lineage, while inhibiting myogenesis. The effect of RA on adipogenesis is mediated by retinoic acid receptor beta (RARb), since it can be mimiked by the addition of CD2314 compound, a specific agonist of this receptor isotype. In contrast, mES cell adipogenesis can be bloked by artificial activation of the Wnt pathway using GSK3 inhibitors such as lithium, or Bio.To decipher the genetic pathways that control the early steps of adipogenesis, we have compared the transcriptome of mES cells stimulated to differentiate using adipogenesis-enhancing (CD2314) or -inhibitory (BIO, or CD2314+BIO) treatments (treatment between day 3 and day 6). We have analysed the transcriptome of these cells at day 3 (before treatment), at day 6 (just after stimulation or inhibition of the early steps of adipocyte commitment) and at day 11 (to investigate later regulators of adipogenesis). Materials and methods: CGR8 at different time point during adipogenic differentiation. - Induction of differentiation by removal of LIF at day 0, hanging drops from day 0 to 3. - At day 3, harvesting of the hanging drops, and start adipogenesis-enhancing (CD2314) or -inhibitory (BIO, or CD2314+BIO) treatments (treatment from day 3 to 6, EBs in suspension on petri dishes).- At day 6, harvesting of the EBs and plating on gelatin-treated TC plates in normal culture medium ( GMEM+10%serum).- At Day 7, treatment with final inducers of adipogenic differentiation (Insuline, T3, BRL). Same medium until end point of differentiation (Day 21). Project description:Deoxynivalenol (DON) and zearalenone (ZEA) are the most common mycotoxins, which are damage to the reproductive system of human and animal. Endometrium cells perform vital roles of female reproduction. In the present study, transcriptomic was conducted on different dosage (0 μM, 10 μM and 30 μM) of DON and ZEA treatment donkey endometrium cells. Transcriptomic analysis showed 7386 differentially expressed genes (DEGs) were identified between DON 10 μM treatment group and control group, with 3655 up regulated and 3731 down; 8694 DEGs were identified between DON 30 μM treatment group and control group, with 4532 up regulated and 4162 down; 1768 DEGs were identified between ZEA 10 μM treatment group and control group, with 763 up regulated and 1005 down; and 3311 DEGs were identified between ZEA 30 μM treatment group and control group, with 1501 up regulated and 1810 down. In DEGs, Caspase-1, GSDMD-NT, ESR 1, PRDX 4, NOX 1, MSH 6 and CD 44, were expressed changed in DON and ZEA treatment group, which related to pyroptosis, hormone secretion, oxidative stress and cancerous. It is unprecedented that DON induced cell death by pyroptosis. This study provides valuable genetic resources for understanding the mechanisms and pathways of the toxin effect of DON and ZEA for Equus asinus’ endometrium cells.

Project description:Endothelial cell and vascular smooth muscle cell were cocultured in hanging droplets to form spheroids representing an inverted vessel lumen. Control or conditioned media from an extravillous trophoblast (EVT) cell line was incubated with vascular spheroids for 24 hours. Spheroid RNA was then analyzed by Illumina Sentrix BeadChip array. Spheroids incubated with EVT conditioned medium showed significant up/downregulation of 101 genes (>1.5-fold; P<0.05), including an upregulation of C-X-C motif chemokine 10 (IP-10). C-X-C motif chemokine 10 expression was confirmed by qualitative real-time PCR and Western blot analysis of spheroids, and immunohistochemistry of first trimester decidua and ex vivo dissected nonplacental bed spiral arteries. EVT conditioned medium reduced VSMC expression of differentiation markers, and both EVT conditioned medium and C-X-C motif chemokine 10 increased motility of VSMC indicating dedifferentiation of VSMC.

Project description:Inactivation of BMP9/10 in mice leads to attenuated contractility of smooth muscle cells and decreased blood pressure.Treatment of de-differentiated PASMCs with BMP9/10 resulted in a strong increase of ACTA2 expression as measured by immunofluorescence and RT-PCR, indicating a switch from the de-differentiated, synthetic to a differentiated, contractile state. To further study the role of BMP9/10 in smooth muscle cells, we isolate PSAMCs and stimulate cells with or without BMP9/10 and perform the RNA-seq analysis.

Project description:This study aims to identify and characterize miRNA expression in aneurysmal smooth muscle cells (SMCs) and M1 and M2 macrophages isolated by laser microdissection from human AAA biopsy samples. The aim of this study was to profile (with microarray technology) miRNAs in M1 and M2 macrophages and Smooth muscle cells (SMCs), isolated by laser capture microdissection (LCM). SMCs isolated from control non-aneurysmal aortas were the control group according to which data were normalized. Areas enriched in M1 macrophages, M2 macrophages and smooth muscle cells were microdissected (9.8 [4.7-16.2] mm2) from two different biopsy samples of human abdominal aortic aneurysm. An area enriched in smooth muscle cells was microdissected from two biopsy samples of non aneurysmal abdominal aortas. Each microdissected samples were analyzed independently on microarray. M1 and M2 macrophages and smooth muscle cells expressed in human abdominal aneurysmal aortas were each analyzed in duplicate (each isolated from different human donors of abdominal aortic aneurysm). Analysis of replicated samples of cells were performed on a second microarray. Data were normalized with smooth muscle cells isolated from human abdominal non aneurysmal aortas.
