Project description:To obtain insight into the genetic basis of the increase of functional activity of memory B cells over time, we compared the gene expression profiles of day 7 and day 40 NP-specific/IgG1 memory B cells, GC B cells and plasma cells in immunized WT mice and naïve B cells, before and after activation in vitro. Total RNAs in three replicates of NP-specific/IgG1 memory B cells, GC B cells and plasma cells in immunized WT mice at day 7 and 40 postimmunization and naïve follicular and marginal zone B cells, before and after activation in vitro, were amplified and labeled according to the Affymetrix's instructions. cRNA was hybridized to Mouse Genome 430 2.0 GeneChips (Affymetrix).

Expression data from murine bone marrow-resident plasma cells and spleen mature follicular B cells

Project description:CD138+ B220- plasma cells were sorted from bone marrow and B220+ CD23+ mature follicular B cells were sorted from the spleens. Plasma cells were sorted from C57BL/6 mice 7 days after boosting with antigen, with mice first primed with an i.p. injection of KLH/IFA followed by boost at day 21 with KLH/PBS i.p. Mature B cells were sorted from antigen-naïve C57BL/6 mice. We compared expression profiles of plasma cells and mature B cells to identify differentially expressed transcripts. Seven days after boosting with KLH, plasma cells were enriched by CD138-PE/anti-PE magnetic bead enrichment and then sorted to purity. Affymetrix microarrays were performed on 50,000-70,000 plasma cells and 2,000,000-3,000,000 naïve B cells.

Clonal structure and dynamics of human memory B cells and circulating plasmablasts

Project description:Using single cell sequencing of multi-year serial blood samples from two healthy donors we found that the memory B cell repertoire was dominated by large clonal families producing IgM, IgG2 and IgA, of which 0.2% share nearly identical VH/VL sequences in different individuals. In contrast, IgG1 families, including those encoding antibodies to T-dependent antigens, were of small size. Longitudinal samples collected over a period of several years showed the overall stability of the memory B cell pool in all its subsets. Surprisingly, clonotype stability was also found in serial samples of circulating plasma cells that largely overlapped with long-term memory B cells and contained recurrent clonotypes of IgA and IgG2 and, at lower frequency, IgG1 specific for recall antigens. Collectively, this study provides a global view of the structure, stability and dynamics of the human memory B cell pool and reveal that a large fraction of the clonal families is active at any time point in the generation of plasma cells involving both T independent and T dependent responses.

Follicular dendritic cells restrict IL4 availability in germinal centers and foster memory B cell generation

Project description:Germinal centers (GCs) are sites of antibody affinity maturation and memory B cell generation. Follicular dendritic cells (FDCs) form a dense stromal network within GCs, but the full extent of their supportive activity is not understood. Here we showed that FDCs expressed IL4Ra and they reduced IL4 availability in GCs. We used scRNA-seq to examine IL4 actions within the GC and identified a subset of light zone cells marked by high IL4Ra and CD23 and lack of a Myc gene-expression signature. Trajectory inference analysis suggested this population could differentiate into pre-memory and pre-plasma cells. Genetic experiments showed that IL4Ra and STAT6 acted intrinsically to restrain pre-memory B cell generation. By reducing IL4 availability, FDCs enhanced GC B cell selection and fostered memory cell generation. This work suggests that GC FDCs provide a niche that limits bystander IL4 activity, focusing IL4 action on B cells undergoing selection and enhancing memory cell differentiation
