Project description:Before the onset of sexual differentiation, male and female external genitalia are morphologically indistinguishable. After the onset of sex-specific production of sex steroids by the gonads, the male and female external genitalia differentiate into either a penis or clitoris. Here we use single cell sequencing to investigate the similarities and differences of cell populations in the external genitalia during the critical sexual differentiation window in the mouse embryo. We found the male and female genitalia are largely similar in cell composition and gene expression at the bipotential stage of development. As sexual differentiation ensues, sex steroid-dependent and -independent differences in cell populations arise, leading to dimorphic establishment of the external genitalia.

Project description:MicroRNAs (miRNAs) are a highly conserved class of small RNAs which function in a sequence-specific manner to post-transcriptionally regulate expression of target genes. Tissue-specific miRNA expression studies have discovered numerous functions for miRNAs in various aspects of embryonic development, but a role for miRNAs in gonadal development and sex differentiation has not yet been reported. Using the chicken embryo as a vertebrate model, differential miRNA expression between male and female embryonic gonads, was analysed at three developmental stages (embryonic days (E) 5.5, E6.5 and E9.5), using custom-designed 4x2K CombiMatrix miRNA microarray. The aims of this study were to: 1-identify miRNAs differentialy expressed by sex; 2-identify sex-specific miRNAs; 3-analyse global changes in miRNA up-regulation in male versus female gonads before, during and after the histological onset of sexual differentiation. This study provides a basis for establishing whetehr miRNAs are involved in either initiating or regulating vertebrate gonadal sex differentiation. Project description:Following sex determination, XY and XX gonads develop into a testis and an ovary, respectively. Depending on the sex of the gonad, resident germ cells will subsequently be committed to either spermatogenesis or oogenesis. In this study we took advantage of the Wv/Wv mouse genetic model, in which gonads are almost devoided of germ cells, to uncover gene expression underlying fetal germ cell development. Male and Female gonads were collected at 12, 14 and 16 days post coitum (dpc) from wild-type and Wv/Wv mouse embryos. Total RNAs were extracted and hybridized on Affymetrix microarrays. Expression signals from male and female gonads or from wild-type and mutant gonads were compared to identify sexually dimorphic genes as well as genes expressed in germ cell during fetal gonad development.

The molecular pathways showing sexual dimorphic patterns have been principally investigated in the gonads. In other organs, much less is known about the gender-associated pattern of gene expression. The brain plays a coordinating role on sexual function in both genders including regulation of reproductive development and maturation through the synthesis of neuropeptide hormones and the regulation of sexual behavior in both males and females. In this study, we aimed at identifying molecular pathways regulated in a gender specific manner in breeding zebrafish in order to generate a greater understanding of the gender specific physiology of the brain. To do so, we measured the gene expression profiles of individual brains of 6 males and 5 females using a 17k oligonucleotide microarray, and interrogated the dataset for genes showing a gender-specific gene expression profile. 42 genes were found to be differentially expressed between males and females from which 18 genes were over-expressed in males and 24 genes were over-expressed in females. Project description:Pirarucu (Arapaima gigas, or A. gigas) is a native fish species to Amazon basin and, economically important in the Brazilian Amazonian for its great potential to aquaculture. In the natural environment the A. gigas is captured only in the sustainable development reserves of the State of Amazonas by applying a system of rotation in fishing in order to avoid overfishing of this important fishing resource. The reproductive biology of A. gigas has long been intriguing; however, very little is known about the molecular pathways underlying their sexual differentiation and determination. Using the SOLiD sequencing platform, a total of 432,058,560 short sequencing reads were produced. An average of ~30% of sequencing reads could be mapped to Asian arowana reference cDNAs. 305 genes showed higher expression in female brain against 8 gene with higher expressing in male brain. In gonad, there are 120 genes higher expressed in female against 10 gene higher expressed in male.

Sexually mature fish (6 males and 6 females) were kept in a 15L tank and allowed to breed naturally. After a 20-day period, fish were sacrificed humanly and the brain was extracted, fixed in RNAlater (Ambion) and stored at -20oC until required for microarray analysis. RNA was extracted using the TRI reagent method followed by a LiCl precipitation (Sigma) according to the manufacturer's instructions. Total RNA was amplified and labeled using the Amino Allyl MessageAmp aRNA amplification kit (Ambion). A reference design was used for the array hybridizations. Array analysis was performed according to established protocols (described in Santos et al., 2007) and the data was extracted and queried for differences in gene expression profiles between genders.
