Project description:Linking the evolution of the phenotype to the underlying genotype is a key aim of evolutionary genetics and is crucial to our understanding of how natural selection shapes a trait. Here we consider the genetic basis of sex allocation behaviour in the parasitoid wasp Nasonia vitripennis using a transcriptomics approach. Females allocate offspring sex in line with Local Mate Competition (LMC) theory. Female-biased sex ratios are produced when one or few females lay eggs on a patch. As the number of females contributing offspring to a patch increases, less female-biased sex ratios are favoured. We contrasted the transcriptomic responses of females as they oviposit under conditions known to influence sex allocation: foundress number (a social cue) and the state of the host (parasitised or not). We found, that when females encounter other females on a patch, or assess host quality with their ovipositors, the resulting changes in sex allocation is not associated with significant changes in whole-body gene expression. We also found that the gene expression changes produced by females, as they facultatively allocate sex in response to a host cue and a social cue, are very closely correlated. We expanded the list of candidate genes associated with oviposition behaviour in Nasonia, some of which may be involved in fundamental processes underlying the ability to facultatively allocate sex, including sperm storage and utilisation.

Project description:Understanding the consequences of thermal and chemical variations in aquatic habitats is of importance in a scenario of global change. In ecology, the sex ratio is a major population demographic parameter. Research carried out so far on environmental perturbations on fish sex ratios has usually involved a few model species with a strong genetic basis of sex determination, analyzing juvenile or adult gonads. However, the underlying mechanisms at the time of commitment are poorly understood. The European sea bass has a mixed genetic and environmental sex determination system, which makes it naturally sensitive to environmental cues. Here, we transcriptomically analyzed developing gonads experiencing either testis or ovarian differentiation as a result of thermal and/or estrogen influences. Elevated temperature masculinized genetic females while estrogen exposure resulted in an all-female population. A total of 383 genes were differentially expressed, with an overall downregulation in the expression of genes involved both in testicular and ovarian differentiation in the estrogen-exposed fish achieved by a shutdown of the first steps of steroidogenesis. GO enrichment analysis uncovered affected pathways related to the immune response, xenobiotic metabolism, response to stimulus, signaling and growth. However, once the female phenotype was imposed gonads could continue their normal development, even taking into account that some of the resulting females were fish that otherwise would have developed as males. The data on the underlying mechanisms operating at the molecular level presented here contributes to better understanding the sex ratio response of fish species subjected to a combination of environmental perturbations.

Project description:Gonad is a bipotential tissue that depending on the genetic and environmental inputs its final phenotype will be determined. Gonadal development is a very complex process because from an undifferentiated gonad the future testis or ovary will be formed. To better understand the complexity of the gonad plasticity throughout development we explored the transcriptomic landscape of gonads from undifferentiated to juvenile fish. For this study we have used turbot (Scophthalmus maximus) that presents one of the most remarkable sexual dimorphism in cultured fish species. With the aim of identifying gene patterns for each gonad developmental stages a microarray species-specific enriched by reproduction- and immune-related genes was assessed. Further genes with sex-differential mRNA expression were mapped in the sex- and growth-QTL (quantitative trait loci) markers. Data revealed that gonadal transcriptome prevail in front of fish biometry as it was possible to classify gonads according to sexual development status by using gene expression profiles rather than their age or size. The same classification was observed with a selection of a subset of reproduction-related genes showing the importance of gene profiles for each of the developing gonadal stage. Overall microarray data showed the importance of the level of expression (intensity) rather than the number of genes found in each development period. Larger numbers of differentially expressed genes (DEG) were found in the differentiating gonads when compared to ovaries and much smaller number resulted when compared to testis. However, more number of DEG was found in testis when compared to ovaries. The earliest presence of cyp19a1a, the key female sex differentiation gene, was found at 90 dpf together with its transcription factor foxl2 showing at the same time a positive correlation with other genes related to extracellular matrix (ecm2), immune system (irf5) or methylation (sall1). The presence of male-related genes like sox9 was evidenced at early developmental stages (70 and 90 dpf) while expression of dmrt3 started not earlier than 150 dpf, coinciding when larger number of male-related genes were found. All developmental stages presented genes related to immune system and epigenetic mechanisms revealing their importance during gonadal development. Sex-QTLs showed larger amount of sex-differential transcripts in the linkage group 6 and 21 both in males and females but transcripts mapped near growth-QTLs were only found in females. Here it is presented the first assessment of the expression profiles observed in the gonads at different developmental stages by identifying genes responsible for male or female traits. It has been identified gene clusters for each developmental stages and gene network interactions for each sexual phenotype. The expression of known male and female-related genes were precisely described throughout gonadal development as well as the expression of other genes with an unknown reproduction function which have been identified for the first time during sexual development process. In addition, it was possible to localize some of these genes on the sex- and growth-QTLs. The data presented here can help to better understand the reproduction system in cultured fish species and improved selection breeding programs to increase their productivity.

Project description:This experiment aimed at characterising the modulatory role of murine induced Neural Stem Cells (iNSCs) and Neural Stem Cells (NSCs) on macrophages (MPs) exposed to LPS in vitro. Naïve MPs were polarized into an M1-like phenotype with LPS, and then co-cultured with 1:1 ratios of iNSCs in a trans-well system that avoids cell-to-cell contacts. Naïve MPs, LPS-stimulated MPs and LPS-stimulated MPs co-cultured with NSCs were used as controls.

Project description:Sex determination is a variable and complex mechanism, yet it can be found all over the plant and animal kingdoms. It creates two morphological different outcomes from one and the same species. Our work demonstrates the involvement of a non-coding RNA in this process for the teleost Danio rerio. Although we were not able to understand the full mechanism behind it, we can provide a piece of the puzzle, which once again highlights the complexity and flexibility of sex determination.
