Splenic immune response to Plasmodium infection initiated by two different routes

Project description:Plasmodium chabaudi parasite obtained from recently mosquito transmitted infections give rise to an attenuated blood-stage infection when compared to serially blood passaged parasites. We compared the immune response in the spleens induced by these two types of parasites in order to determine what may cause this attenuation. Spleens were taken at several timepoints during the blood-stage infection and RNA-seq analysis was performed.

Time series of Balb/c spleen infected with Plasmodium yoelii non-lethal vs lethal strains

Project description:Knowledge on the dynamic features of the processes driven by malaria parasites in the spleen, our biggest lymphoid organ, is lacking. We have implemented intravital microscopy and magnetic resonance imaging of the mouse spleen in experimental infections with the Plasmodium yoelii non-lethal (17X) and lethal (17XL) strains. Notably, there was higher parasite accumulation, reduced motility, lost of directionality and different T2 relaxation times only in spleens of mice infected with the 17X strain. Moreover, these differences were associated with the formation of a strain-specific induced spleen tissue barrier, with macrophage-clearance escape, and with cytoadherence of infected reticulocytes to this barrier. This is a novel spleen-immune evasion mechanism in which parasite-induced spleen remodeling and adherence to this organ allow establishment of chronic infections. We performed time-series global transcriptional analyses from spleens of mice infected with the P. yoelii 17X and 17XL strains at days 3, 4, 5, 10% and 50% of parasitemia post-infection, together with non-infected spleens as a reference day 0, using commercially available arrays representing the complete mouse genome (Agilent Whole Mouse Genome G4122A).

Gene expression profiling of CD4+CD25+Foxp3+ regulatory T cells and CD4+CD25-Foxp3- T cells in the course of Plasmodium yoelii infection of BALB/c mice

Project description:The objective of the present study was to characterize the phenotype of CD4+CD25+Foxp3+ regulatory T cells (Tregs) in the course of parasitic Plasmodium yoelii (P .yoelii) infection of BALB/c mice. Therefore we performed microarray expression analysis of CD4+CD25+Foxp3+ Tregs isolated by FACS from spleens of non-infected mice and from spleens of mice infected with P. yoelii 3 days and 5 days post infection. By comparing the gene expression profiles, we were able to identify molecules which were differentially expressed by Tregs during parasitic infection and thereby might be involved in their immune-suppressive function. Moreover, we included CD4+CD25-Foxp3- T cells from spleens of non-infected and P. yoelii-infected mice in our analysis. It was proposed that immune-suppressive CD4+CD25-Foxp3- T cells might be induced during Plasmodium infection of mice. Thus, detailed gene expression data of these cells in comparison to CD4+CD25+Foxp3+ Tregs would contribute a better understanding in the phenotype.

RNA-seq of primary mouse B cell progenitor populations after activation with LPS

Project description:Naive resting B-cells were purified from bone marrow from C57BL/6 mice using flow cytometry sorting. Naive B cells were activated with LPS and harvested 3 hours, 10 hours or 33 hours after the LPS treatment. Expanded activated B cells (CD22+ CD138-) and plasmablasts (CD22- CD138+) were also harvested on day 4 post-stimulation.

RNA-seq profiling of malaria pre-exposed but resting effector memory γδ T cells

Project description:Plasmodium chabaudi infected mice were drug-treated on day 14 post-infection with an i.p. injection of CQ (10mg/kg) and pyrimethamine (10mg/kg) followed by CQ (6 mg/ml) and pyrimethamine (70 mg/ml) containing water for 5 days. Spleens were removed 12 weeks after completion of drug treatment. Spleens from drug-treated naive mice were used as controls.

Next Generation Sequencing Facilitates Quantitative Analysis of Wild-type (WT) and Uba3fl/fl-Lck Cre+ (KO) splenic CD44hiCD4+ T cell transcriptomes

Project description:We analyze the impact of T cell-specific deletion of Uba3 gene on CD4+ T cell responses during Plasmodium infection using the RNA-seq techonology. We isolated CD44hiCD4+ T cells in spleens from wild-type (WT) and Uba3fl/fl-LckCre+ (KO)mice at day 5 of Plasmodium yoelii 17XNL infection. RNA samples were prepared for deep-sequencing on an Illumina Hiseq platform and 150 bp paired-end reads were generated.Quanlity control was performed to generate high quality data for further analysis.By comparing gene expression levels of different CD4+ T cell lineages, we observed defects in the expression of several signature genes that related to Th1 and Tfh responses in KO mice.
