RNA-seq of the testis sample of ICI mice treated with gut microbiota transplant 2wks

ABSTRACT: We profiled a RNA-seq on ICR mouse testis. The mice were treated with one dose of busulfan at 3 weeks of age. Then the mice were treated with gut microbiota for 2 weeks. Then after another 3 weeks, testis samples were collected. Then RNA was isoltated from the cultured testis to do RNA-seq.

Project description:We profiled a single-cell RNA-seq on ICR mouse testis treated with busulfan and or Alginate Oligosaccharides (AOS) from 3 weeks of age to 8 weeks of age (for 5 weeks) to study the rescue effect of AOS on the spermatogenesis disrupted by busulfan. We collected single cell from testis from the four groups AOS 0 (control), AOS 10 (AOS 10 mg/kg body weight (BW)), B+A 0 (busulfan alone), B+A 10 (busulfan plus AOS 10 mg/kg BW). The the cells were further captured with 10x Genomics based single-cell system. By using single-cell RNA seq, we found busulfan disrupted spermatogenesis by the dramatical decrease in germ cells. AOS significantly increased the number of germ cells and also promoted the germ cell development. Besides, we provide candidate master regulons regulated by AOS. Our data here offers the opportunity for treating of azoospermia or oligospermia.

Project description:We profiled a single-cell RNA-seq on ICR mouse intestine treated with busulfan and or Alginate Oligosaccharides (AOS) from 3 weeks of age to 5 weeks of age (for 2 weeks) to study the rescue effect of AOS on the intestine development disrupted by busulfan. We collected single cell from intestine from the four groups AOS 0 (control), B+A 0 (busulfan alone), B+A 10 (busulfan plus AOS 10 mg/kg Body Weight). The the cells were further captured with 10x Genomics based single-cell system. By using single-cell RNA seq, we found busulfan damaged intestinal cells. AOS significantly improved the intestinal cell junction and microvilin formation. Besides, we provide candidate master regulons regulated by AOS. Our data here offers the opportunity for improving intestine under stress.

Transcriptome profiles of paternal testis and their offspring in response to gut microbiota dysbiosis using RNA sequencing analysis

Project description:Tissue samples harvested from conventionally-raised sire and their offspring or dysbiotic sire and their offspring were profiled for transcriptome changes using total RNA sequencing. Total RNA was extracted from four different tissue types: sire testis, F1 placenta (at E13.5 & E18.5), and F1 brain and brown adipose tissue (at postnatal day P17).Testis were harvested from 11-weeks inbred male mice that were experiencing gut microbiota dysbiosis for 6-week (antibiotics treated, n=5), or drink sterilized water (control, n=5). While, F1 offspring samples were collected from three independent matings per group and two offspring samples per litter.

RNA sequencing analysis of busulfan-treated initial segment-caput epididymis.

Project description:Purpose: The purpose of this study is to characterize genes whose expressions in the initial segment (IS)-caput epididymis were affected by busulfan intraperitoneal injection. Methods: 8-week-old wild-type mice were treated with single intraperitoneal injection of dimethyl sulfoxide (DMSO) or busulfan (40mg/kg body weight) disolved in DMSO and mice were left for 4 weeks until tissue sampling at 12 weeks old. IS-caput epididymal mRNA profiles were generated by deep sequencing, in triplicate, using Illumina NovaSeq6000. Results: RNA-seq data identified differentially expressed transcripts. Conclusions: Our results show that the expression of many genes was affected by busulfan injection.

Global Gene Expression in the Human Fetal Testis and Ovary

Project description:This study describes a temporal profile of gene expression from normal human fetal testes and ovaries. Gonads from 34 fetuses between 9 weeks and 20 weeks of gestation were obtained from the Department of Pathology and the Birth Defects Research Laboratory at the University of Washington. Relative transcript levels were determined using the Affymetrix Human Genome U133A Plus 2.0 arrays. Sex determination occurs in the human gonad at approximately 6 weeks gestation with development of the testis driven by expression of SRY. In this study, SRY transcript was present and elevated at 9 weeks gestation in the testis but absent in the ovary. The transcript levels of other testis-specific factors SOX9, AMH, and the steroidogenic genes CYP17a1, CYP11a1, STAR and HSD17β3 were all significantly higher in the testis. In contrast, transcripts known to be involved in meiosis including STRA8, SPO11, SYCP3, TEX11, TEX14 and STAG3 showed highest expression in the fetal ovary beginning at week 12. These gene expression profiles will be a resource for understanding and defining normal gonad development and provide the opportunity to dissect abnormal development.
