Sham-irradiated vs pool of sham-irradiated keratinocytes

ABSTRACT: CEA_SGF:E00007# This set of experiments was made in order to perform time series analyses. RNAs from sham-irradiated cells at each time point were pooled together. We then hybridized sham-irradiated samples at each time point versus cDNAs obtained from the pooled RNA. Each time point was assessed with two independent dye-swap hybridizations. For this, differentiated cultures derived from a single donor were irradiated at either 1 cGy, 2 Gy or sham-irradiated at T0 and total RNA extracts were prepared at 6 time points post-irradiation. Irradiated samples for each dose were hybridized seperately with sham-irradiated samples at the same time point. Project description:We used transverse aortic constraction pressure overload hypertrophy mouse hearts as a model of cardiovascular disease to study the genetic changes between TAC and SHAM (normal) mouse hearts and over 1 circadian cycle (24h). This is one approach to identify diurnal genetic biomarkers of cardiovascular disease. The micorarray approach allowed to see the gene expression in all genes in cardiovascular disease and sham hearts. There are 36 samples of cardiovascular disease (TAC) and normal SHAM hearts. For TAC: There were 3 mice sacrificed at each time point as biological replicates, for 6 timepoints over 24 hrs. For SHAM: There were 3 mice sacrificed at each time point as biological replicates, for 6 timepoints over 24 hrs.

Expression data in mouse liver exposed to low dose-rate radiation

Project description:Expression profiles in mouse liver exposed to long-term gamma-irradiation were examined to assess in vivo effects of low dose-rate radiation. Three groups of male C57BL/6J mice were exposed to whole body irradiation at dose-rates of 17-20 mGy/day, 0.86-1.0 mGy/day or 0.042-0.050 mGy/day for 401-485 days (cumulative doses were approximately 8 Gy, 0.4 Gy or 0.02 Gy, respectively). Expression profiles were produced for RNA isolated from irradiated individual animals and for pooled RNA from sham-irradiated 3 animals for control. The expression levels of 6 irradiated animals for each dose were compared individually with those of 2 pooled controls (3 irradiated samples to one pooled control in first and second experiments).

Transcription profiling of superior mesenteric arteries from rats that have had portal vein ligation or a sham procedure for different lengths of time

Project description:Twenty-four male Sprague-Dawley rats were used for this experiment, a partial portal vein ligadure (PVL) was carried out in 12 rats, the other 12 rats were sham-operated. Superior mesenteric artery (SMA) samples were sequentially obtained from 2 rats from the PVL and sham groups at 1, 6, and 24 hours, and 3, 5 and 14 days after portal vein ligation or sham procedure. Each PVL sample was paired with its corresponding sham sample using a dye swap experiment design (technical replicates) resulting in 12 arrays (6 time points x 2 dye swap).
