Effects of Overexpression of Dube3a in Drosophila Brain

ABSTRACT: Fly Dube3a was expressed using either the neuronal driver (elav-GAL4) or the glial driver (Repo-GAL4). RNAseq data was compared among groups (elav>Dube3a), (repo>Dube3a) and (Dube3a alone). Expression was measured in whole dissected fly brain.

To understand the roles of glial sl further, we performed bulk transcriptomics. The E3 Ligase CUL3 physically interacts with TANGO10 protein in vivo

Project description:List of protein scores for putative TANGO10 interactors from mass spectrometry analysis of FLAG-tagged TANGO10 using either elav-GAL4 or tim-GAL4 at both ZT10 and ZT 22, as determined using Proteome Discoverer software (ThermoFisher). Fly heads from GAL4 heterozygotes were used as controls. The hits from FLAG-tagged twenty-four (TYF) proteomics were also excluded from the list to increase TANGO10-specificity. Proteins listed are those present in at least one elav-GAL4 sample and at least one tim-GAL4 sample but no negative controls.

RNA-Seq of whole Drosophila larval central nervous systems (mid-3rd instar stage, central brain + ventral nerve cord) with panneuronal ISWI knockdown and controls

Project description:Sleep disruptions are among the most commonly-reported symptoms across neurodevelopmental disorders (NDDs), but mechanisms linking brain development to normal sleep are largely unknown. From a Drosophila screen of human NDD-associated risk genes, we identified the chromatin remodeler Imitation SWItch/SNF (ISWI) to be required for adult fly sleep. To better understand the mechanisms by which ISWI regulates sleep, we performed RNA-Seq analysis on mid-3rd instar larval central nervous systems (central brain + ventral nerve cord) in the setting of pan-neuronal ISWI knockdown (elav-GAL4>UAS-ISWI RNAi) compared to controls (elav-GAL4;+). We chose mid-3rd instar because temporal mapping revealed ISWI knockdown during this pre-adult developmental stage led to adult fly sleep deficits.

RNA-seq identifies genes that are regulated by SAGA deubiquitinase activity in glial nuclei of the Drosophila central nervous system during larval development

Project description:Purpose: Next-generation sequencing (NGS) was used to elucidate genes in Drosophila visual system glia that are sensitive to impairment of the deubiquitinase activity of the Spt-Ada-Gcn5 Acetyltransferase (SAGA) complex. Methods: The nuclear envelope of glial cells was genetically labled with a KASH-GFP fusion protein using the Gal4/UAS binary expression system and repo-Gal4 as driver in wild-type, nonstop and sgf11 third instar larvae. GFP-labeled glial nuclei were released by homogenization and then isolated from the central nervous system/eye antennal disc complex of Drosophila melanogaster third instar larvae from each genotype, and mRNA was extracted. For RNA-seq, dsDNA was generated using the Ovation RNA-Seq System V2 (Nugen technologies), DNA libraries were generated using standard methods (Illumina), and paired-end 100-base fragements were sequenced on an Illumina HiSeq2500 next generation sequencer. Result: EdgeR analysis with significance cut-off of FDR<0.01 identified 629 genes that are significantly downregulated, and 779 genes that are significantly upregulated, in both the nonstop and sgf11 visual system glia as compared to wild-type glia.

Microarray analysis of a Drosophila dopamine transporter mutant, fumin (fmn)

Project description:We previously found a short sleeper mutant, fmn, and identified its mutation in the dopamine transporter gene. In an attempt to discover additional sleep related genes in Drosophila, we carried out a microarray analysis comparing mRNA expression in heads of fmn and control flies and found differentially expressed genes. Next, using the pan-neuronal Gal4 driver elav-Gal4 and UAS-RNA interference (RNAi) to knockdown individual genes, we performed a functional screen. We found that knockdown of the NMDA type glutamate receptor channel gene (Nmdar1) reduced sleep.

An embryonic stage 17 single cell atlas from Drosophila melanogaster

Project description:A major driver of neural circuit complexity is cellular heterogeneity. We sought to profile all of the neural cell types in the Drosophila embryo at stage 17, a period of rapid synapse and circuit formation in the central nervous system. To do so, we performed whole embryo dissociations of Drosophila embryos followed by single cell RNA sequencing. We then computationally segregated neural cell types based on known gene expression for all neurons (elav) and glia (repo). We hope that this dataset will serve as a community resource for other researchers interested in the molecular determinants of circuit complexity.
