Whole-genome single-cell 5mC sequencing of human primordial germ cell-like cells

ABSTRACT: In this study, we evaluated the 5mC content of single cell human primordial germ cell-like cells (hPGCLCs) generated from a male human embryonic stem cell line (UCLA2, 46XY). tilizing a strand-specific, enzymatic based method of sequencing, we compared the 5mC content of the plus strand relative to the whole chromosome. We evaluated just specified hPGCLCs, at day 4 of differentiation (D4) and then D4 hPGCLCs that were cultured on a feeder layer for additional 10 days (D4C10).
