The original goal of the study was to determine if there was compensation from other L1 family members or alterations in cell survival or apoptosis. Project description:Cerebellum from post-natal day 11 L1 knockout mice on the 129Sv background were compared to wild type littermates. The original goal of the study was to determine if there was compensation from other L1 family members or alterations in cell survival or apoptosis. Interestingly no major changes were detected in those families or pathways. The microarray experiment was conducted by extracting total RNA from the cerebellums of three postnatal day 11 congenic 129Sv L1 knock outs and three of their wild type siblings utilizing Qiagen's RNeasy Mini kit (# 74104). Therefore, each genotype, L1KO and wildtype, had three replicates. Samples were processed at the Case Western Reserve University Cancer Center Gene Expression Array Core Facility and hybridized to Affymetrix Murine Genome U74AV2 gene chips.

Target genes of thyroid hormone in cerebellum neurons

Project description:Cerebellar post-natal development is particularly sensitive to thyroid hormone and low levels of thyroid hormone (hypothyroidism) result in permanent defects in cerebellar architecture and function. All cell types of the cerebellum are affected, but the main sign of hypothyroidism in mice is the persistence of the external granular layer, composed of mitotic neuronal precursors at P21. To make the genetic link between thyroid hormone and cerebellar development, we sought to identify new thyroid hormone target genes, in particular in granule cells which represent the vast majority of cerebellar cells.

Project description:The goal of this experiement was to determine what transcriptional differences there are between P195A/p150- (sick) and P195A/p150+ (healthy) mice. Kidney, Liver, and Cerebellum were the most clearly affected on histology and qPCR for IFN. We found an IFN signature and ISR signature were both upregulated in P195A/p150- mice as compared to P195A/p150+ littermates.

Project description:In an attempt to elucidate the molecular mechanisms underlying the multiple roles of L1 in endothelium, we checked whether manipulating its expression affected the transcriptome of lECs. To this purpose, we compared the gene expression profiles of L1-overexpressing and control lECs by Affymetrix, which revealed a remarkable effect of L1 overexpression on lECs transcriptome. Mouse lung-derived EC (lECs), kindly provided by E. Dejana (Milan, Italy), were stably transfected with a pcDNA3.1/Hygro(-) vector containing mouse LICAM full length or with and empty pcDNA3.1/Hygro(-) vector as control. Three independent experimental replicates were performed.

Project description:Inbred Long-Sleep (ILS) and Inbred Short-Sleep (ISS) mice exhibit a large difference in a number of alcohol and drug related behaviors. This study examined the expression levels of transcripts in these strains in the cerebellum, which is a major target of ethanol’s actions in the CNS, in order to find differentially expressed candidate genes for these phenotypes. Cerebellum was specifically chosen due to the fact that Purkinje cell sensitivity to ethanol in these strains is highly correlated to "sleep time", the measure of ethanol sensitivity used with these strains.
