ABSTRACT: Anergic T cells are unable to respond to antigen though their TCR is specific for it. Chronic infections and cancer can induce a dsyfunctional immune response which cannot clear the cause of disease. We asked in how far anergic T cells are similar to antigen-exhausted ones.

Gene expression in anergic and naïve CD4 T cells deficient for Nr4a1 and Nr4a3

Project description:The NR4A family of nuclear receptors are upregulated by acute and chronic antigen stimulation, and play redundant, tolerogenic roles in T and B cells. To bypass their obligate function in the Treg lineage, we generated competitive chimeras with Nr4a1-/-Nr4a3-/- (double knockout or DKO) bone marrow mixed with congenically marked WT bone marrow at a 1:5 ratio. As a result of a defect in Treg production by DKO cells, Treg in mixed chimeras are almost exclusively of WT origin. By contrast, we generate an approximately 1:1 DKO:WT ratio in the peripheral T cell compartment because DKO cells escape negative selection. After reconstitution, we observed pronounced accumulation of anergic DKO CD4 T cells, but a defect in functional tolerance resulting in autoantibody production. Based on these studies and published literature, we hypothesize that the NR4A family regulate TCR-induced transcripts in both naive and anergic CD4 T cells. To identify such targets, we sorted naive and anergic CD4 T cells of either WT or DKO genotype (CD45.1+ or CD45.1-) on the basis of CD73/FR4/CD44 expression from these chimeras directly into RLT buffer. In parallel, we stimulated sorted naive CD4 T cells of each genotype in vitro with plate-bound anti-CD3 and anti-CD28 stimulation for 3 hrs and then generated RLT lysates. We made biological triplicate samples of each condition (naive WT and DKO, naive stimulated WT and DKO, anergic WT and DKO) resulting in 6 different populations x 3 replicates = 18 samples. Samples were subjected to library prep and bulk RNA sequencing to identify DEG.

Project description:During acute viral infections, naïve CD8+ T cells differentiate into effector CD8+ T cells and, after viral control, into memory CD8+ T cells. Memory CD8+ T cells are highly functional, proliferate rapidly upon reinfection and persist long-term without antigen. In contrast, during chronic infections, CD8+ T cells become “exhausted” and have poor effector function, express multiple inhibitory receptors, possess low proliferative capacity, and cannot persist without antigen. To compare the development of functional memory T cells with poorly functional exhausted T cells, we generated longitudinal transcriptional profiles for each. Naive CD44Lo CD8+ T cells were isolated and sorted from uninfected C57BL/6 mice and H2-Db GP33-specific CD8+ T cells were sorted using MHC-I tetramers at d6, 8, 15, and 30 p.i. with either LCMV Arm or LCMV clone 13. RNA from these CD8+ T cells was processed, amplified, labeled, and hybridized to Affymetrix GeneChip MoGene 1.0 st microarrays

We asked how antigen persistence alone affects T helper cell reactivity v. exhaustion and compared the gene expression of CD4+ T cell responding to antigen persisting at different levels for different periods of time. We used microarrays to detail gene expression underlying T helper cell dysfunctionality evolving in response to different levels of antigen at two timepoints.

Identification of an anergic BND cell-derived activated B cell population (BND2) in young-onset type 1 diabetes

Project description:Recent evidence suggests a role for B cells in the pathogenesis of type 1 diabetes (T1D), particularly in individuals who develop T1D at a younger age and demonstrate rapid progression and beta cell loss. However, little is known regarding the specificity, phenotype, and function of B cells in young-onset T1D. We performed a cross-sectional analysis comparing insulin-reactive to tetanus-reactive B cells in the blood of T1D and controls using mass cytometry. Unsupervised clustering revealed the existence of a highly activated B cell subset, we term BND2, that falls within the previously defined anergic BND subset. We found a specific increase in the frequency of insulin-reactive BND2 cells in the blood of young-onset T1D donors, which was further enriched in the pancreatic lymph nodes of T1D donors. The frequency of insulin-binding BND2 cells correlated with anti-insulin autoantibody levels in T1D subjects. We demonstrate BND2 cells phenotypically and functionally are pre-plasma cells and can likely act as antigen-presenting cells to T cells. These findings suggest that activation of insulin-reactive anergic B cells may play a role in the rapid progression of young-onset T1D and warrants further investigation in other autoimmune conditions.

Molecular and functional characterization of alloantigen-specific anergic T-cell suitable for cell therapy

Project description:IL-10 is an immunomodulant cytokine that plays a central role in controlling inflammation, down-regulating immune responses, and inducing immunological tolerance. IL-10 inhibits the expression of costimulatory molecules and the secretion of inflammatory cytokines by antigen-presenting cells (APC), directly suppresses T-cell proliferation, and promotes T-cell anergy. In this study we demonstrate that IL-10 inhibits primary allogeneic proliferation in total PBMC and induces antigen(Ag)-specific T-cell hyporesponsiveness. IL-10-induced anergy is TGF-beta independent, is associated with a decrease of Ag-specific CTLp frequency, and can be obtained among cells with different degree of HLA disparity. The use of tolerogenic dendritic cells (DC) differentiated in the presence of IL-10 (DC-10) allows a more consistent anergy induction, even in the context of HLA matched donors. Importantly, alloAg-IL-10/DC-10-anergized T cells preserve their ability to respond to nominal and third party Ags. Microarray experiments were conducted on different mixed lymphocyte cultures aiming at the identification of gene expression profiles characteristic for the various T cell populations. Independently from the APC used to anergized T cells, the resulting T cell populations exhibit a specific gene signature. Based on the results of this study, we developed a method suitable for GMP scale up and for the generation of a population of anergic Ag-specific T cells for cellular therapy. The dataset comprises 12 samples divided into four sample groups each representing a certain kind of allogeneic mixed lymphocyte culture, i.e. peripheral blood mononuclear cells (PBMC) mixed with CD3-depleted cells (MEC, monocyte-enriched cells) with or without IL-10 treatment, and PBMC mixed with mature or IL-10 treated dendritic cells (DCs). Each group includes three biological replicates with cells collected from different donors.
