Gene expression in human endometrium cells (ATCC 4003) [control]

ABSTRACT: To detect the gene profiles in non-decidualized stromal cells, cultured stromal cells were collected and subjected to RNA-Seq. After aligned to mouse mm10 by HISAT2, RPKM value was calculated by Edger. Our results show that SOX4 is one of the most abundant gene in the whole SOX family. The expression of SOX4 was further confirmed by qPCR, ISH and immunofluorescence. The expression of SOX4 in human endometrium indicated the essential role of SOX4 for reproduction. This RNA-Seq data provides fundamental information for our further physiological study of SOX4.

Project description:To detect the gene profiles in decidualized stromal cells affected by SOX4, cultured stromal cells were transfected with siSOX4 and subjected to RNA-Seq. After aligned to mouse mm10 by HISAT2, RPKM value was calculated by Edger. Our results show that SOX4 was remarkable dimished after siSOX4 transfection. Meanwhile, some decidual marker genes were also changed significantly after SOX4 knockdow. The expression of SOX4 and other downstream genes were further confirmed by qPCR and westerblot. This RNA-Seq data provides fundamental information for our further physiological study of SOX4 during human stromal cells decidualization and fertility.

The expression of HMGB1 was further confirmed by qPCR, ISH and immunofluorescence. The expression of HMGB1 was found expressed in very cells, indicating the essential role of HMGB1 for reproduction. This RNA-Seq data provides fundamental information for our further physiological study of HMGB1.

Transcription profiling of human endometrial stromal cells treated with human trophoblasts vs. control, sampled at multiple time points to investigate stromal cell response to paracrine signals from the trophoblast

Project description:During the invasive phase of implantation, trophoblasts and maternal decidual stromal cells secrete products that regulate trophoblast differentiation and migration into the maternal endometrium. Paracrine interactions between the extravillous trophoblast and the maternal decidua are important for successful embryonic implantation, including establishing the placental vasculature, anchoring the placenta to the uterine wall, and promoting immuno-acceptance of the fetal allograph. Global cross-talk between the trophoblast and the decidua has not been elucidated to date, and the current study used a functional genomics approach to investigate these paracrine interactions. Human endometrial stromal cells were decidualized with progesterone and were further treated with conditioned media (CM) from human trophoblasts (TCM) or, as a control, with conditioned media (CCM) from non-decidualized stromal cells for 0, 3 and 12 hr. Total RNA was isolated and processed for analysis on whole genome, high density oligonucleotide arrays, containing 54,600 genes. Our data demonstrate a significant induction of pro-inflammatory cytokines and chemokines, as well as angiogenic/static factors in decidualized endometrial stromal cells in response to trophoblast-secreted products. The data suggest that the trophoblast acts to alter the local immune environment of the decidua to facilitate the process of implantation and assure an enriched cytokine/chemokine environment, while limiting mitotic activity of the stromal cells during the invasive phase of implantation.

Project description:To detect the direct target genes of SOX4 in differentiated human primary endometrial stromal cells, differentiated human primary endometrial stromal cells cells at the presence of MPA, E2 and cAMP are collected and subjected to ChIP-Seq. After aligned to human Hg38 by STAR, peaks are called by MACS2. Our results show that SOX4 is the key regulator for decidualization by regulating FOXO1, STAT3, PRL and FOSL2. This specific enrichment is confirmed by ChIP-qPCR. Additionally, there are also some other direct genes of SOX4, indicating the essential role of SOX4 in decidualization. This ChIP-Seq data provides fundamental information for our further physiological study of SOX4.
