ABSTRACT: Genome-wide patterns of DNA methylation were quantified using the Illumina Infinium HumanMethylation450 BeadChip (“450K array”) in DNA samples isolated from blood for schizophrenia cases and controls. DNA methylation in postmortem brain samples from autism cases and controls

Project description:We used the Illumina 450KMethylation BeadChip to measure DNA methylation at 485,512 loci across the genome for 40 postmortem brain samples. The purpose of our study was to identify differentially methylated regions (DMRs) associated with autism. Samples included 16 temporal cortex brain tissue samples (6 cases and 10 controls), 11 prefrontal cortex brain tissue samples (6 cases and 5 controls), and 13 cerebellum brain tissue samples (7 cases and 6 controls). We bisulfute converted DNA from the 40 brain tissue samples, processed, and hybridized them to the Illumina Infinium 450K HumanMethylation450 BeadChip as specificed by the manufacturer.

Project description:Genome wide DNA methylation profiling of Neurons derived from human induced pluripotent stem cells (iCell Neurons) at different time points were abtained by the Infinium HumanMethylation450 BeadChip (450K array) Kit (WG-314-1002), which interrogates >480,000 CpG sites in the human genome.In total, 36 samples were profiled using the 450K arrays. 1 ug purified DNA from each timepoint was submitted to the University of Chicago Genomics Core where it was converted with the EZ-96 DNA Methylationâ?¢ Kit (Deep Well Format), as per manufacturer's protocol, followed by array analysis using the Infinium HumanMethylation450 BeadChip (450K array) Kit (WG-314-1002), which interrogates >480,000 CpG sites in the human genome.
