TGF-ß treatment of human melanoma cell lines

ABSTRACT: To analyse whether the activation of TGF-ß signalling (recombinant hTGF-ß, 5 ng/mL; PeproTech) is able to invoke a full phenotype- switch, we performed RNA sequencing of melanoma cultures from two independent melanoma cell lines at two different time point after TGF-β1 (5 ng/ml, preprotech).

Project description:Deep RNA sequceing was performed to explore the expression level of miRNAs in HeLa cell exosomes after treatment with or without 10 ng/ml TGF-β1 for 24 h. The results showed that 48 miRNAs were enriched in exosomes of HeLa cells after treated with TGF-β1 compared with non-treated cells (negative control), proving that TGF-β1 affects HeLa cell exosomes miRNA profile.

Genome-wide identification and analysis of long noncodingRNAs in porcine preadipocytes treated with TGF-β1

Project description:Several studies have revealed thatTGF-β1 may play a key role in epithelial-mesenchymal transition (EMT); however, little is known about the effects of TGF-β1 treatment on porcine preadipocytes. To identify the key lncRNAregulator, two libraries were constructed from porcine preadipocytes were treated with or without 10 ng/mL human recombinant TGF-β1 for 48 h by using the Illumina HiSeqXten platform. RT-qPCR was used to detect the expression of ten differentially expressed lncRNAs, andthe results were consistent with the sequencing results. In addition, the lncRNATargets platform was used to construct an interaction network of lncRNAs and the Smad7 gene.A total of 39 lncRNA transcripts, including 28 upregulated and 11 downregulated transcripts, were significantly differentially expressed in the TGF-β1 group compared with the control group. Functional annotation and enrichment analysisof the host genes revealed that the differentially expressed lncRNAs were related to single-organism process, cell part and cellular process.We constituted the lncRNA/mRNAco-expression network in the TGF-βsignaling pathway.In our study, we systematically investigated the lncRNA contents in porcine preadipocytes were treated with or without 10 ng/mL human recombinant TGF-β1. A total of 8158 lncRNA transcripts were identified, and 39 of these transcripts were significantly differentially expressed between two libraries.

miRNA profiling in primary human peritoneal mesothelial cells (HPMCs) and TGF-β1-stimulated

Project description:TORAY microarray assays were performed on HPMCs derived from four independent donors. miRNA expression was measured in control samples and after 1 ng/ml of TGF-β1 treatment for 48 h. TGF-β1-driven mesothelial-to-mesenchymal transition (MMT) was confirmed in all samples by analysis of the eight molecular markers previously described (data not shown). A total of 1 μg in a volume of ≤2 μl of total RNA from each sample was subsequently used for Toray microarray analysis, performed by Central Biotechnology Services at Cardiff University.

Effect of Tmsb4x knockout on TGF-ß/MRTF signaling

Project description:Myocardin-related transcription factors (MRTFs) are robust co-activator of Serum Reponse Factor, and regulate cell adhesion and motility by controlling the transcription level of cytoskeletal components. In tumor progression, MRTFs has been reported to contribute to metastatic potential. Previously, we reported thymosin-ß4 (Tß4, which is encoded by Tmsb4x gene) as an activator of MRTFs. Enhanced expression of Tß4 is frequently observed during tumor progression, and associated with poor prognosis. However, the relationships between MRTFs and Tß4 in tumor progression are largely unknown. Here we analyze the effect of Tß4 knockout on the transcription levels of MRTFs-target genes. In particular, we focused on their functions in TGF-ß signaling because both MRTFs and Tß4 are down-stream effectors of TGF-ß.
