Project description:Purpose: To demonstrate Runx3's association with organizing the extracellular matrix in SFZ/DZ chondrocytes. Method: RNA samples were collected from SFZ/DZ chondrocytes of 5-day-old Col2a1-Cre;Runx3fl/fl and Runx3fl/fl mice. Results: 18 genes were up- or down-regulated by more than 2-fold in the Runx3 knockout in SFZ. Conclusions: However, genes organizing the extracellular matrix were down-regulated in Runx3 cKO DZ chondrocytes.

Accumulation of exogenous activated TGF-? in the superficial zone of articular cartilage.

Project description:It was recently demonstrated that mechanical shearing of synovial fluid (SF), induced during joint motion, rapidly activates latent transforming growth factor ? (TGF-?). This discovery raised the possibility of a physiological process consisting of latent TGF-? supply to SF, activation via shearing, and transport of TGF-? into the cartilage matrix. Therefore, the two primary objectives of this investigation were to characterize the secretion rate of latent TGF-? into SF, and the transport of active TGF-? across the articular surface and into the cartilage layer. Experiments on tissue explants demonstrate that high levels of latent TGF-?1 are secreted from both the synovium and all three articular cartilage zones (superficial, middle, and deep), suggesting that these tissues are capable of continuously replenishing latent TGF-? to SF. Furthermore, upon exposure of cartilage to active TGF-?1, the peptide accumulates in the superficial zone (SZ) due to the presence of an overwhelming concentration of nonspecific TGF-? binding sites in the extracellular matrix. Although this response leads to high levels of active TGF-? in the SZ, the active peptide is unable to penetrate deeper into the middle and deep zones of cartilage. These results provide strong evidence for a sequential physiologic mechanism through which SZ chondrocytes gain access to active TGF-?: the synovium and articular cartilage secrete latent TGF-? into the SF and, upon activation, TGF-? transports back into the cartilage layer, binding exclusively to the SZ.

Wnt/?-catenin signaling contributes to articular cartilage homeostasis through lubricin induction in the superficial zone.

Project description:BACKGROUND:Both loss- and gain-of-function of Wnt/?-catenin signaling in chondrocytes result in exacerbation of osteoarthritis (OA). Here, we examined the activity and roles of Wnt/?-catenin signaling in the superficial zone (SFZ) of articular cartilage. METHODS:Wnt/?-catenin signaling activity was analyzed using TOPGAL mice. We generated Prg4-CreERT2;Ctnnb1fl/fl and Prg4-CreERT2;Ctnnb1-ex3fl/wt mice for loss- and gain-of-function, respectively, of Wnt/?-catenin signaling in the SFZ. Regulation of Prg4 expression by Wnt/?-catenin signaling was examined in vitro, as were upstream and downstream factors of Wnt/?-catenin signaling in SFZ cells. RESULTS:Wnt/?-catenin signaling activity, as determined by the TOPGAL reporter, was high specifically in the SFZ of mouse adult articular cartilage, where Prg4 is abundantly expressed. In SFZ-specific ?-catenin-knockout mice, OA development was significantly accelerated, which was accompanied by decreased Prg4 expression and SFZ destruction. In contrast, Prg4 expression was enhanced and cartilage degeneration was suppressed in SFZ-specific ?-catenin-stabilized mice. In primary SFZ cells, Prg4 expression was downregulated by ?-catenin knockout, while it was upregulated by ?-catenin stabilization by exon 3 deletion or treatment with CHIR99021. Among Wnt ligands, Wnt5a, Wnt5b, and Wnt9a were highly expressed in SFZ cells, and recombinant human WNT5A and WNT5B stimulated Prg4 expression. Mechanical loading upregulated expression of these ligands and further promoted Prg4 transcription. Moreover, mechanical loading and Wnt/?-catenin signaling activation increased mRNA levels of Creb1, a potent transcription factor for Prg4. CONCLUSIONS:We demonstrated that Wnt/?-catenin signaling regulates Prg4 expression in the SFZ of mouse adult articular cartilage, which plays essential roles in the homeostasis of articular cartilage.

Effect of Moderate Exercise on the Superficial Zone of Articular Cartilage in Age-Related Osteoarthritis.

Project description:This study aimed to evaluate the effect of exercise on the superficial zone of the osteoarticular cartilage during osteoarthritis progression. Three-month-old, nine-month-old, and eighteen-month-old Sprague Dawley rats were randomly divided into two groups, moderate exercise and no exercise, for 10 weeks. Histological staining, immunostaining, and nanoindentation measurements were conducted to detect changes in the superficial zone. X-ray and micro-CT were quantitated to detect alterations in the microarchitecture of the tibial subchondral bone. Cells were extracted from the superficial zone of the cartilage under fluid-flow shear stress conditions to further verify changes in vitro. The number of cells and proteoglycan content in the superficial zone increased more in the exercise group than in the control group. Exercise can change the content and distribution of collagen types I and III in the superficial layer. In addition, TGFβ/pSmad2/3 and Prg4 expression levels increased under the intervention of exercise on the superficial zone. Exercise can improve the Young's modulus of the cartilage and reduce the abnormal subchondral bone remodeling which occurs after superficial zone changes. Moderate exercise delays the degeneration of the articular cartilage by its effect on the superficial zone, and the TGFβ/pSmad2/3 signaling pathways and Prg4 play an important role.
