Regeneration in sponge Sycon ciliatum partly mimics postlarval development

ABSTRACT: We performed high-throughput sequencing on regenerating samples and compared the data with regular embryonic and postlarval development of Sycon ciliatum. Our comparative transcriptomic analysis illuminates that sponge regeneration is equally dynamic as embryogenesis.

Transcriptomic changes during regeneration in demosponge Cinachyyrella cf cavernosa

Project description:In this study, the differential regeneration process in sponge Cinachyrella cf cavernosa was examined by creating a wound at basal and non-basal tissues. The differences in wound healing were investigated based on qualitative and quantitative changes in collagen at both wound locations. The variation at the cellular level was also studied during regeneration. To understand the molecular process of gene regulation during regeneration in C.cf cavernosa, differential gene expression (transcriptomics) analysis was carried out using Illumina NextSeq 500. The basal tissue was found to heal faster than non-basal tissue and collagen was found to be the responsible factor for this increased rate of wound healing. There was no apparent difference in the types and density of cells present in regenerative basal and non-basal tissue. The differential analysis of fast regenerating basal tissue showed up-regulation in collagen-related gene Col and actin-related genes during progressive stages of regeneration. Contrary to other metabolism-related genes, lipid metabolism related genes are expressed in more numbers with consecutive stages of regeneration highlighting their role in stress management of the sponge. This study serves to advance our understanding of the regeneration process, factors affecting the rate of this process and transcriptomic changes in regenerating sponge.

Trial for Retractor Sponge Evaluation in Laparoscopic Colorectal Surgery

Project description:Laparoscopic surgery of the distal colon and rectum requires surgery with an appropriate field of view. A commonly used technique to create a clear exposure is the steep Trendelenburg position in which the patient is positioned in an angle of 15 to 40 degrees with the head down using the effect of gravity to retract the small intestine. This method is associated with haemostatic changes caused by the cranial shift of abdominal organs and blood. Recently, a cellulose compressed sponge was developed as intraoperative retractor, with the aim to keep the small intestines aside while the patient remains in a horizontal position. The safety of the sponge is secured with CE marking. The retractor sponge ensures a clear surgical field and potentially prevents haemostatic instability by avoiding Trendelenburg position. A pilot study in the St Antonius Hospital Nieuwegein has shown that use of the sponge might be associated with shorter hospital stay.

Currently, the identified miRNA sponge genes mostly act on sequestering conserved miRNAs (e.g. miR-7, miR-145), however, the existence, potential function and evolutionary process of miRNA sponge genes for species-specific miRNA, especially for human specific miRNA, are largely unknown. Project description:In order to compare sponge and eumetazoan (higher animal) body plans, we identified and studied expression of a broad range of eumetazoan developmental regulatory genes in Sycon ciliatum (Calcispongiae). In this species, embryonic development is semi-synchronous within a population, synchronous within individuals, and oocytes and embryos occupy a significant fraction of the volume of the sponges during the reproductive period. RNASeq libraries representing non-reproductive (somatic) tissue slices along the body axis, as well as oocytes, embryos and free swimming larvae were generated from material obtained by sampling throughout the life cycle.
