Project description:Defense against attaching and effacing (A/E) bacteria requires the sequential generation of IL-23 and IL-22 to induce protective mucosal responses. While the critical source of IL-22 has been identified as CD4+ and Nkp46+ innate lymphoid cells (ILCs), the precise source of IL-23 is unclear. Here, we use genetic techniques to deplete specific classical dendritic cell (cDC) subsets and analyze immunity to the A/E pathogen Citrobacter rodentium. We find that Zbtb46+ cDCs, and specifically Notch2-dependent intestinal CD11b+ cDCs, but not Batf3-dependent CD103+ cDCs, are required for IL-23 production and immunity against C. rodentium. Notch2 controls cDC differentiation at a terminal step mediated by lymphotoxin signaling. Importantly, these results provide the first demonstration of a non-redundant function of CD11b+ cDCs in vivo. Analysis of differentially expressed genes in ESAM+ and ESAM- CD11b+ and DEC205+ splenic classical DC subsets. Splenocytes were harvested from WT C57Bl/6 or WT Cx3cr1-gfp mice and cDC subsets sorted to >95% purity on the FACSAriaII.

Project description:Classic dendritic cells (cDCs) play a central role in the immune system and consist of two major subsets: CD141+ cDC (cDC1) and CD1c+ cDC (cDC2). The pre-cDCs is the immediate precursors to both cDC subsets. Previous studies showed that there were two pre-committed pre-cDC subpopulations. However, the key molecular drivers of pre-commitment in human pre-cDCs were not investigated. To address this question, we performed both single cell and bulk RNA sequencing (RNA-seq) of two cDC subsets and pre-cDCs. We inferred a list of sixteen candidate master regulator transcriptional factors (TFs) that can indeed separate pre-cDCs into two sub-populations, with one close to cDC1 and the other close to cDC2. More importantly, these two pre-cDC sub-populations are correlated with ratio of IRF8 to IRF4 expression level more than their individual expression level. Our results suggest the concept that the ratio of antagonistic TFs and their competition determine cDC subset differentiation fate.

Project description:Oral tolerance prevents pathological inflammatory responses towards innocuous foreign antigens via peripheral regulatory T cells (pTreg cells). However, whether a particular subset of antigen-presenting cells (APCs) is required during dietary antigen exposure to instruct naïve CD4+ T cells to differentiate into pTreg cells has not been defined. Using myeloid lineage-specific APC depletion in mice, we found that monocyte-derived APCs are dispensable, while classical dendritic cells (cDCs) are critical for pTreg cell induction and oral tolerance. CD11b¬â?? cDCs from the gut-draining lymph nodes efficiently induced pTreg cells, and conversely, loss of IRF8-dependent CD11bâ?? cDCs impaired their polarization, although oral tolerance remained intact. These data reveal the hierarchy of cDC subsets in pTreg cell induction and their redundancy during oral tolerance development. Sorted naïve CD45.1 OT-II CD4 T cells were co-cultured with four dendritic cell subpopulations sorted from mouse mesenteric lymphnodes. 24h later OT-II cells were sorted again and compared in their gene expression profile.

Project description:The goal of this study is to compare the gene profiling of human blood BDCA-1+ cDCs, blood BDCA-3+ cDCs, CB CD172a-pre-cDCs and CB CB172a+ pre-cDCs. RNA from sorted populations was extracted and column-purified using the Arcturus PicoPure RNA Isolation kit (Applied Biosystems). Genomic DNA was removed by on-column digest with DNAse I (Qiagen), according to the PicoPure RNA Isolation kit manual. RNA libraries were prepared using the SMARTer Ultra Low Input RNA for Sequencing kit (Clontech Laboratories) followed with Nextera XT DNA Library Prep kit (Illumina). Libraries were sequenced by 75-bp single-end reading on a NextSeq 500 sequencer (Illumina). Our results show that CD172a+ and CD172a- pre-cDCs represent developmentally discrete populations that differentially express lineage-restricted transcription factors. Moreover CD172a- pre-cDCs cluster with BDCA-3hi cDCs and CD172a+ pre-cDCs with BDCA-1+ cDCs.

Project description:Classical dendritic cells (cDCs) process and present antigens to T cells. Under steady-state conditions, antigen presentation by cDCs induces tolerance. In contrast, during infection or inflammation, cDCs become activated, express higher levels of cell surface MHC molecules, and induce strong adaptive immune responses. We recently identified a cDC-restricted zinc finger transcription factor, zDC, that is not expressed by other immune cell populations, including pDCs, monocytes, or macrophages. Here we define the zDC consensus DNA binding motif and the genes regulated by zDC using chromatin immunoprecipitation and deep sequencing. By deleting zDC from the mouse genome, we show that zDC is primarily a negative regulator of cDC gene expression. zDC deficiency alters the cDC subset composition in the spleen in favor of CD8+ DCs, upregulates activation pathways in steady state cDCs including elevated MHC II expression, and enhances cDC production of VEGF leading to increased vascularization of skin-draining lymph nodes. Consistent with these observations, zDC protein expression is rapidly downregulated after TLR ligation. Thus, zDC is a TLR-responsive cDC-specific transcriptional repressor that is in part responsible for preventing cDC maturation in the steady state. Splenic classical dendritic cells from mice lacking the second exon of the zDC gene were sorted. DEC-205+ and DCIR2+ cDCs from the zDC knockout mice were collected for comparison with wildtype cDCs. Three replicates per group. The wildtype samples have been previously analyzed and are available in the GEO database in Series GSE6259. Project description:Primary Sjögren´s syndrome (pSS) is a heterogeneous inflammatory autoimmune disorder characterized by the infiltration of different immune subsets in exocrine glands and potential development of extra-glandular manifestations. pSS pathology and glandular tissue destruction have been linked to increased IFN responses and the induction of autoreactive T and B cells. However, association of innate immune cells such as Natural Killer (NK) cells and conventional dendritic cells (cDC) with the tissular damage and IFN environment in pSS remains understudied. Here, we identified higher expression of NKG2D, CD107a and superior cytotoxic function on circulating NK cells from pSS which was associated with significant enrichment in a subpopulation of CD16+CD56Hi NK cells in the blood of these patients. In addition, Granzyme B+ NK cells and MICab+ CD1c+ cDC could be found in proximity in tissue sections of salivary glands from pSS patients. Importantly, murine populations of CD64Hi CD11b+CD11c+ cDC and transitional NKG2Dhi CD11b+CD27+ NK1.1 cells, infiltrated in vivo into the salivary gland in a stablished pSS mouse model. Interestingly, transcriptional IFN signatures present on CD1c+ cDC from pSS are characterized by the upregulation of RIG-I and DDX60 sensors and the ISGs IFIT1 and IFIT3. These transcriptional patterns on CD1c+ cDC associated with high basal expression of MICa/b and SLAMF7 ligands for NK receptors, the Fc receptor CD64 and increased ability to induce maturation and CD107a expression in NK cells. Finally, expression of SLAMF7 and MICa/b after Poly I:C exposure was regulated by RIG-I and DDX60 on CD1c+ cDC. Therefore, crosstalk between CD1c+ cDCs and NK cells could contribute to the pathogenesis of pSS, and represent new cellular therapeutic targets for treatment of pSS.
