Expression profiling of plants overexpressing transcription factor WIN1

Project description:T2 progenies of two transgenic lines overexpressing ERF transcription factor WIN1 were grown on soil in parallel under identical conditions. mRNA was extracted from pooled leaves from multiple plants of each line for the microarray experiement. Plants derived from two lines were used in this experiment: line 13, a "medium" overexpressor, and line 22, a strong overexpressor. Control mRNA was obtained from pooled control plants transformed using an empty binary vector. All plants were first selected on medium containing kanamycin before being transferred to soil. Response of Rpp4-silenced plants to soybean rust

Project description:Experimental design: 1 genotypes: PI- (resistant USDA Plant Introduction (PI459025B) line containing SBR Rpp4 resistance gene) 2 treatments: Rub inoculation with BPMV-VIGS construct targeting the LRR region of Rpp4 candidate gene. This BPMV construct and the plant tissue used for microarray was described in Meyer et al. (2009) Plant Physiol. 150:295-307. Control plants were inoculated with empty BPMV vector. At three weeks after BPMV inoculation, all plants were inoculated with Soybean rust (Phakopsora pachyrhizi) isolate LA04-1. Transcript profiling of Bln1 silenced plants (BSMV-VIGS) relative to empty vector and buffer treated controls in barley-powdery mildew interactions

Project description:Barley stripe mosaic virus-induced gene silencing (BSMV-VIGS) was used to identify significant new genes in the regulation of host innate immunity. This experiment was designed to uncover significant changes in Bln1 (Contig12219_at)-silenced plants relative to empty vector and buffer treated controls. Five independent biological replications of a split-plot experimental design were conducted with replications as blocks, treatment with Blumeria graminis f. sp. hordei (Bgh) as the whole-plot factor, and all combinations of genotype (Mla13 and Mla9) and VIGS treatment [Buffer control (mock), BSMV:00 (empty vector), and BSMV:Bln1248] as the split-plot factor for a total of 60 GeneChip hybridizations. Ten seedlings were used as a split-plot experimental unit for each combination of replication, Bgh treatment, genotype, and VIGS treatment. Plants were grown in a controlled 20°C glasshouse prior to VIGS treatment. Twelve days after VIGS treatment, half of the plants in each replication were challenged with the compatible Bgh isolate 5874. Top halves of 5 of the 10 seedling third leaves (about 10 cm) from each split-plot experimental unit were harvested into liquid N2 at 32 hours after inoculation (HAI) - the timepoint with the highest differential Bln1 transcript accumulation (Meng et al. 2009), and after initial establishment of the perihaustorial interface (Caldo et al. 2004). The remaining 5 leaves were used to record infection phenotype 7 days later. The constitutive active form MKK3 (MKK3DD) or MKK4 (MKK4DD) was cloned into the glucocorticoid-inducible vector pTA7002 and transformed using Agrobacterium method into Arabidopsis Col-0 plants. The transgenic plants transformed empty vector was used as control plants (Cont).

Three genotypes: Two lines of Arabidopsis transformed to overexpress QPRT, an enzyme involved in NAD synthesis (NadC4.11 and NadC15.3) and a third control line transformed with an empty vector (162). Leaf discs from the two lines of the QPRT+ plants were incubated with quinolinate, the QPRT substrate, to boost leaf NAD. Discs from the empty vector control were incubated on a buffer withour quinolinate. The experiment was performed in the light and the dark. The aim is to compare the QPRT+ quinolinate samples with the empty vector samples, both for the sample series generated in the light and that generated in the dark. Arabidopsis meiotic-specific recombinase DMC1 IP-MS

Project description:To search for regulators of DMC1 stability, we generated DMC1-FLAG transgenic Arabidopsis plants and conducted immunoprecipitation-mass spectrometry (IP-MS) assay. 6,477 peptides corresponding to 2309 proteins were identified and 1009 proteins remained after removing background interactions using plants transformed with the corresponding empty vector (FLAG). GO term analysis was performed with the obtained data and revealed that ubiquitin proteasome pathway components are significantly enriched among the candidate proteins.
