RNA sequencing reveals the genetic relationship between TOPPs and its regulatory subunit PP1R3 in Arabidopsis thaliana

ABSTRACT: We identified a new regulatory subunit PP1R3 of type 1 protein phosphatase in Arabidopsis by immunoprecipitation coupled with liquid chromatography-mass spectrometry.Through RNA sequencing, it was found that there are few differentially expressed genes (DEGs) between pp1r3-2 and topp septuple mutants (topp-7m).Moreover, the DEGs of pp1r3-2 compared to the wild type (Col) have a great overlap with the DEGs of topp-7m compared to Col, and the change trend of gene expression is very similar.At the same time, we also found that pp1r3-2 can partially restore the gene expression changes in the dominant negative mutant topp4-1.And, the DEGs of pp1r3-2 and pp1r3-3 topp4-1 are less than one hundred, which is less than one-tenth of the number of DEGs of pp1r3-2 compared to Col.These results reveal a strong genetic interaction between PP1R3 and TOPPs at the gene expression level.
